How a Shape-Shifting Nanocomplex Is Designed to Target Tumour Cells

A 2025 preclinical study describes an ursolic-acid and polypeptoid nanocomplex that changes structure in response to tumour-associated conditions. Its reported animal findings are not evidence of benefit in people.

Researchers reported a preclinical nanocomplex that changes its structure in response to conditions associated with tumours and cancer cells. In a 2025 study, the team combined ursolic acid with bioactive polypeptoids: the assembly forms fibres in neutral conditions, clusters in acidic conditions, and is designed to release its components after encountering oxidative conditions inside cells. The reported animal-model results are promising, but they do not show that the treatment works in people or is available to patients.

What the nanocomplex is

The system pairs ursolic acid (UA), a natural pentacyclic triterpenoid, with bioactive polypeptoids that act as assembly cofactors. The researchers say UA’s poor solubility and bioavailability have hindered its clinical use. Their approach is to organize it into a responsive nanoplatform rather than use UA on its own.

Polypeptoids are synthetic molecules related to peptides. Study coauthor Jing Sun told Chemistry World that they “mimic natural proteins but are more flexible, allowing controlled self-assembly of UA.” In this design, that flexibility helps guide how the components assemble and respond to their surroundings.

How the shape changes

“Shape-shifting” describes stimulus-responsive self-assembly, not a particle making decisions. The structure reported in the study depends on the chemical conditions it encounters.

Fibres in neutral conditions

Under neutral conditions, the assemblies retain a helical fibrous structure. This is the form researchers describe before the system encounters the more acidic conditions associated with the tumour microenvironment.

Virus-like clusters in acidic conditions

In acidic tumour-microenvironment conditions, the fibres transform into clusters of assembly subunits described as virus-like. The authors propose that this change helps the material penetrate more deeply into tumours. That is a proposed delivery advantage in the experimental system, not evidence that the material selectively reaches every tumour in a patient.

Release after cellular uptake

After uptake by cells, the nanoparticles are reported to escape into the cytoplasm and accumulate around mitochondria. Oxidation of thioether bonds is reported to trigger release of UA and polypeptoids. The authors associate this release with mitochondrial damage and apoptosis, a form of programmed cell death. Some particles reportedly reassemble into fibres inside cells, which may extend how long the material remains there.

What the experiments reported

The study abstract reports that the combined platform had significantly higher therapeutic efficiency than its individual components, including UA and polypeptoids. It also reports in vivo tumour-growth suppression and reduced metastasis. The abstract available for these findings does not provide numerical effect sizes or enough detail to establish the scale of the effects, so those outcomes should not be translated into a percentage or a prediction for patients.

The distinction between the combined platform and its ingredients matters: the experimental formulation depends on the polypeptoid cofactors and responsive assembly behaviour. It is not equivalent to taking an ursolic-acid supplement, and supplements should not be used as cancer treatment on the basis of this study.

What is not established

This is preclinical research, not a clinical treatment announcement. The reported in vivo findings do not establish human benefit, a safe dose, or how the nanocomplex would perform alongside standard cancer care. The study information described here also does not establish the animal species, numerical efficacy results, or safety outcomes.

Chemistry World quoted nano-biotechnologist Jiban Jyoti Panda as calling for more pharmacodynamic investigation and work on the material’s fate in the body, its clearance route, byproducts, and long-term toxicity. These questions are essential for judging whether a promising responsive design can translate into a treatment.
