Electron spin may offer a clue to how general anaesthetics work, but it is not an established explanation for unconsciousness. In a 2014 study, researchers reported a change in electron spin resonance (ESR) signals from anaesthetic-exposed fruit flies. Because the flies had to be chilled and immobilised, and the proposed connection between the signal and unconsciousness was not demonstrated, the finding remains suggestive rather than conclusive.
What does the electron-spin theory propose?
General anaesthetics include diverse molecules that can produce unconsciousness. Conventional accounts often focus on how these molecules interact with receptors or ion channels. Luca Turin and colleagues proposed a different possibility: an anaesthetic might alter the electronic state of proteins, and that change could contribute to loss of consciousness.

Turin described the idea this way: “What we are suggesting is that somehow the anaesthetic molecule is putting an electronic spanner in the works.” The metaphor captures the proposed disruption; it does not establish that such a disruption is what causes unconsciousness.

Simon Hadlington’s 12 August 2014 report in Chemistry World describes researchers measuring ESR signals from groups of fruit flies. ESR detects signals associated with unpaired electrons. The team cooled roughly 30 flies to a few degrees above freezing so they would be motionless, then compared ordinary flies with an anaesthetic-resistant mutant strain.

- In ordinary flies, anaesthetic exposure was associated with a jump in the ESR signal compared with resistant flies.
- The researchers interpreted the change as an increase in unpaired electrons.
- Melanin-depleted mutants showed a similar result, which addressed melanin as a possible source of the signal.
These are experimental observations reported for chilled flies, not a clinical finding or evidence that the same signal explains anaesthesia in people.
How did the researchers connect the signal to proteins?
Turin proposed that an anaesthetic molecule could enter a hydrophobic pocket in a protein and withdraw electrons, perturbing electron currents. The team also used density-functional-theory simulations with a model hydrophobic peptide and xenon; the report says the simulations indicated that such an interaction was feasible.

That feasibility result does not show that the proposed molecular event occurred in the flies, nor that it caused their behavioural response. The bridge from a possible electronic change in a protein to unconsciousness remained speculative.

Why is the interpretation disputed?
The central concern is the experimental condition needed to obtain the signal: the flies were chilled and immobilised. Michael Perouansky questioned whether the effects occur at normal fly temperature and at concentrations that actually anaesthetise flies, rather than at several times those concentrations. He put the distinction plainly: “A very different question is whether these effects have anything to do with the behavioural state of anaesthesia.”

Stuart Hameroff also argued that chilled, temporarily brain-dead flies were a poor model for anaesthetic action and raised the possibility that the ESR signal could have a metabolic source. He remarked: “Mixing metaphors, Turin et al are in the right ballpark, but barking up the wrong tree.” That was a criticism of the interpretation, not a demonstration of an alternative mechanism.
What was Turin’s response?
Turin conceded that the setup was not ideal and said the team was working on ways to paralyse flies at room temperature. He argued that the difference between ordinary and anaesthetic-resistant flies still suggested an electronic response worth investigating. That defence supports further attention to the observed difference; it does not resolve whether it is relevant to anaesthesia or unconsciousness.

What can readers conclude?
The report presents an intriguing measurement and a proposed mechanism, not a settled account of how general anaesthetics work. The study is Luca Turin et al., “Electron spin changes during general anesthesia in Drosophila,” published in Proceedings of the National Academy of Sciences (PNAS), DOI 10.1073/pnas.1404387111. The key unanswered question is whether an electronic effect seen under the study’s chilled conditions occurs during anaesthesia under biologically relevant conditions and helps cause the loss of consciousness.
Hameroff’s University of Arizona publications page lists his comment on the Turin et al. paper, documenting contemporary discussion rather than establishing later scientific consensus.
