Scientists can probe an individual atom’s electronic and bonding structure by combining a focused scanning transmission electron microscope (STEM) beam with electron energy-loss spectroscopy (EELS). In a 2010 demonstration, Kazu Suenaga and Masanori Koshino measured carbon atoms at a graphene edge and distinguished atoms with one, two, or three neighboring carbon bonds from their spectral signatures. It was a site-specific chemical measurement—not simply a sharper microscope image.

What the 2010 experiment showed

Suenaga and Koshino, researchers at Japan’s National Institute of Advanced Industrial Science and Technology (AIST), reported their results in the Nature paper “Atom-by-atom spectroscopy at graphene edge,” published online December 15, 2010. The experiment targeted carbon atoms along a graphene boundary, where atoms can have different numbers of neighboring atoms than they do inside the sheet.

The researchers combined scanning transmission electron microscopy (STEM) with electron energy-loss spectroscopy (EELS). STEM focuses an electron beam onto a small region; in this experiment, the probe was positioned at individual atoms. EELS measures how much energy the electrons lose after interacting with the specimen. The energy-loss near-edge fine structure—the detailed shape of a spectrum near an energy-loss edge—provided information about the electronic and bonding structure of the targeted carbon atom.

From those spectral features, the team distinguished carbon atoms with single, double, and triple coordination. This is more informative than identifying which element is present: the signal revealed differences in the local bonding environment of carbon atoms at specific sites.

How the atom-by-atom measurement worked

Focus the probe on a chosen site

A STEM instrument scans a focused electron probe across a specimen. For site-specific spectroscopy, the probe is placed on the atom of interest and the energy distribution of electrons that emerge is analyzed. The measurement therefore connects a spectrum to a particular atomic location rather than averaging over a large area.

Interpret the energy-loss fine structure

When electrons pass through a sample, some lose energy through interactions with its atoms. EELS records those losses. Fine structure near an energy-loss edge can carry information about an atom’s electronic state and chemical bonding. In this study, comparing that structure at graphene-edge sites made it possible to distinguish different carbon coordination environments.

The result is not a general-purpose readout of every property of every atom. It demonstrates that electron microscopy paired with EELS can provide local chemical-state information at atomic scale under suitable experimental conditions.

Reported experimental settings—and their limits

A contemporary Chemistry World report described a 0.1 nm-diameter probe and an accelerating voltage of 60 kV for this experiment. The low-voltage setup was intended to provide a suitably short electron wavelength for high-resolution spectroscopy while reducing the risk of sample damage. These are reported parameters for this particular measurement, not universal settings and not a guarantee that an electron beam will leave a sample undamaged.

Why the measurement was difficult

  • Weak signal: Fine-structure spectra from individual light atoms are difficult to obtain because the signal is extremely weak.
  • Beam damage: The electron beam can damage the specimen, limiting how much data can be gathered and under what conditions.
  • Specific interpretation: The demonstrated conclusion concerns carbon atoms at a graphene boundary. It does not establish equivalent performance for every element, material, or specimen.

Those constraints explain why the work mattered: it showed that useful bonding information could be obtained from an individual light atom in its local context despite the challenge of weak signals and specimen damage.

What the technique might enable

Suenaga and Koshino proposed that site-specific spectroscopy could help researchers investigate local electronic structures in nanodevices and individual molecules. Those were prospective applications, not results established by the graphene-edge demonstration. The paper’s demonstrated system was carbon atoms at a graphene boundary.

It enables detailed examination of biological samples, plant structures, circuit boards, scalp, collectibles (coins and stamps), and materials-enhancing research, diagnostics, repairs, and quality control

For researchers, the distinction is between elemental identification and chemical-state analysis. Knowing that a region contains carbon is not the same as determining how a particular carbon atom is bonded. The reported near-edge fine structure supplied that more local bonding information.
