The correct sequence depends on the cell format and chemistry, whether the cell is dry or already filled, whether you are measuring formation, SEI growth or parasitic reactions, and how the calorimeter holds and contacts the cell.
Across published pouch-cell experiments, the preparation usually includes controlled drying when a cell is opened, inert-atmosphere handling, electrolyte filling and sealing, a defined wetting and formation history, optional degassing, and thermal equilibration in the calorimeter before electrochemical measurement. The numerical conditions below are examples from particular studies, not a general standard operating procedure.

Start by defining the measurement, not the oven setting
Sample preparation should be designed around the question the heat-flow experiment must answer. A full cell being monitored during cycling has different requirements from a voltage-hold experiment intended to quantify parasitic reactions. A study of SEI formation may also require a different starting state from a study of high-voltage electrolyte oxidation.
Published examples include NMC442/graphite pouch cells, anode-free pouch cells and wound-cell work focused on SEI formation. These architectures do not share the same current collectors, electrolyte exposure, formation history or gas-generation behavior. Before touching a cell, record:

- Cell architecture, electrode chemistry and nominal capacity.
- Whether the cell arrived dry and sealed, already filled with electrolyte, or was assembled in the laboratory.
- The electrochemical event to be measured: formation, SEI development, voltage-dependent parasitic heat or complete-cell heat generation.
- Whether opening the package, replacing electrolyte or releasing gas is part of the procedure.
- The calorimeter holder, mechanical constraint, temperature and electrical-control arrangement.
These choices determine whether drying, glove-box work, degassing and a particular formation schedule are necessary. They also determine whether two heat-flow datasets are genuinely comparable.
Incoming-cell condition determines the first handling step
Dry, sealed cells
A dry pouch cell that must be filled in-house is not ready to enter the calorimeter. Opening it creates a moisture-control problem, and the drying condition becomes part of the cell history. In the detailed NMC442/graphite example reported by Downie, Hyatt and Dahn, machine-made 260 mAh pouch cells were supplied dry and sealed. The cells were opened before drying and electrolyte addition.
Prefilled cells
A prefilled and sealed cell may not need the same drying and filling sequence. Instead, document the electrolyte already present, the time since filling, storage conditions and any prior formation or cycling. Do not apply a dry-cell protocol to a prefilled cell simply because it was used in another publication.

Cells assembled in-house
For an in-house assembly, capture the electrode, separator, electrolyte formulation and amount, sealing method and rest time before formation. Small differences in fill quantity or sealing can alter wetting, gas evolution and the heat signal that follows.
Drying and atmosphere control when a pouch is opened
If the protocol opens a dry pouch, moisture removal and air exclusion are central preparation controls. In the Downie, Hyatt and Dahn procedure, opened cells were placed in a heated vacuum oven at 80 °C overnight, approximately 14 hours. After drying, they were transferred directly to an argon-filled glove box without exposure to ambient air, where electrolyte was added.
“After drying, the cells were directly transferred to an argon-filled glove box without exposure to ambient air, where they were filled with electrolyte.”
The 80 °C and approximately 14-hour values belong to that cell and study. They should not be treated as a universal drying specification. A different separator, binder, pouch construction or electrolyte may require another validated condition. The reproducible practice is to report the temperature, pressure or vacuum condition, duration, transfer path and atmosphere, rather than writing only “the cells were dried.”

Electrolyte filling and sealing
Fill and seal every cell as consistently as possible. Record the electrolyte identity and formulation, the amount added to each cell, whether the cell was under vacuum during sealing, and the sealing equipment or method. The cited NMC442/graphite work used a specified electrolyte formulation and vacuum-sealed the pouch. A separate manufacturer-described battery calorimetry example also filled and vacuum-sealed dry pouch cells.
Electrolyte amount is not a clerical detail. It affects how quickly the separator and electrodes wet, the quantity of material available for parasitic reactions and the gas volume that may appear during formation. If cells receive different amounts, keep those differences attached to the calorimetry results instead of treating the cells as interchangeable replicates.
Wetting, formation and the initial electrochemical state
Wetting hold
After filling, allow the electrolyte to penetrate the electrode stack before interpreting heat flow. In the NMC442/graphite example, cells were held at 40 °C and 1.5 V for 24 hours to promote complete wetting before staged formation.
An anode-free pouch-cell study used an approximately 24-hour hold at 1.5 V for a different but related reason: ensuring positive-electrode wetting while preventing corrosion of the copper current collector. The same voltage and approximate duration therefore cannot be presented as a universal recipe; the rationale is tied to that cell design.

Formation creates the electrochemical history that the calorimeter will observe. Report every relevant stage: voltage limits, current or C-rate, rest periods, temperature, number of cycles and the point at which the cell is considered formed. “Formed” is not a sufficient description when comparing heat signals from different laboratories.
Why starting history matters
SEI growth, electrolyte decomposition and gas generation can continue during formation. A cell measured immediately after filling is not equivalent to one measured after staged formation and rest. The measured heat may reflect the preparation history as much as the nominal chemistry, so the starting state must be defined before the calorimeter run.
Degassing: include it only when the cell history requires it
Gas release is conditional, not an automatic step for every lithium-ion cell. In the NMC442/graphite protocol, formation was staged and the cells were opened under argon to release gas, then resealed after the formation steps. A manufacturer-published experimental example likewise describes opening cells to release gas and resealing them.

If your protocol includes degassing, document when it occurred relative to each formation stage, the atmosphere used for opening, how the pouch was resealed and whether the cell was weighed or otherwise checked. If the cell design and study do not call for gas release, do not insert a degassing cycle merely to match another paper. Opening a formed cell changes its handling history and may introduce a new opportunity for moisture exposure.

Calorimeter setup and thermal equilibration
Match the holder to the cell
The holder or measurement fixture must accommodate the pouch or wound cell while maintaining the intended mechanical constraint. Report how the cell was positioned, whether pressure or stack constraint was applied, how electrical leads were connected and whether the same arrangement was used for every sample. Mechanical loading can be a confounding variable when comparing cells with different swelling or gas histories.
Set and verify the temperature
In the Downie, Hyatt and Dahn study, measurements used a TAM III calorimeter set to 40.0000 ± 0.0001 °C. That precision describes the reported instrument setpoint for that experiment, not a universal requirement for all battery calorimetry. The appropriate temperature depends on the chemistry, the reaction being isolated and the instrument’s validated operating range.
Allow the cell to reach thermal equilibrium
After formation, the same study allowed 24 hours of open-circuit thermal equilibration before cycling measurements. This rest lets the cell and holder approach a stable baseline so that the subsequent heat signal is less likely to be dominated by installation or temperature transients. State the equilibration duration, whether the cell was at open circuit, the temperature and the criterion used to begin data collection.
Connect electrochemical control
The cited experiment connected the cells to an automated battery tester after equilibration. A complete method therefore specifies both instruments: the calorimeter and the electrical program. Include the voltage or current hold, charge and discharge limits, rest periods, scan or cycling sequence and the timing relationship between electrical control and heat-flow acquisition.
A practical preparation sequence
The following sequence translates the published examples into a reproducibility checklist. Adapt each step to the cell and instrument rather than copying the numbers automatically.
- Define the objective and architecture. State whether the run targets full-cell heat, voltage-dependent parasitic reactions or SEI formation, and identify the chemistry, format and capacity.
- Inspect and document the incoming state. Record whether the cell is dry and sealed, prefilled or assembled in-house, along with storage and prior cycling information.
- Open only when the method requires it. If a dry pouch must be filled or degassed, plan the opening, atmosphere and resealing steps before beginning.
- Dry under a documented condition. For the cited NMC442/graphite example, this was 80 °C overnight, about 14 hours, in a heated vacuum oven. Treat that as a study-specific example.
- Transfer without ambient exposure. Move the dried pouch directly into an argon-filled glove box when the procedure calls for inert handling.
- Add and record electrolyte. Measure the formulation and amount, then vacuum-seal the pouch consistently.
- Wet the stack. Apply the cell-specific wetting hold. Examples include 40 °C at 1.5 V for 24 hours in the NMC442/graphite work and an approximately 24-hour 1.5 V hold in the anode-free study.
- Run the defined formation program. Record all voltage, current, temperature and rest conditions.
- Degas if justified. If gas evolution is part of the cell history, open under the required inert atmosphere, release gas and reseal; otherwise omit this intervention.
- Install the cell in the holder. Document orientation, electrical connections and any pressure or mechanical constraint.
- Equilibrate thermally. Use the instrument- and study-specific temperature and rest period. One published example used 40.0000 ± 0.0001 °C and 24 hours at open circuit.
- Start the electrical program and heat-flow acquisition together. Preserve the timing, voltage/current schedule and data channels needed to relate electrochemical events to heat.
How to compare two preparation protocols
Two datasets should not be compared solely because both use “lithium-ion pouch cells.” Align the following variables first:

What to report so another laboratory can reproduce the run
A useful methods section should let a reader reconstruct the cell’s condition at the instant it entered the calorimeter. Include:
- Manufacturer or assembly route, pouch or wound format, chemistry and capacity.
- Dry, prefilled or previously cycled status on arrival.
- Opening procedure, oven temperature, vacuum condition, duration and transfer atmosphere.
- Glove-box atmosphere, electrolyte formulation, fill amount and sealing method.
- Wetting temperature, voltage and duration.
- Complete formation schedule, including rests and temperature.
- Whether and when the pouch was opened for degassing, plus resealing details.
- Calorimeter model and holder arrangement, mechanical constraint and electrical connections.
- Temperature setpoint, stability specification when relevant, equilibration time and open-circuit state.
- Automated battery-tester program and the synchronization of electrochemical and heat-flow data.
TA Instruments’ technical e-book describes a TAM calorimeter used in battery heat-flow experiments. It is useful as a documented instrument workflow, but it does not establish a universal preparation standard or identify the best calorimeter for every sample type.
Common preparation errors and corrective actions
Treating one paper’s numbers as a standard
Problem: Applying 80 °C drying, a 1.5 V hold or a 24-hour rest to every chemistry.
Correction: Carry the purpose and cell design alongside each condition. Validate the sequence for the specific pouch, electrolyte and holder.
Leaving the incoming state vague
Problem: Calling a cell “fresh” without saying whether it was dry, filled or previously formed.
Correction: Record the state at receipt and all handling before calorimetry.
Exposing dried cells to room air
Problem: Breaking the controlled transfer between oven and filling step.

What is established—and what is not
Published work demonstrates a repeatable pattern: control moisture when a cell is opened, use inert handling where required, standardize electrolyte filling and sealing, document wetting and formation, degas only when justified, and equilibrate the cell in the calorimeter before applying the electrical program.
No consensus procedure establishes one universal moisture threshold, drying schedule, formation sequence, degassing protocol or calorimeter model for all lithium-ion samples. The safest practice is therefore to reproduce the complete cell history and instrument setup, not just one headline temperature or hold time.
