The discovery helps explain why orange cats are usually male and why female cats can have orange-and-dark tortoiseshell or calico patches. It closes a molecular gap dating back to the 1961 X-inactivation hypothesis—not a continuous 60-year search by one team.
What did researchers discover?
In 2025, independent teams led by researchers at Kyushu University in Japan and Stanford reported that orange cat coloration is associated with a deletion in ARHGAP36, an X-linked gene. The deleted stretch is non-coding: it does not remove instructions for building the gene’s protein. OMIA describes the variant as a 5.1-kilobase intronic deletion; the National Institute of Genetics rounded it to about 5,000 DNA bases.

The Kyushu team compared DNA from 18 cats—10 orange and eight non-orange—and reported that the association also held in 49 additional cats. Those are study sample counts, not estimates of the proportion of cats that are orange or male. The team also found higher ARHGAP36 activity in melanocytes from sampled orange patches of calico-cat skin, along with changes in the activity of pigment-related genes. The Stanford-led study independently reported the same gene and a similar deletion.
The finding identifies a genetic variant associated with the color, but the full chain of molecular events that shifts pigment toward orange is not yet understood. The Kyushu University announcement described the discovery as a longtime dream for lead author Hiroyuki Sasaki, a geneticist at the university.

Why are orange cats usually male?
The orange variant is on the X chromosome. In the usual XX/XY pattern, an XY male has one X chromosome; if it carries the orange variant, he can show orange coloration. An XX female has two X chromosomes and generally needs the orange variant on both to be uniformly orange.
This is why orange cats are usually male, while orange females are less common. “Usually” matters: the XX/XY explanation describes the typical inheritance pattern, not every possible chromosomal or developmental case. The available sources do not establish a reliable population percentage for how many orange cats are male.

Why do tortoiseshell and calico cats have patches?
An XX female can inherit the orange variant on one X chromosome and a non-orange version on the other. Early in development, different cell lineages switch off one or the other X chromosome. As those cells multiply, patches of fur reflect which X remains active: some areas express orange, while others express the darker, non-orange coloration. This mosaic pattern produces tortoiseshell coloring.
Calico cats have the same orange-and-dark mosaic, plus white areas. White spotting is a separate factor; it is not the orange variant in ARHGAP36. The size and distribution of white patches contribute to the familiar calico appearance.

What does the discovery establish—and what does it not?
What the evidence supports
- Two independent 2025 studies identified a similar deletion in X-linked ARHGAP36 associated with orange coloration.
- The Kyushu team’s samples linked the deletion with coat color and showed higher ARHGAP36 activity in melanocytes from sampled orange patches.
- The X-linked inheritance pattern explains the usual difference between uniformly orange coats and orange-and-dark mosaics.
What remains open
- The downstream biological process by which altered ARHGAP36 activity changes pigment is not fully known.
- The studies do not show that orange fur predicts a cat’s personality. Sasaki said there is no scientific evidence for such a link yet.
- The findings do not establish a population-wide percentage of orange cats that are male.
How did the search reach this point?
Researchers had known for decades that orange coloration followed an X-linked inheritance pattern, which led to the 1961 X-inactivation hypothesis for patchy coats. Mapping narrowed the location over time; OMIA notes that a 2018 study reduced it to a 1.5-megabase region. The 2025 studies advanced from that broader location to ARHGAP36 and the associated deletion. The 1.5-megabase interval was an earlier mapping region, not the size of the deletion.
Kyushu University announced its findings on May 15, 2025, and the National Institute of Genetics followed on May 16. The Stanford-led paper appeared in the same 2025 issue of Current Biology; its PubMed record lists a June 23 publication date. The papers are independent corroboration, not one combined experiment.
