Molecular motors move cargo by binding to cytoskeletal filaments and using chemical energy, usually from ATP, to generate steps along those tracks. Kinesins and cytoplasmic dynein generally carry cargo on microtubules; myosins move cargo on actin filaments. The motor recruited, the way it is attached and activated, and the arrangement of the tracks together help determine where a vesicle or organelle travels.
What molecular motors do
A cell is not an open, still container in which large structures simply drift to their destinations. Vesicles and organelles need directed transport through the cell interior. Molecular motors provide one way to move them: a motor interacts with a filament, couples chemical energy to changes in its shape, and advances along the filament while connected to cargo.

The two main filament systems involved are microtubules and actin filaments. Kinesins and cytoplasmic dynein generally use microtubules; myosins use actin. A motor therefore does not move freely through the cell in a chosen direction. Its path depends first on which track it can use, and then on how the track is oriented, which motor is active, and how that motor is connected to the cargo.
This distinction matters because “toward the cell edge” and “toward the cell center” are not the motors’ built-in definitions of direction. The motor responds to the polarity of the filament it is on. Since filaments can be arranged differently in different places, the same motor’s direction relative to a track does not by itself identify a destination in the whole cell.

How motors turn chemical energy into movement
A motor has a part that interacts with a filament and, often, other regions that help connect it to cargo. In kinesin, ATP turnover is coupled to conformational changes in the motor and neck domains. Those changes alter how the motor interacts with the microtubule and drive forward stepping. The motor’s tail is associated with cargo, so movement of the track-binding part can carry the attached load along.
This is a useful model for understanding transport, but not a universal blueprint for every motor. Motor families differ in structure and stepping cycle. It is more accurate to say that motors couple chemical energy to track-relative movement than to assume every motor uses the same sequence of shape changes as kinesin.
The cargo does not need to make each step itself. The motor supplies movement at the filament interface; a connection between motor and cargo transmits that movement. If that connection is absent, inappropriate, or not activated, the motor’s ability to move along a track is not enough to ensure that a particular cargo will be transported.

Microtubules and actin filaments are polarized: their two ends are structurally distinct. Motors have characteristic directionality on those tracks. In the common case, kinesins move toward microtubule plus ends and cytoplasmic dynein moves toward microtubule minus ends. Most myosins move toward the barbed, or plus, end of actin; myosin VI is a well-known exception that moves toward the pointed, or minus, end.

| Motor family | Track | Usual direction relative to the track |
|---|---|---|
| Kinesin | Microtubule | Generally toward the plus end |
| Cytoplasmic dynein | Microtubule | Generally toward the minus end |
| Myosin | Actin filament | Most classes move toward the barbed/plus end; myosin VI is a notable pointed/minus-end-directed exception |
These are track-relative directions, not guaranteed routes through the cell. For example, a plus-end-directed kinesin follows a microtubule toward that filament’s plus end. Whether that route leads outward, inward, or elsewhere depends on the local arrangement of microtubules. The same caution applies to interpreting minus-end-directed dynein or the direction of a myosin on an actin filament.
Consequently, a cell’s transport pattern is produced by both motor properties and track geometry. Knowing a cargo has a particular motor gives a directional tendency on its filament; to predict a destination, one also needs to know how the relevant filaments are oriented where the cargo is moving.
How a motor is recruited to the right cargo
Motor attachment is often more selective and more elaborate than a single motor-tail-to-membrane bond. A cargo may recruit a motor through a collection of cargo proteins, adaptor proteins, Rab GTPases, scaffolds, or membrane lipids. These components can help bridge the cargo and motor, and they can contribute to choosing which motor is recruited to which cargo.

Recruitment can also affect whether a motor is ready to move. Some motors are autoinhibited or otherwise less active when they are not associated with cargo. Cargo binding and partner proteins can promote a more active state. Signaling, including phosphorylation or calcium-dependent pathways, can alter motor-cargo association or movement. These are regulatory strategies found in some systems, not rules that should be presumed for every motor or cargo.
This makes transport a regulated process, not merely a matter of motors bumping into filaments. A cargo’s molecular identity and regulatory state can influence which motor is engaged and whether it is active. The resulting movement depends on the combination: an appropriate track, a functional motor-track interaction, and a cargo connection that allows the motor to act on the load.
Why cargo can move in both directions
Some vesicles and organelles carry motors that move in opposite directions on microtubules. A cargo with plus-end- and minus-end-directed motors can therefore make movements in both directions rather than following a simple one-way route. This does not mean the motors necessarily pull with equal strength or are active at the same time.

1Fix the driver behind crashes, sound loss and screen glitches2Clear out junk files and repair common Windows errors3Scan for outdated or missing drivers - takes under a minuteTwo broad explanations are considered. In one, simultaneously active motors compete mechanically, and the balance of their activity influences which way the cargo moves. In another, regulation coordinates the motors, turning down one direction’s motor activity while the other moves the cargo. A particular cargo may involve aspects of both. The mechanism and relative contributions depend on the transport system; there is no single settled explanation for every bidirectionally moving cargo.

Seen this way, a reversal is not necessarily evidence that a cargo has lost its route or that the cell’s transport has failed. It can arise from the regulatory and mechanical behavior of motors attached to the same cargo. To explain a specific case, one would need evidence about the motors present and how their activities are controlled, not just the observed direction at one moment.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone. How transport uses both microtubules and actin
Microtubule-based transport often supports movement over longer distances, while actin and myosins often contribute to local movement and delivery. This is a broad pattern, not a rigid division in which one filament system handles all long journeys and the other only the final step. The relative role of each track depends on the cell and the cargo’s route.
A cargo can also transition from one filament system to another. A general model is that a cargo travels along microtubules and then uses myosin-based movement on actin for a local leg of transport. In inward transport, a route may involve the reverse order. The important point is that a cargo need not remain on one type of track for its whole journey.

Outbyte PC Repair FREERepair Windows errors before they cause bigger problemsFix Now →Outbyte Driver Updater FREEScan for outdated or missing drivers - takes under a minuteDriver Scan →Such a route raises a practical mechanistic question: how does the cargo stop using one motor-track system and start using another? Track intersections and transitions have been observed in several systems, but exactly how motors and cargo are handed off at those junctions remains incompletely understood. It is therefore reasonable to describe microtubules as common longer-range routes and actin as a frequent contributor to local delivery, but not to present a single universal handoff mechanism.

How to reason through a cargo’s route
To understand a transport example without confusing motor direction with cell geography, work through the components in sequence. The route is not determined by a motor name alone.
- Identify the track. Establish whether the movement in question is on a microtubule or an actin filament. Kinesin and dynein generally use microtubules; myosins use actin.
- Determine track polarity. Identify the relevant plus and minus ends, or the barbed and pointed ends for actin. Do not assume an end always points toward a familiar cellular landmark.
- Identify the motor and its directionality. Apply the general family pattern, while accounting for exceptions such as myosin VI.
- Work out the cargo connection. Ask whether attachment is direct or mediated by adaptors, Rab proteins, scaffolds, lipids, or a combination. Avoid assuming one connector is used by all cargoes.
- Consider regulation. Cargo binding, partner proteins, phosphorylation, calcium-dependent signaling, or other system-specific controls may change motor recruitment or activity.
- Check for competing motors or a track transition. Opposing motors can contribute to bidirectional movement, and the cargo may switch between microtubules and actin during its journey.
This sequence separates questions that are often blurred together. “Which way does the motor step?” is about its direction on a filament. “Where does the cargo go?” additionally depends on track arrangement, motor recruitment, motor regulation, opposing activity, and possible transitions between track types.
What is established—and what remains system-specific
The broad framework is clear: molecular motors bind cytoskeletal tracks, use chemical energy to generate movement, and connect that movement to cargo. Kinesins and dynein generally travel on microtubules in opposite directions; myosins move on actin, with direction varying by class. Cargo recruitment and activation are regulated, and some cargoes carry motors capable of opposing directions.
The details should not be generalized beyond the evidence for a particular system. The exact cargo adaptors, regulatory signals, balance of opposing motors, and mechanics of handoff between actin and microtubules vary. Even where a general transport pattern is useful, it does not establish a universal molecular mechanism or an identical route in every cell. The best explanation of a particular cargo therefore combines its motors and cargo-binding machinery with the organization of the tracks it encounters.
