How to Tell Whether a Microscopy Video Was Altered or Generated With AI

No visual tell can prove a microscopy video is authentic or AI-generated. Learn how to preserve native files, compare raw acquisition data, inspect frame continuity and processing, and escalate responsibly when the evidence matters.
Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

There is no visual trick, metadata field, hash, or consumer AI detector that can prove a microscopy video is authentic or synthetic. A defensible answer comes from preserving the earliest native recording and raw acquisition data, documenting the microscope and experiment, and comparing the questioned copy with the original frames, metadata, and processing history.

Unexplained cuts, duplicated structures, inconsistent motion, missing metadata, or a detector score can justify a closer examination. None is conclusive by itself: microscopy artifacts, focus changes, sample movement, software processing, compression, and export conversions can create similar appearances. If the video could affect a publication, clinical decision, research-integrity case, or legal dispute, a qualified scientific-image or media-forensics examiner should review the original files and experiment context.

Start by separating integrity from authenticity

Two questions are often confused:

  • Integrity: Is this file complete and unchanged since the version being examined was created or acquired?
  • Authenticity: Does the recorded sequence accurately represent a real sample, event, and experiment?

A cryptographic hash can show that a copy matches the file that was hashed. It cannot show that the microscope scene was real, that the file was acquired from the claimed instrument, or that the scene was not fabricated before hashing. The UK Forensic Science Regulator describes integrity verification as confirming that data are complete and unaltered since acquisition, while authentication concerns whether the data accurately represent events. Keep those conclusions separate.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The Office of Research Integrity (ORI) states: “Authentication of a scientific image requires access to the original data.” A reposted, screen-recorded, or heavily recompressed clip may be useful as a lead, but it is a weak foundation for a final conclusion.

Preserve the best available evidence before inspecting it

Every conversion can discard information. Before opening a file in an editor, stabilize the evidence set:

  1. Obtain the earliest native video. Ask the person or facility that captured it for the camera’s native sequence, the microscope’s acquisition project, and the raw frames or image stack. Do not re-export the only copy to make it easier to view.
  2. Create a master and working copies. Store the received files read-only or in controlled storage. Analyze duplicates, not the master. Record the file name, size, hash if one is available, date received, source, and every person or system that handled it.
  3. Preserve proprietary acquisition files. Raw microscopy projects can hold instrument settings, timestamps, channel information, stage positions, and other metadata that disappear during export. Keep the original project alongside exported videos.
  4. Record every conversion. Note the software, version, export settings, codec or container, frame rate, resolution, color treatment, and whether the file was uploaded to a platform that recompressed it.
  5. Save the surrounding records. Retain microscope and camera identifiers, acquisition software logs, experiment dates, sample labels, laboratory notebooks, and related views or time points.

ORI recommends retaining original data and performing manipulations on copies. A separate external drive can provide additional storage for a master and working set, but storage itself does not authenticate a video.

  • Who operated the microscope and camera?
  • What microscope model, detector, camera, and acquisition software were used?
  • When and where was the sequence acquired?
  • What objective, magnification, illumination, channels, exposure, gain, frame interval, z-step, and temperature or environmental conditions were used?
  • Was the sample moved, refocused, relabeled, or imaged in more than one session?
  • Which files are raw, which are processed, and which are presentation exports?
  • Were denoising, deconvolution, stabilization, interpolation, false coloring, cropping, contrast changes, or other operations applied?

Metadata can support this account, but it is not a trusted witness by itself. Fields may be removed by export software, rewritten by a later editor, or absent from a legitimate capture. An empty metadata block raises a provenance question; it does not prove fabrication.

Inspect continuity from frame to frame

Play the sequence at normal speed, then step through individual frames. Look for changes that have no plausible acquisition or biological explanation:

  • Hard cuts or jumps in a supposedly continuous time-lapse.
  • Repeated cells, organelles, particles, scratches, or background patterns in different locations.
  • Structures that vanish and reappear without motion, bleaching, occlusion, or a stated intervention.
  • Sudden changes in background texture, illumination, focus, scale, color balance, or noise pattern.
  • Motion that reverses, accelerates, or freezes at a boundary between frames.
  • Frame-rate irregularities, duplicated frames, missing intervals, or a change in image dimensions.
  • A stage, sample, or field of view that shifts while every feature remains unnaturally fixed, or features that move while the background remains inconsistent.

These observations identify a discrepancy, not an intent. A researcher may acquire separate segments, refocus, change illumination, move the sample, or join approved views. Compression and playback software can also make a clean transition look abrupt. Compare each apparent break with instrument logs, acquisition intervals, and the stated protocol.

Examine image structure, not just what the scene depicts

At enlarged scale, check whether texture, shading, color, focus, and boundaries behave consistently across the frame:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Texture and noise: Genuine sensor noise and optical texture usually vary with exposure and local signal. A patch with a different noise character may have been inserted, denoised, or generated separately.
  • Edges and boundaries: Composited regions can have halos, unnaturally sharp borders, mismatched blur, or a different point-spread appearance from neighboring structures.
  • Shading and illumination: A clone or generated object may not follow the illumination gradient, shadow direction, or depth-dependent brightness seen elsewhere.
  • Color and channels: Channel registration, false color, and bleed-through should be consistent with the instrument and stated processing. A color change can be legitimate if a channel or lookup table changed, so seek the acquisition settings.
  • Focus and resolution: Inserted or enhanced detail may be sharper, softer, or more uniformly resolved than structures acquired in the same optical plane.
  • Repetition: Cloned motifs, mirrored shapes, or identical debris patterns are worth documenting, but repeated biological structures can occur naturally and image processing can duplicate pixels accidentally.

General forensic guidance treats these as examination categories, not microscopy-specific proof rules. ORI cautions that first-order image examinations are not foolproof. A meaningful finding requires comparison with raw frames and knowledge of the imaging method.

Trace every processing step

Ask for the complete processing history, including operations that were considered cosmetic:

  • Filtering, denoising, deconvolution, sharpening, or background subtraction.
  • Interpolation, frame generation, motion stabilization, or temporal smoothing.
  • Contrast, brightness, gamma, threshold, or whole-image color adjustments.
  • False coloring, channel recombination, projection, cropping, or resizing.
  • Splicing of fields of view, time points, channels, or separately acquired sequences.
  • AI-assisted enhancement, inpainting, object removal, or synthetic frame generation.

A processed visualization can be scientifically useful if the operation is disclosed and the unprocessed source remains available. ORI guidance treats digital scientific images as data: retain the original, work on copies, disclose processing, and avoid operations that change the scientific meaning. It describes simple whole-image adjustments and cropping as usually acceptable when they do not misrepresent the data, while cloning is highly questionable. Filters are generally discouraged for biological images when they obscure or create structure.

Do not treat the presence of any edit as proof of misconduct. The relevant questions are what changed, whether it was disclosed, whether the raw data support the displayed claim, and whether the operation could alter interpretation.

Account for compression and export damage

Compare the highest-quality available source with every reposted or platform copy. Lossy compression, resolution reduction, sharpening, and format conversion can create block boundaries, ringing, mosquito noise, softened edges, and apparent discontinuities. A screen recording may also change frame timing and color.

Compression is an explanation to test, not a reason to dismiss an anomaly automatically. If a suspicious duplication or discontinuity occurs exactly in a scientifically important feature while surrounding regions remain clean, preserve both versions and have an examiner assess it. ORI warns that compression can affect interpretation and that converting raw data can lose information.

ORI’s archived guidance identifies TIFF as a lossless option for still images and warns that JPEG is lossy and unsuitable for most scientific images. That still-image advice should not be misread as a recommendation for a particular video codec; for video, retain the native acquisition sequence and document every export.

Understand what metadata and hashes can prove

Signal What it can establish What it cannot establish
Matching cryptographic hash The examined copy matches the file that was hashed. That the depicted microscopy event was real or that the hashed file was honestly acquired.
Camera, microscope, or software metadata Some consistency with a claimed acquisition workflow. That the fields were not edited, removed, or copied from another file.
Missing or inconsistent metadata A reason to investigate provenance and export history. Standalone proof that the video is fabricated.
Visible duplicate or discontinuity A documented discrepancy requiring explanation. Intent, misconduct, or an AI origin without contextual review.
AI-detector score At most, a lead for specialist analysis when the method is validated for comparable data. A universal “real” or “AI” verdict for microscopy footage.

Hash the preserved master and each working copy separately if your procedure calls for it. Record the algorithm and the exact file to which it applies. A hash becomes meaningful only with a documented chain of custody and a clear statement of what was hashed.

Do not overinterpret automated AI detection

NIST separates manipulation detection and localization from deepfake detection as different media-forensics tasks. That distinction matters: a system designed to flag a face-swap benchmark is not automatically validated for fluorescence time-lapse, electron microscopy, phase contrast, or volumetric projections.

No validated accuracy figure for arbitrary microscopy videos is established here. Do not convert an unvalidated probability or label into certainty, and do not assume that a “real” result clears a clip. Compression, denoising, interpolation, unusual optics, low light, repeated textures, and legitimate false coloring can all move a microscopy video outside a detector’s training distribution.

Use automated output as triage:

  1. Preserve the original and document the detector version, settings, input file, and output.
  2. Ask whether the method was validated on the same microscopy modality, file type, resolution, frame rate, and processing pipeline.
  3. Check whether it localizes a suspected region and explains the basis of its finding.
  4. Have a qualified examiner review the result alongside raw data and instrument records.

The UK Forensic Science Regulator’s code treats automated scanning as an indication that specialist analysis may be needed, with authenticity conclusions treated as expert opinion rather than an automatic label.

Compare the questioned clip with independent records

The strongest comparison is not video versus video; it is the questioned presentation versus independent acquisition evidence:

  • Raw frames or the native time-lapse sequence.
  • Other fields of view, channels, z-planes, or time points from the same experiment.
  • Microscope and camera logs, stage coordinates, exposure records, and software project files.
  • Laboratory notes, sample identifiers, and records from collaborators.
  • Replicate acquisitions made under the same protocol.

Look for explanations that predict the observed change. A focus adjustment should affect sharpness in a characteristic way; a stage movement should shift the field coherently; a channel change should follow the acquisition record; a recompression should affect broad regions rather than one biologically important object. These are hypotheses to test, not universal rules.

Know when to escalate

Seek an independent scientific-image-integrity or digital-media-forensics review when:

  • The video supports a publication, patent, grant, clinical decision, safety claim, or regulatory submission.
  • The original native files or acquisition records are unavailable, disputed, or incomplete.
  • A suspected edit changes the apparent biological result or quantitative conclusion.
  • Different versions conflict and the chain of custody matters.
  • The dispute may lead to a formal research-integrity or legal proceeding.

Choose an examiner who can demonstrate experience with the relevant microscopy modality and media-forensics methods. Ask for the validation data, expected false-positive and false-negative behavior on comparable material, ability to localize and explain findings, treatment of compression and legitimate processing, practitioner qualifications, reproducibility, and chain-of-custody procedures. A generic image analyst may not understand acquisition artifacts; a microscopy specialist may not have validated forensic methods.

Use a disciplined observation record

For each finding, record the frame number or time, region of interest, file version, display conditions, and a neutral description. For example: “Frames 184–196 show a background boundary and a repeated bright feature at coordinates X–Y; the same feature is absent in the native sequence.” Keep the observation separate from an interpretation such as “the object was cloned.” Attach the raw and working-file hashes, screenshots only as illustrations, and the software used to inspect them.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

This separation prevents a discrepancy from being promoted prematurely into an allegation. ORI puts the point plainly: “The identification of a discrepancy is only the allegation, and it does not by itself demonstrate an intent to falsification of data.”

A practical decision path

  1. Only a social-media or presentation copy exists: Preserve it, obtain the source from the creator or facility, and treat visual findings as provisional.
  2. Native files exist but metadata is incomplete: Compare frames with instrument logs and related acquisitions; missing fields alone do not decide authenticity.
  3. A visible anomaly has a plausible acquisition explanation: Test that explanation against settings, timing, and raw data, then document the result.
  4. An anomaly remains unexplained and affects the scientific claim: Stop editing the files and refer the complete evidence set to a qualified examiner.
  5. An automated tool reports “AI” or “real”: Preserve the exact input and output, check method validation for the microscopy material, and treat the result as a lead only.

What a responsible conclusion sounds like

A careful report states the scope and limits: which file was examined, what original data were available, what processing was documented, which discrepancies were observed, which explanations were tested, and what remains uncertain. It should distinguish “the exported copy differs from the native sequence” from “the biological event was fabricated” and from “AI generation was used.” Those are different claims requiring different evidence.

Without original data, instrument context, and a validated method, the most accurate conclusion may be that authenticity cannot be determined from the available copy. That is a legitimate forensic outcome, not a failure to choose between “real” and “AI.”
