DP4-AI: Automating NMR Analysis to Compare Candidate Structures

DP4-AI automates NMR signal assignment to help compare proposed structures. Here’s what its workflow does—and why it is not an unconstrained structure generator.

DP4-AI was designed to automate part of NMR-based structure elucidation: it processes raw ¹H and ¹³C spectra, assigns experimental signals against calculated chemical shifts, and estimates which of a set of proposed structures best fits the data. It is a way to test candidate structures—not a system that independently invents an unknown molecule from a spectrum.

What DP4-AI does

When two proposed structures have similar one-dimensional NMR spectra, distinguishing them can require careful assignment of many signals. DP4-AI addresses that problem by automating analysis and assignment for candidate molecules, then comparing the resulting evidence. Its intended use includes resolving uncertainty between alternatives such as regioisomers or diastereomers.

The workflow begins with trial structures. For each candidate, the method uses density functional theory (DFT) to calculate chemical shifts for its atoms. It processes raw NMR data into experimental multiplet shifts and integrals, assigns calculated shifts to experimental peaks, and uses those assignments to produce a DP4 probability for each candidate.

That distinction matters: DP4-AI evaluates proposed structures against the spectrum. It does not search an unlimited space of possible molecules and return a structure without candidate input.

How it differs from standard DP4

Standard DP4 requires users to provide experimental peak locations and identify which atoms in a candidate molecule are chemically equivalent. DP4-AI’s stated purpose is to automate that human-prepared input by starting with raw NMR data and carrying out signal processing and assignment within the candidate-comparison workflow.

Automation changes the preparation and analysis steps; it does not remove the need to formulate plausible candidates. The result is useful when a chemist has structures to compare and wants computational evidence to help resolve uncertainty.

DP4-AI and Mnova have different roles

Aspect DP4-AI, as described in 2020 Mnova, as described in 2020
Primary role Assigns experimental signals and compares proposed structures using calculated shifts. Helps users process and interpret spectra.
Starting point Raw NMR data and trial structures. Spectrum processing and interpretation; the report does not specify a single required input format.
Calculated DFT shifts Part of the candidate-comparison workflow. Not described as part of the role discussed in the report.
Question addressed Which proposed structure best fits the data? How should the spectrum be processed and interpreted?

This is a limited distinction from Hannah Kerr’s 6 April 2020 Chemistry World report, not a current feature-by-feature product review. The report does not establish current software features, compatibility, availability, or commercial terms for either program.

What the reported evaluation and timing show

Chemistry World reported that the evaluation covered 47 molecules, averaging 3.49 stereocentres per molecule. It also reported that DP4-AI could complete a full calculation in about 60 seconds per molecule, compared with a manual process estimated to take up to eight hours.

Rank #3

Those figures describe the report’s 2020 account; they are not an independent benchmark or a guarantee of present-day speed. The report cites A. Howarth, K. Ermanis and J. M. Goodman’s paper, “DP4-AI automated NMR data analysis: straight from spectrometer to structure,” in Chemical Science (2020), DOI 10.1039/D0SC00442A. The performance figures here are attributed to Chemistry World’s report, rather than presented as independently verified measurements.

Why raw-data context matters

Automating signal analysis depends on being able to connect spectra to usable experimental and structural context. Goodman noted that raw NMR data may be retained while labels and corresponding structures remain in lab books, making that information difficult to retrieve alongside the data. Without durable, accessible labels and structure records, a raw spectrum can be harder to interpret, reproduce, or reuse later.

Rank #4

The point extends beyond any one analysis program: preserving raw spectra together with clear metadata and the relevant candidate structures makes computational and human review more practical.

Does automation replace learning to interpret spectra?

No. DP4-AI was presented as a way to speed up and support assignment, not as a reason to abandon chemical judgment. The user still needs to propose appropriate structures and assess whether the result makes sense in the context of the chemistry. Goodman compared the role of such tools to calculators: they can make complex work faster and more accurate without making the underlying skill irrelevant.

What is established—and what is not

The 2020 account describes a method for automating ¹H and ¹³C NMR analysis and comparing candidate structures through assignments informed by DFT-calculated shifts. It also reports an evaluation and timing figures, and identifies DP4-AI as open-source at that time. Ariel Sarotti of the National University of Rosario praised the group’s work on tools for structural and stereochemical assignment and predicted in 2020 that the open-source method would become popular.

That historical description does not establish whether DP4-AI is currently maintained, compatible with present-day systems, or available under unchanged terms. Those details, as well as current adoption, are not established by the 2020 report.
