How Germanium Nanospheres Revealed Kinesin’s Tiny Steps

Germanium nanospheres helped researchers resolve kinesin-1’s 4-nanometer center-of-mass steps and observe microsecond-long slipping under hindering force.

In a 2021 laboratory study, researchers used germanium nanospheres as optical-trapping probes to measure kinesin-1 motion at finer resolution. They reported 4-nanometer center-of-mass steps and found that, under hindering force, the motor could slip along a microtubule in rapid 8-nanometer events before detaching or resuming directed movement.

What the study found

Kinesin-1 is a motor protein that transports cellular cargo along microtubules. Swathi Sudhakar and colleagues measured single kinesin motors in vitro and reported that they took 4-nanometer steps in their center of mass. The result addressed whether such fine substeps could be resolved in kinesin’s movement.

The measurements also captured a different behavior under hindering load: instead of detaching immediately, kinesin could slip along the microtubule in 8-nanometer steps lasting microseconds. Motors remained in this slip state before either detaching or reengaging in directed movement. The authors reported that reengagement and rescue of directed motion occurred more often than they expected.

How germanium probes enabled finer measurements

The approach used germanium nanospheres as probes in optical tweezers, which can track movement while applying or measuring force. Chemistry World’s 2021 account reports that the team made regularly sized spheres 70 nanometers in diameter by reducing germanium oxide, coated them with a lipid bilayer to mimic vesicles, and bound them to kinesin.

Probe size presents a measurement tradeoff: a smaller probe can reduce drag and improve resolution, but it scatters less light and is harder to trap with sufficient force. The team used germanium’s high refractive index to capture more light momentum while using weaker lasers, according to the contemporary Chemistry World report. The study abstract independently describes the nanospheres’ role as ultraresolution optical-trapping probes. A 2022 protocol chapter by Sudhakar, Pasupathi Rajendran, and Erik Schäffer later described synthesizing 70-nanometer germanium probes and using them to demonstrate the 4-nanometer kinesin-1 center-of-mass steps.

Why the slip state matters—and what it does not show

The results add a slipping state to the picture of kinesin’s motion under hindering force: the motor may continue to engage with the microtubule in short backward or slipping events rather than simply detach. Because some motors reengaged and returned to directed motion, the authors argue that slipping and rescue should be considered when understanding long-range transport by teams of motors.
