“Killing three parasites with one stone” refers to a 2012 report on a laboratory-stage molecule that joined the antimalarial drug amodiaquine to a nitric-oxide-releasing furoxan group. The hybrid showed activity in assays involving malaria, schistosomiasis and hookworm parasites, but the work did not establish that it is safe or effective in people.
What the phrase refers to
The phrase was the title of a Royal Society of Chemistry Medicinal Chemistry Blog post by James Anson, published on 19 October 2012. The post discussed a paper by Bryan T. Mott and colleagues, “A furoxan–amodiaquine hybrid as a potential therapeutic for three parasitic diseases,” published in MedChemComm in December 2012. Read the paper.

The researchers covalently joined amodiaquine, an antimalarial drug, with a furoxan group designed to release nitric oxide. The aim was to combine activity through more than one mechanism in a single molecule. The paper describes the hybrid as a potential therapeutic, but that wording is a research hypothesis—not evidence that it became a medicine.

Which parasites were studied

These findings are not interchangeable: in vitro assays test effects under laboratory conditions, while an ex vivo assay examines material outside a living host. Neither demonstrates that a treatment works in an infected person.

What the hookworm experiment found
For the adult-worm survival assay, the paper reports testing hybrid 15 at 100 μM for 120 hours. The researchers observed a greater percentage of worm death than with the albendazole control; the difference was statistically significant at 72 hours (P < 0.05). This is a result from that specific ex vivo experiment, not evidence of human benefit or a recommended dose.
What the study does—and does not—show
The paper reports in vitro and ex vivo activity. It does not report clinical efficacy or establish safety in humans. Its conclusion describes the hybrid as an early lead for further structure–activity exploration, with the possibility that additional chemical work could improve activity. The reported results therefore support continued laboratory investigation, not use as a treatment.

The paper’s introduction cited a 2012 World Health Organization estimate that more than 400 million people were infected with malaria, schistosomiasis or hookworm. That is a historical figure as reported in the paper, not a current prevalence estimate.
Do not confuse it with the 2016 “three diseases” story
A similarly worded Novartis story published on 1 December 2016 described a separate experimental compound studied against parasites associated with Chagas disease, African sleeping sickness and leishmaniasis. That was a different project, compound and disease set. Its findings—including the story’s report of work in mice—should not be attributed to the 2012 furoxan–amodiaquine study.

What is known about the hybrid now
The 2012 paper establishes the reported laboratory findings and the authors’ call for more medicinal-chemistry work. The cited sources do not establish the hybrid’s clinical or development status as of 7 October 2026. It would be inaccurate to say, on this evidence, that it advanced into clinical use, failed development or is available as a medicine.
