Haemoglobin Mimic Mops Up Cyanide in Promising Rat Study

A 2013 report described a synthetic haemoglobin mimic that bound cyanide quickly in vitro and revived poisoned rats. Human benefit and safety remain unestablished.

A synthetic molecule called Met-hemoCD3 was reported to bind cyanide faster than methaemoglobin or hydroxocobalamin in laboratory tests, and to revive cyanide-poisoned rats after respiratory arrest. Those are promising preclinical results—not evidence that it works safely in people or is available as an antidote.

How the haemoglobin mimic is built

In a 2013 report, Chemistry World described work by Koji Kano and colleagues at Doshisha University in Kyoto. Their synthetic supramolecule, Met-hemoCD3, was designed to imitate methaemoglobin’s ability to bind cyanide. It consists of an iron(III)-containing porphyrin held inside a capsule made from cyclodextrin dimers.

The report says the complex binds cyanide to form a non-toxic cyanide-bound complex under conditions described as those in the human body. That describes the reported experimental chemistry; it is not a finding about safety in patients.

What the experiments showed

Laboratory binding tests

In vitro tests reported in 2013 found that Met-hemoCD3 bound cyanide faster than methaemoglobin or hydroxocobalamin. Kano said: “The rate of cyanide binding is fast, which is important for emergency therapy.” A faster laboratory reaction is not, by itself, proof of faster or better treatment in people.

Rat poisoning experiment

In the reported animal experiment, rats poisoned with sodium cyanide received Met-hemoCD3 intravenously. The article says the treatment revived animals that had gone into respiratory arrest, while untreated rats died. This result is from rats, not a human trial, and does not establish that the compound would rescue people.

Why the result is not yet a better antidote

The Chemistry World report contrasted the candidate with nitrite antidotes, which work by oxidizing haemoglobin iron to produce methaemoglobin that can bind cyanide. It characterized that approach as slow and associated with harmful side effects. This is context from the 2013 report, not a current clinical comparison or treatment recommendation.

  • Event Memory records the last time carbon monoxide was detected or when the unit was last tested

Gerard Boss of the University of California San Diego acknowledged that “The compound can function as a cyanide scavenger,” but questioned whether it offered an advantage: “they have not compared it in vivo to currently available antidotes. All of their comparisons have been in vitro.” In other words, the reported laboratory speed comparison did not establish superiority over existing antidotes in living animals or people.

Boss also argued that intramuscular delivery would be more useful than intravenous treatment in a mass-casualty scenario. The reported rat treatment used an intravenous route; the article did not establish that the compound can be given intramuscularly.

Clinical status and limits of the 2013 report

Kano also said, “And we found no evidence of toxicity.” That was a remark reported in 2013, not proof of human safety. The report describes a research candidate and says further collaboration with medical doctors and industrial partners was needed to bring it into actual use. It does not establish human trials, approval, or clinical availability.

Chemistry World identified the original study as K. Watanabe, H. Kitagishi and K. Kano, “Angew. Chem., Int. Ed.,” 2013, DOI 10.1002/anie.201302470. The report is Emma Stoye’s article, published 3 June 2013: “Haemoglobin mimic mops up cyanide”. Its findings support further study of cyanide binding, but do not show that Met-hemoCD3 became a treatment.
