A roughly 3-gram Chinese red-headed centipede subdued a mouse weighing about 45 grams within 30 seconds in an observation reported by researchers. They traced much of the venom’s effect to a peptide they named Ssm Spooky Toxin, or SsTx, which blocks certain potassium channels. The finding helps explain the toxin’s potent effects in experimental models; it does not mean SsTx is the only active ingredient in centipede venom or establish a treatment for human bites.
What is the deadly component of centipede venom?
SsTx is a peptide purified from the venom of the Chinese red-headed, or golden-head, centipede Scolopendra subspinipes mutilans. In their 2018 study, Luo and colleagues identified it as a major contributor to the venom’s activity, particularly its effects on blood vessels and the cardiovascular system. The paper’s title, “Centipedes subdue giant prey by blocking KCNQ channels,” describes the mechanism the authors investigated.

The mouse observation was striking: the researchers reported that a centipede weighing about 3 grams subdued a mouse weighing about 45 grams within 30 seconds. That is a result from this study, not a typical or guaranteed time for an attack, and it should not be generalized to every centipede species or bite. Read the study in PNAS.

The mature SsTx peptide consists of 53 amino acids and has a reported molecular weight of 6,017.5 daltons. The precursor protein contains 76 amino acids; removal of a 23-amino-acid signal peptide produces the mature toxin. The researchers reported two disulfide bridges and solved the peptide’s structure using solution NMR. The sequence is recorded as GenBank accession MG585384, and its structure is deposited as PDB entry 5X0S. View the SsTx structure in the Protein Data Bank.

How does SsTx affect KCNQ potassium channels?
KCNQ channels help regulate the movement of potassium ions across cell membranes. By blocking these channels, SsTx can disrupt electrical activity in tissues that depend on them. The study found inhibition of KCNQ1, KCNQ2, KCNQ4 and KCNQ5 channels in laboratory assays, with reported IC50 values of about 2.5–2.8 micromolar across the tested channel types. An IC50 is the concentration that produces 50% inhibition under the assay conditions; it is not a measure of how much venom a person would receive in a bite.
The authors’ experiments pointed to the channels’ outer pore region as the toxin’s interaction site. They also found that changing either of two positively charged SsTx residues, arginine 12 or lysine 13, substantially weakened inhibition. Those results support a role for these residues in the toxin’s binding surface.

What evidence links SsTx to the venom’s effects?
The study connected several kinds of evidence, from isolated channels to tissue and animal experiments:
- Channel assays: Purified SsTx inhibited several KCNQ channel types, and the authors tested how changes to the toxin affected that inhibition.
- Vessel assays: Removing SsTx from crude venom greatly reduced its activity in the vessel assay reported by the researchers. This supports SsTx as a major cardiovascular-active component of this species’ venom, not as its only biologically important component.
- Animal and other experimental models: The paper reported cardiovascular effects as well as nervous-system and respiratory effects. These findings characterize experimental effects; they do not establish the course or severity of every human bite.
The evidence therefore supports a potent role for SsTx within the venom mixture. It does not show that every effect of the whole venom comes from this one peptide.

Does retigabine treat a centipede bite?
The researchers tested retigabine, a drug that opens KCNQ channels, and reported that it reversed effects in their experimental work. They proposed that the approach might have therapeutic relevance. This is an experimental result, not evidence that retigabine is a proven or approved treatment for centipede bites in people. The study does not establish a human dose, safety profile, or clinical protocol, so it should not be used as advice to take retigabine after a bite. The PNAS paper describes the experiments.

Luo and colleagues’ paper was published in PNAS on January 22, 2018. Its findings concern one centipede species and laboratory, tissue and animal models. They explain a mechanism by which SsTx can contribute to venom toxicity, but they are not a survey of all centipedes, a prediction for an individual bite, or proof of an effective human treatment.

For wider context, Chemistry World reported on the study on January 25, 2018. The article quoted Natural History Museum venom expert Ronald Jenner describing the work as the first study to show that a potassium-channel-blocking neurotoxin may be a major weapon for overwhelming large vertebrate prey.
