Neither a tmux session nor a separate TTY, on its own, is a security boundary. Choose tmux for persistent, organized terminal work that can be detached and reattached; choose a separate TTY when you need a different terminal endpoint or console context. For security separation, use account permissions and operating-system isolation controls appropriate to the threat.
What does “isolate sessions” mean?
The answer depends on whether “isolate” means keeping terminal work organized and distinct, preserving it through a disconnect, or preventing another user or process from accessing it. A multiplexer and a TTY address terminal interaction; neither choice alone establishes that one workload cannot inspect or affect another.

A TTY is a terminal endpoint. A pseudo-terminal (PTY) is a pair of virtual character devices that provides a terminal interface to programs; it is commonly used by terminal emulators and tools such as tmux. Linux documents PTYs separately from process credentials and session control, so a different terminal endpoint should not be confused with different process privileges. See the Linux PTY manual, credentials(7), setsid(2), and ioctl_tty(2).
How tmux sessions differ from separate TTYs

The tmux project describes each program inside tmux as getting its own terminal managed by tmux, accessible from the terminal where tmux is running. In practical terms, tmux organizes windows and panes into sessions, with a tmux server managing the programs and their terminal output. A client attaches to that server to interact with the work. The tmux project guide explains the model, and the tmux manual describes its server, clients, and communication socket.

A separate TTY gives you another terminal endpoint
Using a separate TTY gives a program a different terminal context, but does not by itself change its user identity, credentials, or permissions. The surrounding console, terminal emulator, login session, and operating-system configuration determine how those endpoints are created and managed.

Why tmux is useful for continuity, not security isolation
When a tmux client detaches or a remote connection is lost, programs managed by the server can continue running and later be reattached from the same or another terminal. That makes tmux useful for long-running remote work. It separates the lifetime of the work from the lifetime of the terminal connection, rather than creating a new security domain.

Sharing matters for access decisions: the tmux guide says a session can have multiple attached clients. The manual documents communication through a socket. For the default socket location, the manual says the per-UID directory must not be world readable, writable, or executable. This is a documented consideration for that setup, not a guarantee for every custom socket path or configuration. Review the actual socket location and permissions when access to a tmux server matters.
What to use when security separation matters
Start with the threat you need to address. If the concern is another user, use separate accounts and appropriate file and process permissions. If workloads need stronger separation, choose operating-system isolation controls suitable for that threat model. Terminal layout alone—whether a tmux session, a new TTY, or a new session ID—does not establish that processes cannot inspect or affect one another.
