Its predicted selectivity reportedly agreed well with experiment. The result is a focused demonstration for one reaction—not evidence that the method can design ligands reliably across chemistry.
What the model was designed to predict
The researchers, affiliated with Yale University and Boehringer Ingelheim Pharmaceuticals, modeled enantioselectivity in a Negishi cross-coupling reaction. Enantioselectivity describes how strongly a reaction favors one of two mirror-image products over the other, an important outcome when those forms can behave differently.

Rather than treating ligand selection as a structure-only problem, the approach used calculated information about how candidate ligands interact with the reaction’s transition states. A transition state is the high-energy arrangement of atoms along a reaction pathway; comparing transition-state free energies can help estimate which product-forming path is favored.
How 17 ligands became training data
The reported training set covered 17 phosphorus ligands. For those ligands, the model used density functional theory (DFT)-derived transition-state free energies, geometries, and electronic features. The Chemistry World report also says attractive dispersion effects were considered. The available account does not specify how the dataset was assembled or give the ligand structures, computational settings, or model architecture.

This is a different kind of input from a screen based only on measured experimental outcomes: the model drew on computed molecular and transition-state descriptors. That does not establish that calculations replace experiments. In this case, the predicted selectivity was checked against experimental results, and the report says the agreement was good.
What the model identified—and what mattered
The model helped researchers identify a ligand reported to be more enantioselective than the ligands used as inputs. Chemistry World describes the prediction as being in good agreement with experiment, but does not provide a numerical prediction error, detailed reaction conditions, or the extent of substrate testing. The result therefore supports the reported candidate in this reaction context; it does not quantify how accurately the model would perform on other ligands or reactions.

Among the features described as influential were:
- Valence electron population at a particular carbon center.
- Deviation from planarity by the metal center.
- A bond angle at a ring junction adjacent to phosphorus.
These features suggest that both electronic effects and three-dimensional geometry contributed to the model’s predictions. The report does not provide enough detail to assign them a universal role in ligand design or to interpret their importance beyond this model and reaction.

How far the finding can be generalized
The evidence described is bounded: one Negishi cross-coupling reaction, a training set of 17 phosphorus ligands, and a candidate whose predicted selectivity reportedly matched experiment well. The available report gives no numerical validation metric and does not establish transfer to other reaction classes. The researchers proposed that the approach could be useful where experimental data are limited, but that is a potential extension rather than a demonstrated capability.

Jonathan Goodman, a chemical informatics expert at the University of Cambridge who was not involved in the work, said the study showed that experimental data, DFT calculations, and human insight could be combined to generate an effective selectivity model, and that it was likely to inspire others. His comment describes the promise of the approach, not proof of broad performance.
