Some methods can track particles moving through a fluid; others deliberately concentrate their measurement near a sensor surface. To understand what “beyond a sensor surface” means, distinguish the particle’s position from the optical signal being measured: a particle can be detected in a volume, or its presence can be inferred from a change in light at an interface.
What “label-free” means—and what it does not
In fluorescence microscopy, a fluorescent label attached to a target supplies a bright signal. In label-free optical detection, the signal instead comes from the target’s own interaction with light: it may scatter light, change its phase, absorb it, or alter the local refractive index. The particle does not have to be made fluorescent for the instrument to detect it.

That distinction does not mean the sample is untouched or the sensor is always plain. Researchers may prepare or dilute a sample, immobilize a particle, functionalize a surface so a target binds to it, or use a specialized substrate or optical resonator. “Label-free” describes the source of the measured signal, not the whole preparation or measurement process. The 2025 review of label-free single-molecule optical detection describes how weak intrinsic signals remain a central challenge and surveys methods used to enhance them (npj Biosensing, 2025).

A technique may detect a particle while it is suspended and moving in a fluid, or it may sense changes caused by particles close to an interface. These arrangements are not interchangeable: they answer different questions about where a target is, how it moves, and whether it interacts with a surface.

How a faint particle signal becomes detectable
A small particle generally scatters only a small amount of light. Simply illuminating it and measuring its scattered intensity can leave a weak signal against background light and other sources of noise. Label-free methods address this problem by choosing a more informative optical measurement or by enhancing the field that interacts with the particle. There is no single enhancement strategy that suits every sample.
Interferometric scattering compares the particle with a reference field
Interferometric scattering microscopy, commonly abbreviated iSCAT, combines light scattered by a particle with a reference field, often reflected light from an interface. When the two fields overlap, they interfere; the resulting intensity can change in a way that reveals the particle even when its scattered light would be difficult to detect on its own. The instrument is not turning the particle into a bright emitter. It is measuring how the particle’s faint scattered field changes the combined optical signal.
The recorded pattern can be analyzed to locate a particle and, depending on the setup and experiment, to track it or quantify properties of nanoscale objects. A 2019 review describes iSCAT as an all-optical method for label-free imaging, tracking, and quantitative characterization, with applications including particle tracking, biomolecular assembly, and mass measurement (Annual Review of Physical Chemistry, 2019). Those capabilities do not establish one universal detection limit: the observed signal depends on optical configuration and sample conditions.

Surface plasmons and evanescent fields emphasize an interface
An evanescent field is concentrated near an interface rather than extending uniformly through the sample. Surface-plasmon sensing uses an optical field associated with surface plasmons to detect local changes, such as a change in refractive index when material approaches or binds near the surface. A 2023 review reports that the surface-plasmon field penetrates approximately 30–200 nm into the adjacent liquid (Chemical Society Reviews, 2023).
That range describes the field’s reach into the liquid, not a particle-size limit or a general sensitivity rating. This geometry can be useful for studying surface binding and near-surface changes. It should not be described as tracking freely suspended particles throughout the bulk fluid: a particle well beyond the field’s near-interface region will not contribute in the same way as one close to the sensor.
Holography reconstructs information about the optical field
Holographic methods record information that can be reconstructed as a complex optical field, providing more than a single brightness value. The reconstructed field can help estimate a particle’s size and refractive index and, in some cases, distinguish material classes from their optical response. A 2025 review of label-free microscopy for dielectric nanoparticles discusses holographic and interferometric characterization (Nanoscale, 2025).

More kinds of information in a measurement do not automatically mean a lower detection limit. The Nanoscale review identifies detection limit as a historical drawback of holographic microscopy relative to some other approaches. Holography can be attractive when characterization is the goal, but the richer field description alone is not proof that it will detect the weakest particle signal in a particular experiment.

In a volume-based measurement, particles can be observed or tracked while suspended in a liquid rather than first landing on, binding to, or being tethered to a sensor. Their Brownian motion—the random movement caused by collisions with molecules in the surrounding fluid—or other motion can be followed optically. A 2024 review distinguishes scattering approaches that track particles without requiring them to land on a surface from approaches that use tethering or surface interactions (Quarterly Reviews of Biophysics, 2024).
“Away from a surface” does not mean that every part of the instrument is surface-free. An objective, coverslip, chamber, or reflective interface may still be part of the optical path or provide the reference field. The relevant point is whether the target itself must be at the sensing interface to generate the measurement, or whether it can be detected as it moves in the fluid volume.

Particle tracking is also a different outcome from detecting that something is present. A detection measurement may establish that a signal consistent with a particle occurred. Tracking requires following its position over time. Characterization goes further by estimating properties such as size, refractive index, or mass. Each additional aim places different demands on signal quality, spatial and temporal resolution, and analysis.
Instrumentation is a configured system, not just a microscope. A NIST review describes optical tracking systems as combinations of light sources, sensors, optics, and stages, and discusses how choices in illumination and detection affect their capabilities (NIST, 2016). The appropriate configuration depends on the particle, sample, and measurement question.

How to compare the main approaches
The methods are easiest to compare by asking what generates the signal and where the target must be. The table describes broad method families, not guaranteed performance for every instrument or sample.
Best Value
| Approach | Measured signal | Surface dependence | Useful when the goal is | Important qualification |
|---|---|---|---|---|
| Interferometric scattering microscopy (iSCAT) | Interference between particle-scattered light and a reference field | Often uses reflected light from an interface as a reference, but the particle can be imaged or tracked in a fluid | Label-free imaging, tracking, or quantitative study of nanoscale objects | Signal and sensitivity depend on optical configuration and sample conditions (Annual Review of Physical Chemistry, 2019) |
| Surface-plasmon or evanescent-field sensing | A local refractive-index change or related change in the near-surface optical field | Strongly emphasizes the interface; the reported plasmon-field penetration into adjacent liquid is about 30–200 nm | Measuring changes close to a surface, including surface interactions | The penetration range is not a universal detection limit or a measure of bulk-volume tracking (Chemical Society Reviews, 2023) |
| Holographic microscopy | A recorded and reconstructed complex optical field | Depends on the specific optical arrangement; holography is not inherently a surface-binding measurement | Estimating size or refractive index and characterizing optical response | Richer field information does not guarantee the best detection limit (Nanoscale, 2025) |
| Surface-tethered or binding-based scattering methods | Scattering from targets interacting with or held near a surface | Requires surface interaction or tethering for the measured arrangement | Observing individual targets in a surface-based assay | It is not equivalent to following freely suspended particles in a bulk fluid (Quarterly Reviews of Biophysics, 2024) |
Other enhancement strategies discussed in reviews include resonators and nanoscale apertures. They can modify or concentrate the optical interaction, but their usefulness depends on the design and measurement. A 2022 overview of optical microscopy systems for unlabeled nanoparticles surveys a range of techniques rather than establishing a universally superior one (Optical Microscopy Systems for the Detection of Unlabeled Nanoparticles, 2022).
A practical way to choose a measurement geometry
Start with the scientific question rather than the label-free method name. A surface sensor is relevant if the question is whether a target binds or changes the local environment at an interface. A volume-tracking method is more relevant if the question is how particles move in solution without first being tethered. If the objective is to estimate size, refractive index, or another property, check whether the method’s optical signal and analysis actually support that inference.
- Decide where the target needs to be. Specify whether it may move freely in the fluid, must approach a surface, or must be immobilized. This rules out geometries that cannot answer the question.
- Define the required result. Separate detecting presence from tracking a trajectory or estimating material properties. Do not assume that a method optimized for one outcome automatically delivers the others.
- Check the sample’s optical context. Particle concentration, optical contrast, and background affect whether a weak intrinsic signal can be distinguished. Reviews discuss limitations across techniques, but the available evidence does not support a universal ranking across all samples.
- Match signal type to the question. Consider whether the experiment needs scattered intensity, an interference signal, a near-surface resonance or refractive-index change, or a reconstructed optical field. These signals encode different information.
- Evaluate the whole instrument and analysis. Account for illumination, detection, optics, stages, surface preparation where applicable, and the analysis needed to interpret the signal. A method name alone does not specify a complete or reproducible setup.
- Validate under the intended conditions. Performance should be assessed for the actual particle and sample, with the intended geometry and measurement objective. A sensitivity figure from another configuration cannot be assumed to transfer.
What label-free detection avoids—and what remains difficult
Without fluorescent tags, measurements avoid photobleaching of those labels: fluorescent signal can fade under illumination. A 2025 review notes that label-free methods do not suffer from label photobleaching and can enable prolonged observations and real-time tracking with minimal perturbation (npj Biosensing, 2025). This is not a guarantee that every label-free experiment leaves a sample entirely unperturbed; illumination, surface interactions, and sample handling can still matter.

The central trade-off is that removing a bright label also removes a strong source of optical signal. Weak scattering or small refractive-index changes can be difficult to separate from background, and improvements in one property may come with practical costs in another. The 2025 review identifies sensitivity, robustness, and versatility as continuing challenges, while the 2023 Chemical Society Reviews article characterizes label-free optical biosensing as predominantly a research tool at the time of publication. That biosensing statement should not be generalized to all optical particle tracking or treated as a claim that no systems are used outside research.
Accordingly, neither “label-free” nor “nanoscale” identifies a single consumer-ready instrument. These are specialized optical measurements, and a general-purpose microscope cannot be assumed sufficient for nanoscale label-free detection. The sources describe research instrumentation and method families, not one product that reliably reproduces them.
