In a 2025 study, researchers at the University of Naples Federico II used filter paper with a 1:1 ethanol–water solvent mixture to distribute reagents while solvent evaporated during the reaction. They reported yields as high as 99.5%, but that figure is the study’s maximum—not a result for every compound.
How the filter-paper method works
The researchers applied the reactants in a 1:1 mixture of ethanol and water to filter paper. The paper acted as a support that spread the reagents across the reaction area; as the reaction proceeded, the solvent evaporated. A 3D-printed housing held the paper in place. The team selected the filter-disc size for the scale: their reported setup used a 2 cm disc at 0.14 mmol and a 6 cm disc at 1.4 mmol.

The rationale is to combine reagent distribution on a solid support with solvent evaporation, rather than carrying out the reaction in a conventional batch mixture and then removing water during work-up. In the study’s comparison, the authors reported fewer purification steps for the paper-based approach. These are results and explanations for the tested procedure, not evidence that paper will simplify every synthesis. The full study in Green Chemistry describes the experimental method and comparison.
What the study demonstrated
Alfano, Kalligosfyri, Baia, Brindisi and Cinti reported preparing 36 squaramide-based compounds, including 30 newly characterized chemical entities. The scope included symmetric and asymmetric squaramides made with substituted benzylamines, anilines and selected aliphatic amines. Substituents included methoxy, trifluoromethyl, nitro and halogen groups. Results varied across reactions; the reported maximum yield should not be read as a typical or universal outcome.

Separately, Chemistry World reported 97% yield after five minutes in small-scale benzylamine tests at room temperature. That is the news report’s summary of specific tests; it should not be substituted for the primary paper’s broader experimental context. How it compares with batch synthesis
The paper compares its platform with batch reactions under the study’s tested conditions. Its reported distinction is not simply that paper makes the reaction faster: the setup changes how the reagents are supported and how solvent is handled. In the authors’ account, evaporation during the paper-based reaction avoids the water-removal work-up used in the batch comparison and reduces purification steps. The reported times and work-up differences apply to those specific experiments, not to batch chemistry as a whole.

For a chemist considering the approach, the practical comparison is whether the solvent, substrate and scale suit the paper platform, and whether its simpler reported work-up offsets the effort of adapting the setup. This study establishes a laboratory demonstration and a tenfold scale increase; it does not establish industrial-scale feasibility.
Where the method may not fit
The solvent is water-based, which can be incompatible with some compounds. Corresponding author Margherita Brindisi told Chemistry World: “A potential limitation is the use of water-based solvents, which may not suit all compounds. However, this can be further optimised through ongoing investigation.” The study therefore supports a promising option for the tested squaramide reactions, not a universal replacement for conventional synthesis.

Brindisi also cautioned that reaction setups need case-by-case evaluation, describing the approach as a starting point for further applications in chemical synthesis. The published scope and scale-up are meaningful demonstrations, but do not establish compatibility with every substrate or prove that the method transfers directly to other reactions.
Paper and equipment considerations
The material described is laboratory filter paper, used as filter-paper discs in a reaction platform. The study does not establish a particular brand or paper grade as necessary. Anyone reproducing the procedure should follow the paper’s protocol and select materials that meet their laboratory’s chemical-compatibility and safety requirements; the disc dimensions reported for the two scales are experimental details, not universal specifications.
