Some gall-forming aphids can alter how a host plant’s genes are expressed as a gall develops. The clearest aphid-specific example links the aphid gene dgc to changes in witch-hazel gall color: greater dgc expression is associated with reduced activity of plant genes involved in anthocyanin pigment production, helping explain why some galls are green and others red. This is gene-expression manipulation, not demonstrated editing of plant DNA. The full sequence by which aphids induce a gall’s shape, tissues and protective nursery environment remains unresolved.
What does it mean for an aphid to manipulate plant genes?
A gall is plant tissue whose growth and development have been altered in association with an insect that feeds or develops there. Rather than simply consuming an existing leaf or bud, a gall-inducing insect can be associated with the production of a new structure that supplies food and protection. In some systems, the insect’s presence is also linked to changes in which host genes are active as the structure forms.

Gene expression is the process by which information in a gene is used to produce a functional product, often a protein. A change in gene expression can alter a plant cell’s behavior without changing the underlying DNA sequence. In this context, saying that aphids “manipulate plant genes” means that their activity is associated with changes in host gene expression and development. It does not mean that aphids are known to rewrite the plant’s genome.

The witch-hazel system provides an aphid-specific molecular link between an aphid gene, host pigment-related gene expression and gall color. Other studies, including work on grape phylloxera, show broad changes in host gene expression and development, but phylloxera is an aphid relative, not the same aphid system. Neither example supplies a universal recipe for how all galls are made.

How do aphids make galls? The best-studied clue is gall color
Hormaphis cornus and witch hazel
The clearest aphid-specific example in the available evidence concerns the witch-hazel cone gall aphid Hormaphis cornus. In the gall-inducing generation, the aphid gene called dgc, short for “determinant of gall color,” is highly expressed in the salivary glands. The gene belongs to the aphid bicycle family, a group associated with candidate proteins that may be secreted.
Researchers found that greater dgc expression is associated with reduced transcription of host plant genes encoding enzymes in the anthocyanin biosynthesis pathway. Anthocyanins are pigments that can contribute red coloration. When those plant genes are less active, less anthocyanin is produced; when dgc expression is lower, anthocyanin can accumulate and the gall can appear red. This relationship helps explain variation between green and red galls.
The finding is a link among aphid gene expression, host gene expression and a visible gall trait. It is not evidence that the aphid gene directly edits the plant’s DNA, nor does the association alone establish every step of the causal chain. The proposed mechanism is that the aphid produces a DGC protein in its salivary glands and delivers it into plant tissue, where it influences the host response. That delivery and mode of action remain a mechanistic interpretation rather than a complete, established account of gall formation.

Bicycle-family genes are candidates, not a catalogue of proven gall builders
Transcriptomic analysis of aphid salivary glands found more than 400 genes encoding predicted proteins with shared bicycle-family structure. A 2023 review presents these as a potentially large set of secreted effectors that could take part in host manipulation. “Predicted” matters: the count describes genes identified from sequence and expression analysis, not more than 400 proteins individually shown to build galls.
Salivary-gland expression makes a gene a plausible candidate for a role in communicating with or influencing plant tissue, but it does not by itself prove that the protein is secreted into the plant, identify its target, or show what happens when the protein is absent. The evidence for dgc and gall color is more specific than the general candidate status of the bicycle-family set; those claims should not be treated as interchangeable.
What changes in a plant as a gall develops?
Gall development is not just a color change. Plant cells divide, expand and specialize to form tissues with different roles. Reviews of aphid galls discuss altered cell fate and host gene expression as useful ways to think about early development, while emphasizing that the first insect signals and the causal pathways they trigger are difficult to identify. Hormone changes, candidate secreted proteins and plant developmental responses may all contribute, but evidence does not establish one standard pathway shared by all gall-forming aphids.

A complementary example comes from grape phylloxera, Daktulosphaira vitifoliae, an aphid relative that induces leaf galls on grape. In a 2019 study by Schultz and colleagues, gall development involved altered cell division and expansion and the formation of a developing nutritive zone. Under the conditions described in that study, a complete leaf gall formed in four to five days. The researchers also reported that development stopped if the insect was removed before the final stage. These observations support the importance of the insect’s continued presence in that particular system; they do not establish that every aphid gall follows the same timetable or depends on the insect in precisely the same way.
Large-scale expression changes in grape phylloxera galls
Across sampled stages of grape phylloxera gall development, Schultz and colleagues reported differential expression in 11,049 grape genes at least once. Their study used a threshold of more than 1.5-fold change and P<0.01. This is a result for that grape–phylloxera system and the study’s sampled stages and criteria, not a count that can be applied to aphid galls generally.
The affected genes included those associated with auxin synthesis and signaling, vascular cambium activity and reproductive-development programs. Auxin is a plant hormone involved in growth and development; the cambium is a tissue that produces new plant cells. The researchers interpreted persistent cambial activity and changes in reproductive-development genes as clues to how gall tissue might acquire unusual developmental capacity. These are clues about possible processes, not proof that any one pathway alone causes the gall.

There are limits to what a gene-expression survey can establish. A measured change in RNA abundance does not by itself show that a gene’s activity caused a particular tissue feature. Inferring function from related genes in other organisms can also be uncertain, and transcript measurements do not capture every layer of regulation. Those caveats apply especially when moving from a broad pattern of altered expression to a claim about the exact molecular trigger.

A gall can provide a feeding site as well as physical protection. In social gall-forming aphids, colonies can pass through multiple parthenogenetic generations inside the gall. In this context, “nursery” is a useful description: the gall is a sheltered setting where a colony can feed and successive generations can develop.

This description has a defined scope. The social-aphid review concerns aphids with this kind of colony organization; not every aphid species forms a gall, and not every gall houses multiple generations in the same way. A gall’s contents and function depend on the insect and host system. The nursery role is therefore a feature of some gall-forming aphid life histories, not a universal property of all galls.
Best Value
The structure also connects the insect’s development to the plant’s. A gall is made of host tissue, but it can become a specialized environment used by the insect. The plant’s altered growth supplies the tissue, while the insect feeds and develops within it. The precise signals that coordinate those outcomes are still under investigation; the nursery function does not by itself reveal how the plant was induced to make the structure.
How the main gall examples compare
| System | Insect and host tissue | Feature or process studied | What the evidence supports |
|---|---|---|---|
| Witch-hazel cone gall | Hormaphis cornus on witch hazel | dgc expression, host anthocyanin-pathway transcription and green versus red coloration | An aphid-specific association connecting insect gene expression, host gene expression and a gall trait; delivery and action of DGC as a secreted protein are proposed, not fully resolved. |
| Grape leaf gall | Grape phylloxera, Daktulosphaira vitifoliae, on grape | Cell division and expansion, nutritive-zone development, gall timing and host transcript changes | A comparative example of insect-associated development and extensive host expression changes; phylloxera is an aphid relative, so its findings do not directly prove the mechanism in H. cornus. |
| Related aphids on Japanese witch hazel | Four related aphid species on Hamamelis japonica | Gall phenotype and gene-expression patterns | Shows that related gall inducers and their phenotypes can be compared; it does not establish one mechanism that explains every gall. |
The comparison highlights why gall research needs to identify both the insect and the host. A gene associated with pigment production in a witch-hazel gall cannot automatically explain a grape gall’s cell division or nutritive tissue. Different studies also measure different evidence: a visible trait, transcript levels, tissue development or the effect of removing an insect. Those observations answer related but distinct questions.
Do aphids change plant genes or edit plant DNA?
The evidence described here supports changes in host gene expression and development, not direct editing of plant DNA by aphids. For example, the H. cornus finding concerns the transcription of plant genes involved in pigment production. The phylloxera study measured host transcription across gall stages and interpreted patterns in developmental pathways. Neither result means the aphid rewrote the plant’s genetic sequence.
This distinction matters because the word “manipulate” can imply more certainty or direct control than the evidence warrants. Researchers observe associations and expression patterns, then test possible mechanisms. A proposed salivary effector could influence how host cells behave without changing the host’s DNA. The exact molecular interaction between candidate aphid proteins and plant processes is not yet established for the complete gall-building process.
What scientists still do not know
The full chain from aphid signal to a finished gall has not been resolved. The current evidence supports sophisticated host developmental manipulation, but leaves key questions about the first signals, their targets and how tissue-level outcomes are coordinated.
- Which candidate proteins have a demonstrated function? The bicycle-family survey identifies many possible effectors, but the predicted proteins are not all functionally validated.
- How does a candidate signal reach and affect plant cells? DGC is proposed to be secreted from aphid salivary glands into the plant, but the complete delivery and action mechanism remains uncertain.
- How do molecular changes produce particular gall forms? Expression data point to pathways that may matter, but do not alone prove how a given shape, tissue or developmental sequence arises.
- How far can results be generalized? The witch-hazel aphid and grape phylloxera examples involve different insects and hosts. They are useful together as evidence that insects can be associated with major changes in plant development, but one system cannot stand in for all others.
The most reliable account is therefore specific rather than universal: in H. cornus, dgc expression is linked to host pigment-gene transcription and gall color; in grape phylloxera, gall development coincides with extensive host gene-expression changes and altered developmental processes; and in some social aphids, galls shelter colonies across generations. These findings illuminate different parts of the problem without yet explaining every step that turns plant tissue into a gall and nursery.
