A 2011 report described a strategy for making six alkaloids, including strychnine, from one common synthetic precursor. The approach—called collective total synthesis—used a multifunctional tetracyclic spiroindoline intermediate and different catalysts to steer cascade reactions toward different products. “Parent molecule” here means that shared chemical starting point, not a universal route or a molecule from which the products are biosynthesised.
What does “one parent molecule” mean in this synthesis?
In this context, it means a common synthetic intermediate: a molecule prepared during the synthesis that can be converted into several distinct target compounds. It is not a claim that all natural products descend from one biological parent, or that the same route can produce any natural product.

The 2011 work reported by Chemistry World used a tetracyclic spiroindoline precursor bearing multiple reactive features. The researchers designed those “redundant functionalities” so that different catalysts could engage different sites and set off cascades leading to different alkaloids.
How did the researchers branch from the shared intermediate?
Instead of designing and completing six entirely separate routes, the team first made the common scaffold, then used catalyst-directed reactions to guide it along different paths. A cascade links successive transformations, so one catalyst-guided sequence can build on changes made in earlier steps.

The central idea was to design the intermediate for flexibility rather than for just one final structure. David MacMillan, who led the team, described the aim as constructing an intermediate with “many redundant functionalities.” In practical terms, those features provided alternative chemical handles for directing the precursor toward different products.
What did the 2011 report say the strategy achieved?
Chemistry World reported six alkaloid products, including strychnine, and attributed the step-count comparison to the researchers: 36 steps in total, or an average of six per product, compared with about 12 steps per molecule in the best literature systems at the time. This is a historical comparison reported in 2011, not a current, across-the-field benchmark. Step counts can depend on how routes and shared operations are counted, so the figures should not be read as a universal measure of synthetic efficiency.

The report quoted natural-products chemist Alan Armstrong of Imperial College London calling it “very impressive” to access six structurally diverse alkaloids from a common core. He also noted its potential, if applicable more widely, for preparing diverse compound collections for biological screening. That potential is distinct from proof that the method works broadly across natural-product families.
How does this differ from later enzyme-guided scaffold editing?
Later chemoenzymatic methods also start from a shared molecular scaffold, but they solve a different problem. The 2011 strategy builds several target alkaloids from a purpose-designed synthetic intermediate. A later skeletal-editing approach instead modifies existing natural-product-derived scaffolds using engineered cytochrome P450 enzymes together with chemical reactions.

- Where products diverge: In the 2011 report, catalyst choice directs cascades from the common spiroindoline precursor. In the later method, enzyme variants select particular, sometimes remote, aliphatic C–H sites on a scaffold.
- How the scaffold changes: The later study oxidised selected C–H positions to ketones, then used Baeyer–Villiger rearrangement or ketone homologation to expand rings.
- Reported scope: The skeletal-editing study reported 17 edited derivatives across the substrates it examined. That count belongs to that study and should not be combined with the six alkaloids in the 2011 report.
The enzyme-guided chemistry also had substrate-dependent limits. Epoxidation competed with ring expansion for some substrates; 2-oxo-micheliolide did not undergo the desired ring expansion under the tested conditions, and a totarol-derived phenol decomposed under the rearrangement conditions. These results show why scaffold editing is not a universal substitute for route design.

The skeletal-editing study reported in vitro cell-line assays, not clinical evidence. Most edited compounds showed no detectable anticancer activity in the panel. Two analogues had selective activity against H1155 cells in the reported assays: parthenolide derivative 20 had an ED50 of 21 ± 3 μM, and artemisinin derivative 30 had an ED50 of 25 ± 4 μM. These are specific laboratory observations, not evidence of efficacy or safety in people.

How is the separate CeDOS approach related?
A distinct chemoenzymatic diversity-oriented synthesis study, abbreviated CeDOS, began with parthenolide and reported producing a collection of about 50 complex natural-product-like molecules through P450-catalysed oxyfunctionalisation followed by divergent chemistry. It is another way to generate molecular diversity from a common scaffold, but it is separate from both the 2011 collective total synthesis and the later skeletal-editing study.
