Scientists identify an unknown grain or metallic inclusion in glass by combining methods, not by relying on one instrument. Microscopy can locate and image a feature; energy-dispersive X-ray spectrometry can show which elements are present in the analyzed area; and techniques such as X-ray diffraction or Raman spectroscopy can help identify a mineral phase. The right sequence depends on what is being asked, how much sample is available, and the sensitivity and spatial detail needed.
What does “identify” mean in this context?
A tiny feature in glass can raise several different questions. Is it inside the glass or merely on its surface? What does it look like? Which elements are present? Is it a particular crystalline mineral, a mixture, or a metal-bearing material? Does it resemble material in another sample? Those are related questions, but they are not interchangeable—and no single measurement necessarily answers all of them.

Three levels are especially useful to separate:
- Observation: locating the feature and describing its color, shape, texture, and position.
- Composition: measuring or detecting elements, such as silicon, iron, or copper, in a selected area or sample.
- Phase identification: determining the material’s mineral or crystal structure, where it is crystalline and the measurement can resolve it.
A fourth question—whether two materials share a source—requires comparison and interpretation beyond simply finding the same elements or a similar-looking grain. The conclusion should match the evidence: a measurement may characterize an inclusion without uniquely naming its origin.

1. Inspect the glass and locate the feature
Examination begins by documenting the specimen and the suspected material. Optical or petrographic examination can record color, shape, texture, and whether a particle appears embedded or lies on the surface. That distinction matters: surface contamination can be mistaken for an inclusion if location is not established. Sample preparation depends on the specimen’s size and condition and on the question the laboratory intends to answer, so there is no universal preparation recipe.

Forensic glass examination also considers physical and optical properties when comparing fragments, as described by the National Institute of Standards and Technology (NIST). That forensic task is related to, but different from, naming a single geological grain inside glass.
2. Image the feature and examine local chemistry
A scanning electron microscope (SEM) can image a small region at high magnification. In many workflows, the image is paired with energy-dispersive X-ray spectrometry (EDS): the instrument records characteristic X-rays produced by elements in the analyzed volume. The result can help show whether a feature contains elements consistent with a metal-bearing particle, a silicate, or another composition.
Backscattered-electron (BSE) imaging can reveal textural and compositional contrasts in a sample. A 2024 U.S. Geological Survey (USGS) study of Meteor Crater impact melts used BSE imaging and EDS to characterize glass, mineral phases, and metallic inclusions. It is an example of a combined approach in one geological setting, not a protocol that automatically applies to every glass specimen.

EDS is local and useful for characterizing inclusions, but its elemental result does not itself establish a unique mineral phase. Nor is SEM-EDS sufficiently sensitive for every trace-element comparison. NIST’s Materials Trace Interpretation Document, surfaced as draft guidance in 2025, cautions about that sensitivity limit and points to more sensitive elemental techniques when enough sample is available.
3. Test for a mineral phase if the question requires it
Elemental chemistry and mineral identity are different kinds of evidence. A list of detected elements does not reveal, by itself, how atoms are arranged in a crystal. X-ray diffraction (XRD) is used to identify crystalline phases. Raman spectroscopy provides molecular and vibrational information that can help distinguish materials with similar elemental composition.
The distinction can be important even among forms of the same substance. In its 2024 impact-melt report, the USGS notes that without Raman spectroscopy or another suitable method, it can be difficult to determine which silica polymorph is present. The USGS’s 2011 overview of mineralogical analyses describes the complementary use of XRD, petrography, SEM, and microprobe methods.

4. Measure composition at the needed sensitivity
If the question concerns the glass’s elemental composition or a comparison between glass samples, laboratories may use X-ray fluorescence (XRF), including micro-XRF, or inductively coupled plasma mass spectrometry (ICP-MS), including laser-ablation ICP-MS (LA-ICP-MS). NIST’s Glass Evidence Analysis program describes XRF and LA-ICP-MS as techniques commonly used by forensic glass analysts. These methods address elemental composition; XRF does not directly name a mineral phase.
Method choice depends on the target, spatial scale, required sensitivity, sample amount, and condition. The exact preparation and whether a measurement alters or consumes material depend on the instrument and laboratory protocol. Ask the laboratory about those specifics before submitting a valuable or very small specimen.
Which method answers which question?
| Method | What it can contribute | Important limit |
|---|---|---|
| Optical or petrographic examination | Documents visible appearance, texture, and whether a feature appears internal or on the surface. | Appearance alone does not establish elemental composition or crystal structure. |
| SEM imaging, including BSE | Shows fine-scale morphology and textural or compositional contrast; can guide analysis of a selected feature. | An image alone does not identify all elements or uniquely determine a mineral. |
| SEM-EDS | Detects elements in a local analyzed volume and can characterize inclusions alongside SEM imaging. | Element detection is not phase identification; sensitivity may be inadequate for some trace-element comparisons. |
| XRD | Identifies crystalline phases through diffraction evidence. | It addresses crystalline-phase identification, not every question about a trace inclusion or its source. |
| Raman spectroscopy | Adds molecular or vibrational evidence that can help distinguish phases, including silica polymorphs. | Its role is complementary; the available evidence does not establish that it answers every inclusion question by itself. |
| XRF or micro-XRF | Measures elemental composition; micro-XRF can be used for spatially targeted analysis. | Elemental composition does not directly identify a mineral’s crystal structure. |
| ICP-MS or LA-ICP-MS | Supports elemental characterization and, where needed, more sensitive trace-element comparisons. | Suitability depends on the sample and laboratory method; it does not by itself prove a unique source. |
The table describes the methods’ roles, not a guaranteed sequence or universal performance ranking. Spatial scale, sensitivity, preparation, and material alteration depend on the instrument configuration and laboratory procedure. The 1996 USGS Analytical Methods Manual for the Mineral Resource Surveys Program is useful background on validated methods and quality assurance, but it is not a current, instrument-specific protocol for an unknown glass inclusion.

How should you choose an analysis?
Start with the question rather than the instrument. A laboratory needs to know whether the goal is to locate and describe a grain, distinguish a crystalline phase, measure trace elements, compare glass samples, or investigate several of these at once. A staged workflow can prevent an expensive or intrusive analysis from being chosen before the feature is understood.
- Describe the specimen and the uncertainty. Tell the laboratory what is known about the glass, where the feature appears to be, and whether the immediate concern is identity, composition, or comparison.
- Ask what can be established non-destructively or with minimal sampling. The answer depends on the specimen and lab protocol; do not assume that a method leaves the sample unchanged.
- Locate and document the target. Optical or petrographic examination and microscopy can help establish whether the material is internal and guide a local analysis.
- Use local elemental analysis when composition is the first question. SEM-EDS can characterize a small feature, but discuss its detection limits if trace quantities matter.
- Add phase-sensitive analysis if a mineral name is needed. Ask whether XRD, Raman, or another suitable method can resolve the particular material and whether the amount and condition of sample are adequate.
- Choose a more sensitive elemental method for trace comparisons when justified. XRF or ICP-MS-family techniques may be relevant; the laboratory can advise on sample requirements and whether the technique suits an inclusion, the surrounding glass, or both.
- Request an interpretation tied to the evidence. A useful report distinguishes observations, measured composition, phase assignment, uncertainty, and any source-comparison conclusion rather than treating them as one claim.
This is a decision framework, not a fixed protocol. The appropriate order can change with sample size, preservation needs, and whether the target is a single inclusion or the bulk glass.

What can the results establish—and what can’t they?
Elements are not a unique mineral name
EDS or XRF may show that certain elements are present, but elemental overlap is common among different minerals and mixtures. To claim a specific phase, the laboratory needs evidence suited to phase identification, such as diffraction or vibrational information, and the result must be interpretable for that sample. A list of elements should not be promoted into a definitive crystal-structure identification.

A similar composition is not proof of a common source
Two glass samples can share measured features without the analysis proving that they came from the same object, location, or manufacturing event. Source comparison requires an appropriate framework for interpreting the degree of similarity and the variability of the materials being compared. NIST has noted that standardized interpretation of glass evidence remains an issue.
A 2021 NIST interlaboratory study assessed refractive-index, micro-XRF, and laser-induced breakdown spectroscopy (LIBS) data for forensic comparison of glass samples. Seventeen laboratories participated. That number describes study participation, not an accuracy rate for mineral identification or a probability that a particular match is correct. The study recommends verbal scales and/or background databases, where available, to express the significance of comparison results.
One successful geological example is not a universal recipe
The USGS Meteor Crater work demonstrates how imaging and composition can be combined to study impact-melt glass and inclusions. It does not establish that every glass type, inclusion size, or investigative purpose should receive the same analysis. Forensic glass comparison, geological characterization, and examination of an isolated unknown grain have overlapping tools but distinct goals.
What should you ask a laboratory before submitting glass?
- Can you determine whether the suspected material is embedded or on the surface?
- Which result are you being asked to provide: visual characterization, elemental composition, mineral phase, or a comparison?
- What is the minimum sample size or condition required, and can the feature be targeted at its scale?
- Will preparation or analysis consume, remove, coat, or otherwise alter any part of the specimen?
- What are the detection limits relevant to the elements of interest, and is SEM-EDS sensitive enough for this question?
- Can the report separate measured observations from interpretation and state the limits of any source-comparison conclusion?
These questions help align the analytical plan with both the scientific question and the specimen’s preservation needs. Because preparation and material alteration are protocol-specific, the laboratory—not a generic instrument description—is the appropriate source for those details.
