How Strained Molecules Make a Radical Reaction Work Without an Initiator

A proposed diradical pathway lets two highly strained carbon frameworks couple to form functionalized bicyclo[2.1.1]hexanes without an external radical-generating stimulus.

A 2025 reaction joins a transient cyclic allene with a bicyclo[1.1.0]butane (BCB) to build a bicyclo[2.1.1]hexane (BCH). The unusual part is its proposed mechanism: the severely distorted reactants have inherent diradical character, allowing a σ-bond insertion pathway without an external radical-generating stimulus such as light or an initiator. The authors describe a proposed pathway, not proof that every member of these molecule classes behaves identically.

What reacts, and what does the reaction make?

The reaction couples two highly strained carbon frameworks. One partner is a cyclic allene, a ring containing a carbon–carbon–carbon unit with two adjacent double bonds. Because cyclic allenes are unstable, the reported method generates one in the reaction mixture rather than isolating it first. The other partner is a bicyclo[1.1.0]butane, a compact four-carbon structure with two bridgehead atoms joined by a short, strained bond.

The coupling produces a functionalized bicyclo[2.1.1]hexane: a six-carbon framework arranged in a bridged, three-dimensional shape. The Nature paper by Tena Meza and colleagues reports these products as a way to access BCH scaffolds from the two strained fragments. Read the Nature paper.

How can a radical pathway proceed without light or an initiator?

In many radical reactions, light, heat, a catalyst, or a chemical initiator is used to create radical species. Here, the authors attribute the reactivity to the substrates themselves: severe geometric distortion gives the cyclic allene and BCB diradicaloid character. In other words, their electronic structures are proposed to make a diradical pathway accessible without first using an external stimulus to generate radicals.

#1 Best Overall

“Diradical character” is a careful description, not a claim that every molecule in the mixture is a freely diffusing pair of radicals. The paper proposes σ-bond insertion through a diradical pathway; the strength and role of that character can depend on the particular structures and conditions.

The reported sequence

Chemistry World’s account says the team generated the cyclic allene in situ from a silyl triflate precursor, and that it reacted with the BCB at room temperature. Its summary of the team’s computational analysis describes attack at a BCB bridgehead, cleavage of the bridgehead bond to form a diradical intermediate, and recombination that creates the second carbon–carbon bond. These are mechanistic details attributed to the computational proposal, rather than a sequence directly established for every possible substrate. Chemistry World’s report.

What does strain contribute?

Strain is stored energy associated with a molecule’s constrained geometry and bonding. The Nature article gives approximate strain energies of about 30 kcal mol−1 for cyclic allenes and about 60 kcal mol−1 for BCBs. These are literature-grounded approximate values, not exact measurements that apply to every substituted example.

When the strained starting structures reorganize into a less constrained product, releasing strain can help drive the transformation. The authors’ interpretation is that distortion also contributes to the reactants’ diradicaloid character, linking the reaction’s unusual electronic pathway with the energetic cost of their geometries.

Why make bicyclo[2.1.1]hexanes?

BCHs are three-dimensional, saturated frameworks that have drawn interest as aromatic bioisosteres: structures chemists may explore as alternatives to flat aromatic rings in drug-discovery molecules. A method that installs functionalized BCHs can therefore expand access to molecular shapes researchers may want to investigate.

That is a synthetic opportunity, not evidence that this reaction has produced an approved medicine, a clinical benefit, or a drug candidate. The paper demonstrates access to a scaffold; any biological value would require separate evaluation.

What is established—and what requires the full paper?

The Nature article describes the conditions as mild and the protocol as operationally simple. Its supplementary information, linked from the paper, contains experimental procedures, characterization, and computational methods and data. Those materials are needed to assess exact reagents and equivalents, yields, substrate-by-substrate scope, and reproducibility.

  • The accessible abstract and specialist news account do not establish that every cyclic allene or BCB will react.
  • They do not support a complete scope table or a general yield claim.
  • They do not establish that this route outperforms other ways of making BCHs.

A fair comparison with another BCH synthesis would need to consider the specific substrates, how the reactive allene is generated, whether an external stimulus or catalyst is needed, reaction conditions, functional-group compatibility, product substitution, and measured yields. The published abstract and its reference list do not provide a normalized head-to-head comparison across methods.
