Personal Bioaerosol Samplers: How They Measure Airborne Exposure

Personal bioaerosol samplers collect particles in a person’s breathing zone for later analysis. The right design depends on the target, assay, collection medium, and sampling conditions.

A personal bioaerosol sampler collects airborne biological particles from a person’s breathing zone for later laboratory analysis. There is no single best sampler for every organism or test: the target, collection medium, sampling conditions, and intended assay determine which design is suitable. A collected sample is not, by itself, a pathogen identification or a measure of infection risk.

What a personal bioaerosol sampler does

A personal sampler is carried or worn to collect particles in a person’s breathing zone or personal exposure zone. Depending on its design, it deposits particles onto a filter, into liquid or a tube, onto a plate, or onto a charged surface. The collected material can then be examined using methods such as culture, microscopy, or molecular analysis.

NIOSH describes its Personal Bioaerosol Cyclone Sampler as collecting airborne particles “into relevant respiratory size and into appropriate tubes to facilitate analysis.” The collection device gathers material; the subsequent analysis determines what can be detected. NIOSH’s Health Effects Laboratory Division describes the sampler and its licensed technology.

How the main collection approaches differ

Approach How particles are collected What to check
Filter Air passes through a filter that retains particles for later analysis. Whether the filter material and handling method suit the planned assay, and whether dust could obstruct airflow.
Cyclone Airflow separates particles into a collection tube or, in some designs, a liquid film. Target particle sizes, collection medium, flow requirements, and compatibility with the intended analysis.
Impaction Airborne particles are directed onto a collection surface, such as a plate. Whether the collection surface and recovery procedure support the organisms or measurements of interest.
Electrostatic collection An electric field draws particles onto a collection surface or into a collection medium. Whether the device is a prototype or a currently available instrument, plus its demonstrated performance for the specific assay.

These categories are not interchangeable. The NIOSH Manual of Analytical Methods, Chapter BA discusses bioaerosol sampling methods and equipment. Its list of commercial samplers is non-exhaustive and explicitly does not constitute NIOSH endorsement.

How to choose a sampler for a study

  1. Define the measurement. Decide whether the study concerns viable or culturable organisms, spores, total biological material, or a molecular endpoint. These outcomes are not equivalent.
  2. Match the medium to the assay. Confirm that the filter, liquid, tube, plate, or charged surface can be processed by the laboratory method you plan to use.
  3. Check aspiration and capture. Consider whether the sampler draws in particles representative of the exposure of interest and how much of that material it retains. Physical collection efficiency alone does not establish biological recovery or viability.
  4. Confirm flow and duration. Assess the required airflow, sampling time, pump or battery needs, and whether the unit is practical to wear during the activity being studied.
  5. Account for dust and other loading. In high-particulate settings, consider the risk that a filter or collection path will clog and change sampling performance.
  6. Look for comparable field evidence. Prefer evaluations that resemble the intended environment, target, and analysis. Laboratory tests and results from a single field setting should not be generalized automatically.

What published comparisons show—and what they do not

Field comparison of filters and sampler methods

A NIOSH-hosted 2015 field evaluation found polycarbonate filters had the highest relative efficiency among the filters tested, particularly for bacteria. In that evaluation, Button and IOM samplers using those filters had the highest relative efficiencies for fungi and bacteria among the compared methods, followed by the NIOSH cyclone. The Button sampler was more prone to clogging in high-dust conditions. These are findings from that evaluation, not a universal ranking for every sampler, organism, or workplace. Read the field evaluation.

Electrostatic prototype results

A 2018 NIOSH-hosted report on the Personal Electrostatic Bioaerosol Sampler (PEBS) reported more than 80% physical collection efficiency at 10 L/min in its stated testing. Compared with the BioSampler, the prototype produced similar or higher culturable-bacteria results, lower culturable-spore results, and higher airborne ATP in the reported comparisons. The differences by endpoint matter: the physical-efficiency figure is not a guarantee of biological recovery across other conditions. See the PEBS report.

A 2016 NIOSH-hosted conference record reported 70–90% collection efficiency for tested particles under tested operating conditions, with bacterial aerosol experiments reported in the 70–80% range. Those figures describe a prototype and specific tests, not a specification for every electrostatic sampler or evidence that the device is broadly available commercially. Read the conference record.

Cyclone aspiration testing

A 2012 wind-tunnel study tested two cyclone-based personal samplers with recirculating liquid films. Using fluorescent test aerosols from 0.5 to 20 micrometres and wind speeds of 0.5 and 2.0 m/s, the study reported aspiration efficiencies that closely agreed with the ACGIH inhalable convention within the tested particle-size range. This is evidence about aspiration under those test conditions, not proof of equal biological recovery for all aerosols. Read the wind-tunnel study.

What a sampler result can tell you

A sampler provides collected material for a specified analytical method. Whether a result indicates viable organisms, biological components, or a particular organism depends on the assay and sample handling. It does not by itself identify a pathogen, establish a universal indoor-air threshold, or determine a person’s infection risk. Those conclusions require appropriate analysis and study design.

The evidence cited here does not establish a consumer-grade real-time monitor that identifies all airborne pathogens. A personal sampler should therefore be treated as part of a sampling-and-analysis workflow, rather than as a stand-alone diagnostic or health-risk device.

Before buying or specifying equipment

  • Verify the exact sampler model, collection medium, and compatible laboratory methods.
  • Check whether published performance applies to your target organism or endpoint and to conditions like your own.
  • Confirm required flow, run time, power, wearability, and maintenance needs.
  • For dusty environments, look specifically for evidence on loading or clogging.
  • Distinguish a tested prototype from a commercially available instrument; a prototype result does not establish current availability.
  • Use NIOSH’s equipment listings as a reference, not as an endorsement or complete buying guide.
