How a 2006 C-Glycoside Route Avoided Sugar Protecting Groups

A University of York route used an anomeric sulfone and a Ramberg–Bäcklund reaction to make C-glycosides, with a reported one-pot extension that avoided hydroxyl protection.

A University of York team reported a route to C-glycosides that uses a sulfur-containing intermediate and a Ramberg–Bäcklund reaction. Its key practical advance, as described in a March 8, 2006 Chemistry World report, was extending the sequence to a one-pot process that did not require protecting the sugar’s hydroxyl groups.

Why replace the oxygen in a sugar linkage?

Many natural carbohydrate units are joined through an O-acetal bond. Because such bonds can be susceptible to hydrolysis, chemists have explored C-glycosides, in which carbon rather than oxygen forms the linkage to the sugar. The aim is a more robust carbohydrate analogue.

Making these analogues can involve a practical complication: sugars have several hydroxyl groups, and conventional approaches may require temporarily protecting some of them, then removing those protections later. Each protection and deprotection operation adds manipulation to the synthesis.

How the reported route makes a C-glycoside

The news report describes an example starting from 2-deoxy-D-ribose, which has three hydroxyl groups. The sequence first installs a sulfur-containing group at the sugar’s anomeric carbon, then uses that intermediate to form the carbon-linked product.

  1. Install an anomeric sulfone. A sulfonyl Wittig reagent reacts selectively at the anomeric carbon, replacing its hydroxyl with a CH2SO2R group. This creates the sulfone intermediate.
  2. Convert the intermediate by Ramberg–Bäcklund reaction. Treatment with base and a halogenating agent is reported to replace the sulfone group with a double-bonded carbon, producing the C-glycoside.

The Chemistry World account says Richard J. K. Taylor’s group at the University of York extended the procedure to a one-pot process without hydroxyl protection. The report does not provide a full mechanism or experimental protocol, so the outline above should not be read as a recipe.

What the one-pot extension changes

The stated advance is avoiding the need to manipulate hydroxyl-protecting groups in this route. That could simplify preparation of C-glycosides and related compounds, since the protection and later restoration of sugar hydroxyls can cause difficulties. Paul Murphy of University College Dublin described the prospect as “the synthesis of a wide variety of novel C-glycoconjugates of biological relevance without the need to manipulate protecting groups, which can cause difficulties.”

“One-pot” describes the reported way of carrying out the sequence; it does not by itself establish a particular yield, shorter overall production time, or superiority to every other method. The report does not give comparative measurements or enough detail to assess those questions.

What the 2006 report does—and does not—establish

The report presents the route as a potential way to make robust analogues of natural carbohydrates and derivatives relevant to fields such as drugs and biosensors. Those are prospective applications, not evidence that the method produced a particular medicine, commercial biosensor, or clinical product.
