RNA interference (RNAi) has reduced activity of the bovine PRNP gene in cultured cells and in one reported calf, but it has not been shown to make cattle resistant to BSE. In that calf, the gene’s messenger RNA fell substantially while the measured prion protein fell much less, and the calf was studied only to 20 days of age—not challenged with BSE.
What RNAi does—and what the cattle studies found
PRNP encodes cellular prion protein, PrPC, which is relevant to prion propagation. RNAi uses short RNAs matched to a target sequence to reduce the target messenger RNA. Lowering PRNP RNA is therefore a molecular result, not by itself evidence that an animal cannot become infected.

Cell-culture work tested ways to lower PRNP
A 2007 study tested bovine PRNP-targeting small interfering RNAs in cultured cells, varying the promoter or vector, target site, and RNA length. Under the conditions tested, a 21-nucleotide sequence produced the strongest knockdown. The authors described the findings as a basis for further studies in vivo; they did not report BSE-resistant cattle. Read the 2007 study.

The 2011 calf had different RNA and protein results
In a 2011 study, researchers combined RNAi with somatic cell nuclear transfer (SCNT) to produce a calf with reduced PRNP expression. They transferred 42 reconstructed embryos to 11 recipients. Only embryos using the tRNA-promoter vector established pregnancies; the report describes four aborted fetuses, one stillbirth, and one live-born calf.

At 20 days old, the calf was killed for analysis. In sampled nervous tissues, average PRNP transcript levels were 35% of the control calf’s level, but measured PrPC protein averaged 86% of the control. The authors concluded that RNAi reduced the messenger RNA but did not reduce the protein sufficiently under those conditions. These are measurements from one experiment, not estimates of disease resistance. Read the 2011 study.
Why lower PRNP RNA does not prove BSE resistance
Messenger RNA is used to make protein, but a reduction in the RNA does not necessarily produce an equivalent reduction in the protein measured in tissue. The 2011 calf illustrates that gap: its transcript measurement was substantially lower than the control’s, while its measured PrPC level was much closer to the control value.

Neither result shows whether an animal resists infection. That requires evidence about disease-relevant outcomes, such as a properly conducted live-animal challenge. The cited RNAi calf report does not establish that the calf was exposed to BSE, and it provides no challenge result. It should not be described as BSE-resistant.

The knockout study’s in-vitro finding is meaningful but narrower than a live-animal test: it shows that brain homogenates from those cattle resisted prion propagation under laboratory conditions. It does not establish the outcome of infecting a living animal. Read the 2007 knockout study.

The 2025 Cas9 work is a separate genome-editing model, not an RNAi experiment. Its description of the animals as a resistance model should not be mistaken for a reported BSE challenge result. Read the 2025 study.
What would be needed to show resistance?
Evidence of reduced PRNP RNA or PrPC is not the same as evidence that cattle resist BSE. A stronger claim would require results that connect the genetic change to outcomes after exposure, alongside adequate follow-up and assessment of animal health. The studies summarized above used different methods and endpoints, so they cannot be treated as a head-to-head comparison or ranked as if tested under one protocol.

A USDA National Agricultural Library project description says that a portion of cattle in the described work would be challenged with infectious BSE or bovine scrapie to assess resistance. That page describes planned work; it is not proof that the challenge was completed or that resistance was demonstrated. Read the USDA project description.

Genetic engineering has been used to reduce or disrupt PRNP in cattle-related research, but the evidence must be described by the intervention and endpoint. RNAi showed gene-expression knockdown, including in one short-lived calf; the cited RNAi work did not show that the animal resisted BSE. Knockout and CRISPR-based studies are distinct approaches, and the cited findings do not amount to a live-animal BSE challenge result.
