Its key workaround is to install a stable benzofuran mask during peptide synthesis, then use ozonolysis to reveal a reactive salicylaldehyde ester for ligation. The work demonstrates a flexible synthetic method—not a validated drug or treatment.
How the peptide ligation strategy works
The method addresses a practical synthesis problem: salicylaldehyde esters on aspartate or glutamate side chains can be unstable during peptide synthesis and handling, making them difficult to install directly. The researchers instead used benzofuran as a masked form of that reactive group.

- Build the peptide with the masked handle. The benzofuran-containing group and a hydroxyl-amino partner—Dap–Ser or Lys–Ser—are incorporated at peptide side chains using standard Fmoc solid-phase peptide synthesis (Fmoc-SPPS).
- Remove protecting groups. After global deprotection, the benzofuran remains available for conversion.
- Reveal the reactive ester. Ozonolysis converts the benzofuran moiety into a side-chain salicylaldehyde ester.
- Join the peptide components. The resulting handle can react through Ser/Thr ligation or Cys/Pen ligation, forming a native peptidic linkage at the ligation site.
In this sequence, benzofuran is a synthesis-compatible precursor rather than the final ligation group. The strategy postpones formation of the reactive ester until the peptide has been assembled and deprotected. The primary paper in Chemical Science describes the chemistry and its demonstrated products.

The paper also reports joining peptides bearing the side-chain salicylaldehyde ester to partners carrying Dap/Lys–Ser or an N-terminal Ser, Thr, Cys, or Pen. These results show that the chemistry can create more than a simple linear peptide product; they are synthetic demonstrations of structural diversity.
Why benzofuran is useful—and what it cannot solve
Directly installing a salicylaldehyde ester on an Asp or Glu side chain is challenging because the active ester may not tolerate the steps and handling involved in peptide synthesis. A benzofuran mask provides a less reactive form that can be incorporated by Fmoc-SPPS and converted to the desired ester later.

The conversion step also imposes a sequence constraint. Ozone is a strong oxidant, and the study identifies cysteine, methionine, and tryptophan as incompatible with its ozonolysis conditions because these residues react during oxidation. As a result, the method cannot be applied indiscriminately to every peptide sequence.

Does this establish a therapeutic application?
No. The work establishes a method for making structurally varied peptides; it does not demonstrate clinical efficacy, a marketed therapeutic, or a validated protein–protein interaction (PPI) inhibitor. Constrained peptide architectures can be useful starting points for exploring PPI inhibition, but that is a potential application, not an outcome shown by this study.

Chemistry World’s coverage of the study describes the team’s future interests in higher-order cyclic peptides, PPI inhibitors, and peptide–drug conjugates. Those directions remain prospective. The distinction matters: demonstrating a route to a new molecular shape is an enabling chemistry result, not evidence that the resulting molecule works as a drug.
