A free scan shows the junk files, broken settings and background clutter dragging Windows down - then fixes them in one click.Free scan · Windows 10 & 11To check whether a microscopy image has changed since a known point, compare its current file checksum with a trusted checksum recorded earlier. To assess whether it faithfully represents an experiment, you also need the original acquisition files, metadata, instrument records, and experimental context. Neither a matching checksum nor plausible metadata proves that an image is authentic.
What “image integrity” can—and cannot—mean
There are two different questions behind an image-integrity check:
As an Amazon Associate I earn from qualifying purchases.
- Have the file’s bytes changed since a known checkpoint? A cryptographic checksum, or digest, can answer this when you have a trusted digest for comparison. It detects a later difference in the file; it does not explain why the file differs.
- Does the image faithfully represent the acquisition and experiment? That requires examining the original data, metadata, processing history, instrument and acquisition records, and experimental context. A checksum cannot authenticate how an image was created, and metadata can be incomplete or inaccurate.
For scientific work, preserve the original acquisition files and assess any processed or published image against them. A discrepancy is a reason to investigate, not proof by itself of falsification or intent.
Do these 3 things before closing this tab:
1Repair Windows errors before they cause bigger problems2Fix the driver behind crashes, sound loss and screen glitches3Clear out junk files and repair common Windows errorsPreserve the original acquisition record
Copy the complete microscope file set to managed storage before viewing, converting, or editing it. Retain the unaltered source and work from a separate copy. Include sidecar files, acquisition logs, and vendor project files: relevant metadata may be stored outside the image file. U.S. Department of Health and Human Services Office of Research Integrity (ORI) guidance states: “Manipulations of digital images should always be done on a copy of the raw image data. The original must be retained.” ORI also advises against discarding files that contain metadata and recommends a second copy because storage media can fail.

Follow your institution’s storage and backup policy. Where appropriate, keep a separate backup and periodically verify stored copies against a trusted reference digest or another approved preservation mechanism. Record the digest and the date or checkpoint it represents in a way that remains associated with the preserved file. Comparing against a reference is useful only if that reference is itself trustworthy; the comparison establishes byte-level consistency from that checkpoint, not authenticity at acquisition.
Inspect metadata against the experiment
Use the microscope’s native software where available, or a compatible reader such as Bio-Formats in Fiji. Review the fields relevant to the experiment, then compare them with acquisition logs, instrument records, and the methods description. Treat missing, uncertain, or conflicting values as points to investigate; they are not automatic evidence of misconduct.

Fields to review
- Image structure: width and height, pixel type or bit depth, and the number and arrangement of channels, Z planes, and time points.
- Spatial information: pixel calibration and units, including whether dimensions and scale make sense for the specimen and acquisition.
- Acquisition details: date and settings, microscope and detector information, and other recorded parameters relevant to the modality.
- Annotations and context: experiment notes, regions of interest, and any identifiers needed to connect the file to the sample or acquisition record.
OME’s data model represents image parameters such as XYZ dimensions and pixel type, as well as acquisition, annotation, and region-of-interest metadata. The relevant fields depend on the experiment: not every field applies to every imaging method. A complete-looking metadata record is useful for interpretation and reproducibility, but does not prove that the pixels are authentic.
Choose a format without losing the source
Bio-Formats can read and write many life-sciences image formats and map metadata into the OME data model. OME-TIFF stores image planes in TIFF and embeds an OME-XML metadata block in the TIFF header. These options can help labs exchange data, but conversion is not a substitute for retaining the original vendor files or their companion metadata.

| Option | Useful for | What to verify or preserve |
|---|---|---|
| Original vendor file set | Preserving the acquisition record, including proprietary details and companion files. | Keep the native files, sidecars, logs, and project files together. Confirm that the receiving lab has software capable of opening them. |
| Bio-Formats | Reading or writing many life-sciences formats and translating supported metadata into the OME data model. | Check support for the instrument format and compare the converted result with the source. Record the Bio-Formats version and conversion settings. |
| OME-TIFF | Exchanging TIFF image planes together with an embedded OME-XML metadata block. | Confirm that dimensions, planes, channels, calibration, and relevant metadata survived conversion. Do not assume every proprietary field has an equivalent in the destination. |
After conversion, compare image dimensions, channels and planes, physical calibration, and metadata with the original; compare pixel values where possible. Preserve the source alongside the converted file, and record the tool version and conversion settings so another lab can understand what changed. Software and format support evolve, so check the official documentation for the version you plan to use rather than treating a documentation version as the current software release.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Review processing and publication figures
Keep a reproducible record of the input files, software, operations, and parameters used to produce an analysis or figure. Compare the final figure with its source data, preferably with an independent reviewer for high-stakes work. Global display adjustments should preserve the represented information and comply with journal and institutional policies. Avoid selective edits that add, remove, move, or obscure scientific features, including local enhancement, cloning, and erasure. Avoid lossy compression for scientific source data because it can alter pixel values and introduce artifacts.

If a figure combines separate acquisitions, label and visually separate the panels so the boundaries are clear. Retain the exact displayed figure as well as its source images and processing record. ORI’s image guidance emphasizes that interpretation of suspected discrepancies depends on original data and context; an observed difference alone does not establish falsification or intent.

For a figure or dataset, keep the source images, relevant metadata, analysis workflow, and final displayed figure together. Provide clear scale information and annotations, and describe relevant imaging and analysis methods. Make originals or suitable losslessly compressed data available when policy, consent, privacy, licensing, and repository arrangements allow. Community-developed image checklists emphasize clear formatting and annotation, data availability, and reporting image-analysis workflows to support interpretation and reproducibility.
When choosing a storage approach, follow institutional requirements and consider redundancy, access control, durability, and whether copies can be checked periodically. A backup copy supports preservation, but does not replace retaining the original acquisition record or documenting the workflow.
How to interpret a mismatch
A changed checksum means the file differs from the one represented by the trusted reference digest; it does not identify the cause. Metadata can be absent, incomplete, transformed during conversion, or inconsistent with records. Forensic screening can flag discrepancies or patterns for investigation, but scientific interpretation requires the original data, relevant technical expertise, and knowledge of the experiment and instrument. Follow up by checking the preserved source files, acquisition records, conversion or processing history, and experimental context before drawing conclusions.
