Conventional circular dichroism (CD) measures the difference in extinction or absorption for left- and right-circularly polarized light from an ensemble. Circular differential scattering (CDS) adds dark-field microscopy and polarization-resolved spectra to measure that difference from an individual scattering nanoparticle. CD microscopy measures single-object extinction, while photothermal CD measures differential heat generated by absorption. None of these signals is automatically a map of local optical-chirality density.
The practical workflow is therefore: define the signal, decide whether an ensemble or one object is required, stabilize and calibrate the polarization optics, control orientation and illumination geometry, and use achiral and linear-dichroism controls before assigning a signal to intrinsic structural chirality.

What “optical chirality” measurement actually means
Optical chirality is not one universal instrument channel. In nanophotonics, researchers use several related observables:
- Ensemble CD: the difference in absorption or extinction between opposite circular polarization states, normally reported as a spectrum from many particles.
- Single-particle CDS: the difference between scattering spectra recorded from one nanoparticle under opposite circular polarization states.
- CD microscopy: a spatially localized extinction measurement that combines scattering and absorption contributions.
- Photothermal CD: a differential heat-generation signal associated with absorption.
- Near-field, electron, emission, or vibrational methods: measurements of localized fields, electron-coupled loss, circularly polarized luminescence, or vibrational optical activity rather than ordinary far-field CD.
A positive or negative differential signal is meaningful only with its observable, polarization convention, geometry, and normalization stated. A far-field CD or CDS spectrum should not be described as a direct spatial image of optical-chirality density.

The 2017 review by Smith, Link, and Chang surveys the progression from ensemble polarimetry to nonlinear, near-field, and single-particle methods. The 2026 review by Li and Renero-Lecuna focuses on CDS fundamentals, instrument configurations, artifacts, and alternatives.
How ensemble circular dichroism works
In a conventional CD experiment, a spectrometer illuminates the sample with left- and right-circularly polarized light and records the difference in the resulting optical response. Depending on the instrument and sample presentation, that response is treated as differential absorption or extinction. Cuvettes, colloids, films, and other ensemble formats can be measured efficiently, which makes CD useful for screening a population or following a spectral feature across a preparation.
What an ensemble spectrum includes
The spectrum is an average over every particle in the illuminated volume. Size dispersion, shape variation, random or biased orientations, aggregation, substrate interaction, and mixtures of opposite handedness all contribute to the measured curve. If right- and left-handed objects occur in equal proportions, their chiroptical signals can cancel even though each particle is strongly chiral. A weak ensemble result therefore does not prove that individual objects are achiral.
When a commercial CD result can be interpreted directly
A 2022 review on single plasmonic nanostructures cautions that treating a commercial CD spectrometer output as a nanoparticle CD spectrum is justified only under restrictive conditions. The authors specifically discuss objects typically below 30 nm in diameter and random orientation. That figure is a reported interpretive condition from the review, not a universal size limit for every instrument, particle, or sample geometry. Larger particles, anisotropic aggregates, oriented films, and samples with substantial scattering require an explicit optical model or a different measurement strategy.
How to measure one nanoparticle with circular differential scattering
CDS is the most direct route when the question concerns the scattering response of an individual nanoparticle or nanostructure. It combines a dark-field microscope with circular-polarization-resolved scattering spectroscopy. Instead of measuring an ensemble extinction signal, the system isolates one bright scattering object against a dark background and compares its spectra under opposite circular polarization states.

1. Prepare and locate isolated objects
- Immobilize or sparsely deposit the particles on a substrate that gives adequate mechanical stability and an acceptably low optical background.
- Use dark-field illumination so the direct illumination is excluded from the collection path while scattered light from the particle enters the objective.
- Locate an isolated scatterer and record its position, focus, substrate region, and acquisition settings. Avoid changing focus or moving to a different field between polarization states.
Dark-field systems commonly use a hollow cone of illumination. The condenser, objective numerical aperture, illumination angle, and collection geometry vary by design, so there is no single universal CDS layout.
2. Generate the two circular polarization states
One documented excitation arrangement places a linear polarizer and a broadband quarter-wave plate before the sample. Rotating the relative orientation of these elements generates the two opposite circular states. The quarter-wave plate must provide the intended retardance over the complete measurement band; a component specified at one wavelength can become a source of ellipticity elsewhere in the spectrum.
Another arrangement analyzes circular components in the detection path. Simultaneous two-channel detection can reduce errors caused by alternating measurements, but the channels must be calibrated for unequal throughput, detector response, and spectral sensitivity. Alternating illumination states are simpler, yet they are more vulnerable to drift, particle motion, focus change, and lamp fluctuations.
3. Acquire matched scattering spectra
- Measure the background at a nearby location or with the particle excluded, using the same polarization state and optical settings.
- Record the scattering spectrum for one circular polarization.
- Switch only the polarization state, keeping the particle position, focus, illumination power, integration time, and collection path fixed.
- Record the opposite-polarization spectrum, then repeat the pair if signal-to-noise or drift assessment requires it.
- Subtract backgrounds consistently and calculate the differential scattering using a stated convention, such as the spectrum for left circular polarization minus the spectrum for right circular polarization.
Report whether the result is an absolute difference, a normalized difference, or an asymmetry-like quantity. The normalization must be defined because different conventions can change the numerical scale and sign presentation without changing the underlying spectra.
4. Verify the polarization state across the spectrum
Measure or model the actual polarization delivered to and collected from the sample, rather than assuming that a quarter-wave plate is ideal. Retardance errors, polarizer extinction, objective birefringence, mirrors, windows, and wavelength-dependent transmission can leave residual linear or elliptical polarization. Characterize the optics over the band used for the particle spectrum and document the orientation of every polarization element.

Controls that separate chirality from artifacts
Use an achiral reference
Measure an achiral particle or structure with comparable size, material, substrate, and scattering strength. A nonzero differential signal from that reference indicates instrumental asymmetry, imperfect polarization, alignment error, or geometry-induced effects that must be addressed before interpreting a chiral sample.
Check linear dichroism and linear birefringence
An anisotropic particle can respond differently to two orthogonal linear polarization components. Imperfect circular polarization can convert that linear response into an apparent circular differential signal. Rotate the sample or polarization basis where possible, measure linear-polarization responses, and include the resulting contribution in the uncertainty or model. A CDS peak that changes strongly with azimuthal orientation is not automatically evidence of intrinsic handedness.
Control illumination and collection symmetry
An achiral object illuminated obliquely from one direction can become extrinsically chiral as a combined object-plus-illumination system. The 2026 CDS review notes that illumination from all directions can cancel this extrinsic contribution in the discussed setup. In practice, vary the incidence direction, reverse the sample or illumination geometry when feasible, and report the angular arrangement. A signal that reverses or disappears under a geometry change may be extrinsic rather than a property of the object alone.
Control substrate, focus, and drift
- Measure particles on the actual substrate, because an interface can alter modes, polarization, and radiation patterns.
- Keep the particle centered and in focus for both states; a small lateral displacement can change the collected scattering.
- Monitor lamp intensity and detector drift during the pair.
- Repeat measurements on several nominally identical particles to distinguish a reproducible population feature from one-object variability.
- Record whether the particle is immobilized and how its orientation was established; an immobilized object is not equivalent to a randomly oriented ensemble.
What each method can and cannot tell you
CD versus CDS
CD and CDS both compare opposite circular polarization states, but they do not measure the same signal. CD in a transmission or extinction configuration can contain absorption and scattering. CDS is explicitly a scattering difference from a selected object. A CDS spectrum cannot be substituted for an ensemble extinction spectrum, and an ensemble CD null cannot rule out strong single-particle scattering chirality.
CD versus photothermal CD
Photothermal CD detects differential heat generation. It is therefore tied to how the object absorbs and converts optical energy, whereas CDS responds to the scattered field. Two particles can have similar photothermal CD but different CDS, or vice versa, because absorption and radiation are separate channels.

CD microscopy
CD microscopy localizes an extinction response to individual objects in a microscope image. It is useful when spatially identifying objects matters, but the measured extinction still combines scattering and absorption. Pair it with independent scattering or absorption information if the physical origin of a feature is important.
Near-field and electron-based mapping
Near-field optical microscopy can probe localized fields with nanoscale spatial resolution. Electron energy-loss spectroscopy (EELS) and cathodoluminescence provide electron-coupled information and can access modes that are not resolved in ordinary far-field spectra. A primary 2020 Nano Letters study used a nanometer-sized focused electron beam scanned across an optically excited chiral nanostructure; electron energy spectra acquired at successive positions produced a high-spatial-resolution map of near-field dichroism. This is a specialized mapping experiment, not a replacement for routine ensemble spectroscopy.
Emission and vibrational methods
Circularly polarized luminescence (CPL) compares emitted light in opposite circular polarization channels, usually under optical excitation. Surface-enhanced Raman optical activity (SEROA) and related Raman methods address vibrational optical activity. Hyper-Rayleigh scattering optical activity and circular differential photocurrent mapping answer still different nonlinear or device-level questions. Select them only when emission, vibration, nonlinear scattering, or photocurrent is the signal of interest.

- State the polarization convention and define which spectrum is subtracted from which.
- Show raw or background-corrected spectra for both polarization states, not only the differential curve.
- Describe the optical path, including polarizer and wave-plate locations, objective and condenser geometry, illumination angle, numerical apertures, detector channels, and substrate.
- Give wavelength range, spectral resolution, integration time, incident power, and whether the states were recorded alternately or simultaneously.
- Explain polarization calibration, channel balancing, background subtraction, and drift correction.
- Report particle size, shape, material, orientation or immobilization method, and the number of individual objects measured.
- Include achiral references and any tests for linear dichroism, sample rotation, incidence-angle dependence, or substrate effects.
- Separate instrumental uncertainty, particle-to-particle variation, and model uncertainty. Do not present a review-reported detection limit as an expected performance guarantee.
The 2026 review reports CDS detection limits as low as 1.5% and notes more common values around 10%, but the available review passage does not establish a protocol-independent sample basis. Treat those figures as literature context, not as a specification your setup will necessarily achieve.

Common failure modes and fixes
The differential spectrum is large from an achiral control
Check polarizer extinction, wave-plate retardance, objective and mirror birefringence, channel balance, and illumination centering. Repeat with the particle removed to measure the optical background. If the effect follows the instrument rather than the sample, correct the calibration before measuring chiral structures.
The signal changes when the particle is rotated
Test for linear dichroism and extrinsic chirality. Record the orientation, incidence direction, and collection geometry for every spectrum. A rotation-dependent response may be real for the complete measurement geometry, but it should not be labeled intrinsic structural chirality without additional symmetry controls.
The two spectra do not overlay outside the resonance
Look for lamp drift, focus change, particle motion, detector saturation, or unequal integration times. Acquire polarization states in rapid succession or use simultaneous channels, then calibrate each channel independently. Recheck background subtraction at wavelengths where the particle scatters weakly.

Inspect the orientation distribution, size dispersion, aggregation, and handedness balance. Opposite-handed populations can cancel. Strong scattering can also make a simple absorption interpretation inappropriate; consider CDS, CD microscopy, or an optical model that includes extinction components.
A far-field signal is being used as a local-field map
Do not infer a spatial distribution of optical-chirality density from one far-field CD or CDS number. Use near-field microscopy, EELS, cathodoluminescence, or another spatially resolved method when the claim concerns nanoscale localization.
A practical decision path
- Define the observable: absorption, extinction, scattering, heat, emission, vibration, photocurrent, or local near field.
- Define the sampling scale: ensemble average, one particle, or a spatial map across one structure.
- Choose the matching family: CD for efficient ensemble response, CDS for one scatterer, CD microscopy for localized extinction, photothermal CD for absorption-linked heat, and near-field or electron methods for local mapping.
- Specify geometry before acquisition: illumination direction, collection angles, substrate, orientation, and whether the arrangement can generate extrinsic chirality.
- Calibrate and control: characterize polarization optics over wavelength, balance channels, measure backgrounds, and test achiral and linear-dichroism controls.
- Interpret conservatively: identify the measured signal in the title, caption, and analysis; distinguish intrinsic structure from object-plus-illumination effects; and avoid turning a review caveat into a universal threshold.
Minimum equipment and sample requirements
- A stable spectrometer and detector covering the resonances of interest.
- For CDS, a dark-field microscope with a condenser/objective arrangement that rejects direct illumination while collecting scattered light.
- Linear polarizers and a broadband quarter-wave plate, or a calibrated polarization analyzer in the detection path.
- A mechanically stable sample mount and a substrate with characterized optical background.
- Software or procedures for background subtraction, channel balancing, spectral registration, and uncertainty estimation.
- Achiral reference structures and, where relevant, a way to rotate the sample or vary illumination direction.
- For local mapping, the specialized near-field, electron-beam, or cathodoluminescence platform appropriate to the chosen signal.
The most defensible result is not the largest CD-like peak. It is a reproducible differential response whose measured channel, polarization state, geometry, calibration, and controls are all explicit.
