How Blocking Bacterial Hydrogen Sulfide May Boost Antibiotics

A proposed bacterial hydrogen sulfide defense may be targetable, but the evidence is preclinical and later work challenges the resistance claim for Pseudomonas aeruginosa.

Some bacteria produce hydrogen sulfide (H2S) as part of a stress-defense system. A 2021 study proposed that blocking a key enzyme involved in making it could make certain antibiotics more effective. The findings came from laboratory experiments and mouse models, not human treatment; a 2024 study also challenged the resistance finding in Pseudomonas aeruginosa under the conditions it tested.

What the “self-poisoning” headline means

Hydrogen sulfide is often known as a toxic gas, but bacteria can produce it themselves. The headline’s “self-poisoning” phrase is shorthand for this bacterial H2S production and the proposed role it plays in defense against antibiotic stress. It does not mean bacteria deliberately poison themselves, nor that this pathway works identically across all species.

In 2021, Shatalin and colleagues reported that cystathionine γ-lyase (CSE) was a primary H2S-producing enzyme in the studied strains of Staphylococcus aureus and P. aeruginosa. They tested small molecules that inhibit bacterial CSE, aiming to weaken this defense and help bactericidal antibiotics kill bacteria. The study in Science reported antibiotic-potentiating effects in laboratory experiments and mouse infection models.

What the 2021 experiments reported

The authors reported several outcomes, which should not be collapsed into a single claim about “resistance.” CSE inhibitors potentiated bactericidal antibiotics in vitro and in mouse infection models. The study also reported reduced biofilm formation and fewer persister bacteria surviving antibiotic treatment.

  • IMMUNE SUPPORT: Herbal blend designed to support the body's natural defense system.
  • KEY INGREDIENTS: Includes Echinacea, Goldenseal, Marshmallow & more, traditionally used for wellness.

  • CONVENIENT PACK: 1 bottle, 100 capsules for extended use, ensuring continuous support.
  • QUALITY YOU CAN TRUST: Made with high-quality, natural ingredients, crafted with care.
  • Antibiotic potentiation: the inhibitors were tested alongside antibiotics and reported to improve their bactericidal effect in the studied experimental systems.
  • Biofilms: the researchers reported less biofilm formation. Biofilms are bacterial communities that can affect how bacteria respond to treatment.
  • Persisters: the researchers reported fewer persister cells surviving antibiotic exposure. Persisters can survive treatment without necessarily being genetically resistant; their survival is associated with antibiotic tolerance and persistence.

The paper’s authors concluded, “Our results establish bacterial H2S as a multifunctional defense factor, and CSE as a drug target for versatile antibiotic enhancers.” That is the authors’ interpretation of preclinical evidence—not proof of an effective human treatment or an available therapy.

Resistance, tolerance and persistence are different

Antibiotic resistance generally means bacteria can grow despite an antibiotic at concentrations that would inhibit susceptible bacteria. Tolerance describes surviving antibiotic exposure without the same change in the concentration needed to stop growth. Persisters are a subpopulation that survives exposure and can resume growth later. A 2021 Nature Chemical Biology highlight likewise notes that persisters may survive antibiotic exposure even when they are not resistant: the highlight on the study.

These distinctions matter because an experiment showing fewer survivors after treatment does not automatically show that inherited or measurable resistance has been reversed. The 2021 paper addressed antibiotic potentiation as well as biofilms and persister survival; those outcomes involve different measurements and should be described separately.

Why a 2024 study complicates the claim

A later study reported a different result for P. aeruginosa. In 2024, researchers found that changing H2S production did not affect antibiotic resistance in their minimum inhibitory concentration (MIC) assays. Their analyses of cystic-fibrosis isolates also did not support H2S protecting the bacteria from antibiotics during chronic lung infection. See the 2024 study in Antimicrobial Agents and Chemotherapy.

This challenges a broad claim that H2S production weakens antibiotic resistance in P. aeruginosa. It does not, by itself, disprove every result about H2S, CSE inhibitors, biofilms or persistence. The studies address different outcomes and experimental settings: MIC assays measure growth inhibition, while treatment-survival and persistence experiments examine which bacteria remain alive after exposure. Organism and strain, antibiotic, exposure conditions and model also matter when interpreting differences.

What the findings mean for patients

The proposed strategy remains preclinical in the evidence described here. The reported inhibitor effects were in vitro and in mouse models; they do not establish that a CSE inhibitor is safe, effective or available for people, or that patients should change antibiotic treatment. The 2024 findings further caution against treating the proposed resistance effect as settled across species or conditions.

The most accurate takeaway is narrower than the headline: the 2021 study identified bacterial CSE as a potential target for enhancing antibiotic effects in experimental models, while later work did not find an H2S effect on P. aeruginosa resistance in the tested MIC assays and chronic-infection isolate analyses.
