Rather than building a mainly linear connection through azide–alkyne coupling, the method adds different groups in sequence around sulfur, creating a tetrahedral arrangement with up to three connections from the hub.
How the sulfur-based method works
The reported process begins with SOF4 gas. Its sulfur center has fluorine atoms that can be displaced in a controlled sequence through sulfur(VI) fluoride exchange, or SuFEx, chemistry. The sequence allows researchers to attach different functional groups to one central point rather than joining components only end to end.

First connection: a primary amine
A primary amine can displace two fluorine atoms in the presence of a tertiary amine catalyst, forming the initial connection at the sulfur hub.
Adding a third group
A secondary amine, an amino acid, or a protected phenol can displace a third fluorine under basic catalysis. If the third group is a protected phenol, the remaining fluorine can then be displaced either by a secondary amine directly or by another protected phenol using a phosphazene base catalyst. These are reported reaction sequences, not a general recipe for every substrate.

What makes the linkage three-dimensional
The important distinction is the geometry: the attached groups extend from a tetrahedral sulfur center. The report presents this as a way to create up to three sequential connections from a single hub, offering a different arrangement from the more linear linkage commonly associated with azide–alkyne click chemistry. John Moses of the University of Nottingham described the result as three sequential reactions, each creating a unique connection, extending “from a single sulfur hub into the 3D world.”
The report called the sulfur(VI) fluoride compounds “sleeping beauties,” describing them as stable until exposure to tertiary amines, which can initiate controlled reactions. That phrase characterizes the researchers’ account; it is not a general guarantee of stability, safe handling, or suitability for storage.

Why chemists may find it useful
A central hub with several distinct connections could give chemists another way to assemble and modify complex molecules. The 2017 account identified biomolecule tagging, fragment-based drug discovery, and functional polymers as relevant research directions. Eric Anslyn of the University of Texas at Austin described the approach as an extension of click chemistry that could offer more controllable three-dimensional organization at the conjugating agent, and noted its potential relevance to assembling fragments around a single core.

These are prospective applications, not evidence of clinical use or commercial deployment. The report does not provide a full head-to-head performance dataset against conventional azide–alkyne methods, so it does not establish that the sulfur approach is faster, more efficient, or broadly preferable.

Availability and safety context
Chemistry World reported in February 2017 that SOF4 was not commercially available at that time and that the research team was working with a Chinese company toward a ton-scale process. This is historical information, not a statement about current supply. Current commercial availability and applicable handling or regulatory restrictions are not established here; anyone considering the reagent would need up-to-date supplier and safety information.
