Choose a xenograft model for the biological question it can answer—not because one model is universally “best.” CDX models are often practical for controlled screening; PDX models can better represent patient-tumor features; humanized mice are suited to selected questions involving human immunity; and organoid-derived xenografts can carry patient-derived culture findings into an in vivo setting. Each has limitations that affect interpretation, and no single reporting standard reviewed here covers all four model classes.
What distinguishes the four model types?
The key differences are the source and history of the tumor material, the host’s immune context, and which parts of tumor biology the study is designed to represent. The comparative strengths and limitations below are described in Franklin et al.’s 2022 review of preclinical immuno-oncology models.

How should you choose a model for a study?
Treat model selection as a set of trade-offs, not a ranking of fidelity. A model is useful when the biology it represents aligns with the question and the features the experiment needs to test.
Start with the biological question
- For controlled screening or pharmacology: a well-characterized CDX may be a practical choice when a cell-line-based tumor and the host’s immune deficiency are compatible with the question.
- For patient-tumor features or heterogeneity: consider a PDX, while accounting for availability, establishment demands, and the absence of a human immune system in standard PDX hosts.
- For human immune components or human-specific targets: consider a humanized xenograft only when those components are central to the question. Humanization is not a general upgrade for every xenograft experiment.
- For extending organoid findings in vivo: consider an organoid-derived xenograft, and document both how the organoid was generated and how the resulting model was validated.
Check practical and interpretive constraints
Before committing to a model, weigh biological fit alongside immune context, model availability and characterization, establishment time, study duration, cost, and reproducibility. These factors can change what is feasible and what conclusions the resulting experiment can support; they do not establish that one model class is universally preferable.

What should researchers standardize and report?
Reporting should let readers assess where the tumor material came from, how the model was produced and maintained, what the host can represent, and how outcomes were defined. PDX-MI, the 2017 Minimal Information for Patient-Derived Tumor Xenograft Models standard, is a reporting baseline specifically for PDXs. It addresses tumor clinical attributes, implantation and passage procedures, host strain, quality assurance, and model use; it is not a standard spanning every model class in this article.
Report core details for every model class
- Tumor source and identity: state the source and identity of tumor material, plus authentication or characterization methods.
- Model history: describe passage or culture history, baseline tumor characteristics, and model provenance. For PDXs, include passage information and sufficient deidentified tumor and treatment history when available.
- Host and implantation: identify the host strain and immune status, implantation site, and implantation method.
- Study conduct: report allocation and treatment details, monitoring, exclusions, and outcome definitions.
- Rationale: explain why the chosen model answers the stated biological question and identify its relevant limitations.
The NCI Patient-Derived Models Repository illustrates the value of access to early-passage, molecularly characterized, clinically annotated patient-derived models and repository-linked information. Such characterization and provenance can help readers judge comparability, but do not remove the need to report how a particular experiment was conducted.

Apply PDX efficacy recommendations to PDX studies
The NCI PDXNet consensus recommendations published in Molecular Cancer Therapeutics in 2024 call for clinically relevant doses and schedules when known, tolerability monitoring, and evidence across at least two clinically relevant models for PDX efficacy studies. They also recommend at least two measures of antitumor activity, including a tumor-volume measure referenced to baseline and a second measure such as tumor-volume change or event-free survival. The consensus states: “Antitumor activity is best assessed using a combination of two or more metrics.” These are recommendations for PDX efficacy assessment, not a universal protocol for every xenograft experiment.

The 2024 OBSERVE guideline addresses refinement and reporting in rodent cancer studies. It is relevant as general animal-model guidance; PDX-MI remains the PDX-specific reporting baseline. The sources discussed here do not establish one standard that covers CDX, PDX, humanized, and organoid-derived xenografts together.

How should results be interpreted across models?
Differences in tumor provenance, culture history, host immunity, and model establishment affect what a result means. A response in a clonal cell-line tumor does not by itself establish how heterogeneous patient tumors will respond. A PDX can better preserve some patient-tumor features, but a standard immunodeficient PDX host cannot answer a question that depends on a human immune system. A humanized model adds selected human immune components, but incomplete reconstitution, donor effects, and graft-versus-host disease remain relevant constraints. An organoid-derived xenograft connects culture and in vivo work, but neither setting represents every component of the tumor microenvironment.
Accordingly, distinguish the result observed in the chosen model from any broader biological or therapeutic claim. Report enough information about the model and experiment for readers to judge how far that result can reasonably be generalized.
