A 2010 laboratory study found that two compounds isolated from the leaves of Piper sanguineispicum showed activity against a parasite that causes leishmaniasis. The findings are an early drug-development lead—not evidence that the plant or its compounds can prevent or treat the disease in people.
What researchers found
In a paper published in 2010, Billy Joel Cabanillas and colleagues reported isolating 10 compounds from leaves of Piper sanguineispicum: three previously undescribed caffeic acid esters, four previously undescribed lignans, and three known compounds. They tested the compounds against axenic amastigote forms of Leishmania amazonensis, a parasite species associated with leishmaniasis.

Two caffeic acid esters—identified in the paper as compounds 1 and 3—had the strongest reported activity in that assay:
| Compound | Reported IC50 | What the figure describes |
|---|---|---|
| Caffeic acid ester 1 | 2.0 μM | In-vitro assay against axenic L. amazonensis amastigotes |
| Caffeic acid ester 3 | 1.8 μM | In-vitro assay against axenic L. amazonensis amastigotes |
IC50 is the concentration that inhibits a measured biological response by 50% under the conditions of an assay. These values describe laboratory activity against parasite forms grown without host cells; they do not show that the compounds work in an infected person, or establish a safe and effective dose.

- CAYENNE - THE SPICY PUNGENT ESSENCE: Firmly established in national cuisines worldwide, Cayenne pods serve as a key ingredient in spice blends, sauces, and marinades. Delivering an intense, moderately hot, and spicy flavor with a pleasant pepper aroma, they can be enjoyed fresh, dried, or ground. A tiny dose enhances any dish, offering rich nutrition and strong antioxidant properties.
- GHOST PEPPER - THE SMOKY EXTREME: Conquering global gastronomy with its fiery and complex flavor profile. It features the signature sweet taste typical of super-hot peppers, complemented by subtle fruity, citrus notes and a delicate smoky undertone. Recognized as one of the most flavorful extreme-heat peppers, it inspires entire restaurant menus and serves as a core base for hot sauces.
The paper also reports moderate cytotoxicity on murine macrophages. That finding matters because activity against a parasite is not enough for a medicine: a candidate must also be evaluated for its effects on host cells and for safety and efficacy in further studies.
Why this is not a treatment yet
The study tested isolated compounds in vitro. It did not establish that eating, handling, or applying the plant treats leishmaniasis, nor did it report a clinical trial in people. In a 2010 Royal Society of Chemistry report, study author Nicolas Fabre described the results as “very preliminary” and said in-vivo investigations were needed to confirm the esters’ potential.

The same report quoted natural-product synthesis professor Felipe Otálvaro saying the compounds had activities “more or less similar” to amphotericin. This was a contemporaneous expert reaction to early laboratory findings—not a head-to-head clinical comparison or evidence that the compounds are equivalent to amphotericin as medicines.

The species was Piper sanguineispicum, a member of the Piperaceae family. It is not culinary black pepper, Piper nigrum, and it is not a chili pepper. The Royal Society of Chemistry’s 2010 report said the Chayahuitas people in Peru used the leaves of P. sanguineispicum as an anti-inflammatory. That traditional-use account helps explain why researchers examined the plant; it does not show that the plant treats leishmaniasis.

When was the study published, and what happened next?
The paper, “Caffeic Acid Esters and Lignans from Piper sanguineispicum,” appeared online on October 18, 2010, and in the November 29, 2010 issue of Journal of Natural Products (volume 73, issue 11, pages 1884–1890; DOI 10.1021/np1005357). The Royal Society of Chemistry’s news report followed on October 27, 2010.
The cited sources establish the initial laboratory findings, but do not establish whether the compounds later advanced to animal efficacy studies, clinical trials, or an approved medicine. The specific chemical names and structures of esters 1 and 3 are also not provided in the abstract-level study information cited here.
