A 2011 study described an enzyme-assisted way to make defined, short-chain heparin analogues in fewer steps than conventional chemical synthesis. The researchers reported rabbit in-vitro pharmacological properties comparable to fondaparinux (Arixtra), but this was preclinical work—not evidence of a treatment proven in people, a cheaper medicine, or a commercially established manufacturing process.
What the enzymatic route was designed to make
The work focused on ultra-low molecular weight heparin (ULMWH): short, structurally defined heparin-like chains. Unlike unfractionated heparin, which the 2011 account says is obtained mainly from porcine intestinal lining, the short-chain synthetic agent discussed in the study was made through a lengthy chemical route.

The researchers’ aim was not simply to reproduce a broad mixture of heparin molecules. They sought to assemble homogeneous oligosaccharides—chains with a defined structure—so that specific variants could be made and studied.

- Build a short sugar-chain backbone. The reported process used uridine diphosphate (UDP) sugars and enzymes derived from Escherichia coli to assemble the oligosaccharide backbone.
- Modify the backbone enzymatically. Epimerase and sulfotransferase enzymes altered the chain, producing a homogeneous oligosaccharide analogue of fondaparinux.
- Make defined products. The account says the approach produced two homogeneous ULMWHs in fewer steps, with higher yield and purity than the standard chemical synthesis route, as described by the researchers.
The accessible account supplies no exact step counts, yields, or purity measurements, so those advantages cannot be quantified from it. Chi-Huey Wong also pointed to regeneration of PAPS and sugar-nucleotide cofactors as a way that could reduce costs and avoid inhibition problems; the account does not provide a cost calculation.

What the evidence does—and does not—show
The 2011 news account reports pharmacological properties comparable to Arixtra in rabbit in-vitro tests. This is a preclinical comparison, not a human trial or proof that the synthetic compounds prevent or treat thrombosis in patients.
- Supported by the account: an enzyme-based route for making defined short-chain analogues, and a reported rabbit in-vitro comparison with fondaparinux.
- Not established by the account: efficacy or safety in people, regulatory approval, routine clinical use, manufacturing at commercial scale, or replacement of porcine-derived heparin.
- Still prospective: lower production costs and the development of variants with different half-lives or reversibility. These were goals or possibilities, not demonstrated clinical improvements.
Why the approach mattered to researchers
Enzymatic assembly offered a potential way to explore structures that could be difficult to produce through the conventional chemical route. Robert Linhardt, identified in the report as a research-team leader, said: “Since our method is faster and simpler than standard chemical synthesis we can also explore many new structures allowing us to improve on and expand on the current properties of ULMWHs.”

Wong, described as a chemoenzymatic synthesis researcher at Scripps Research Institute, called it “the best and most practical way of making certain types of short-chain heparins on large scales.” That is an expert assessment quoted in the report, not evidence that large-scale production was subsequently achieved.

Study identification and limits on current status
The news account cites Y. Xu and colleagues, Science 334, 498 (2011), DOI 10.1126/science.1207478. The account does not establish what happened to the route after the study, so it cannot support claims about its present-day clinical, regulatory, or commercial status.
