What Psychedelics Do to the Brain: Neural Connectivity, Perception and the Limits of Current Research

Human brain-imaging studies show that classic psychedelics such as psilocybin, LSD and DMT are associated with measurable changes in brain activity and functional connectivity. The clearest current pattern is not a single “psychedelic network,” but altered communication between normally distinct systems: higher-order transmodal networks communicate more with visual and motor networks, while connectivity within some networks can fall. These effects vary by substance, dose, timing, participant and analysis method.

Those findings help explain why perception, attention, self-processing and meaning can feel different, but they do not provide a complete mechanism. Imaging usually measures correlations in groups during a limited time window; it cannot by itself prove that one network change causes a particular visual effect, predict an individual’s experience or show that the brain has been permanently “reset.”

What brain imaging can—and cannot—tell us

Most human psychedelic neuroscience uses functional magnetic resonance imaging (fMRI). Rather than watching neurons directly, fMRI tracks blood-oxygen-level-dependent (BOLD) changes, an indirect signal related to local neural activity. Researchers can analyze the signal in two broad ways:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Activation: whether a region’s BOLD signal rises or falls during a task or compared with a baseline.
  • Functional connectivity: whether signals in separated regions or networks fluctuate together over time. Connectivity is statistical coupling, not a physical wire and not proof that one area sends information to another.

Resting-state scans measure spontaneous activity while participants do not perform a structured task. Task fMRI asks participants to view images, listen to sounds or perform another activity. Acute scans occur during the drug’s active effects; follow-up scans occur later and answer a different question. Results from these designs should not be treated as interchangeable.

Imaging is therefore one window on a psychedelic state. Subjective reports, behavior, pharmacology and other measurements add different information. A connectivity map can show that two systems became more statistically coupled, but it cannot by itself identify the thoughts, emotions or visual content that accompanied that change.

The connectivity patterns reported so far

Default-mode connectivity is often reduced, but not universally

The default mode network (DMN) includes interconnected regions involved in self-related thought, autobiographical processing and internally directed attention. Reviews have frequently reported reduced connectivity within the DMN during psychedelic states. This result helped motivate the popular idea that psychedelics “quiet” the brain’s self system.

That shorthand is too simple. A reduction in within-DMN coupling is an average statistical finding, not evidence that the network stops working. It can coexist with stronger communication between the DMN and sensory or motor systems. Newer pooled analyses also find that within-network reductions are selective and vary across drugs and networks rather than appearing as one universal signature.

Between-network communication can increase

Several reviews describe increased connectivity between normally segregated systems, including sensory networks and thalamocortical circuits. The thalamus helps route and coordinate information between subcortical and cortical regions, so altered thalamic coupling is relevant to changes in sensory integration. However, “more connectivity” does not automatically mean more accurate perception or more information. It describes stronger statistical coordination under the specific scan and analysis conditions.

The 2026 international mega-analysis

The most recent large synthesis in this evidence base is a Nature Medicine study published online on 6 April 2026. It combined 11 independent resting-state fMRI datasets from five countries and three continents. The pooled sample included 273 participants and more than 550 connectomes, covering psilocybin, LSD, mescaline, DMT and ayahuasca.

Using a uniform preprocessing pipeline and a Bayesian hierarchical model, the researchers found a core pattern of increased connectivity between transmodal networks—default, frontoparietal and limbic systems—and unimodal networks, especially visual and somatomotor systems. They also observed altered coupling of the thalamus, caudate, putamen and cerebellum with sensorimotor networks.

Within-network connectivity reductions were weak to moderate overall, selective rather than global, and substantially variable across substances and networks. The authors explicitly concluded that “the neural mechanisms underlying psychedelic effects remain unclear.” The mega-analysis is a stronger current anchor than any single small experiment, but it does not eliminate uncertainty or establish a one-to-one map between a network result and a conscious experience.

What major reviews found

Source and year Evidence base and approach Main pattern reported Important qualification
2023 systematic review 91 full-text functional-imaging papers representing 51 unique samples Recurring effects along the sensorimotor–association cortical axis, particularly in psilocybin and LSD studies 57 studies (54%) did not meet the review’s contemporary standards for Type I error correction or motion-artifact control; designs, doses, timing and analyses were highly heterogeneous
2024 scoping review 24 articles selected from 492 initial records; healthy participants; ayahuasca, psilocybin, LSD and MDMA Decreased within-DMN connectivity and increased sensory and thalamocortical connectivity, with some associations to subjective reports The review called for direct clinical evidence before drawing conclusions about clinical outcomes
2024 meta-analysis Synthesis of phenomenology, neuroimaging and pharmacology studies using varied techniques Increased between-network connectivity alongside reduced within-network connectivity Phenomenology, connectivity and pharmacology profiles showed only weak correspondence; BOLD activation could not be validly pooled because procedures and data were too heterogeneous
2026 international mega-analysis 11 resting-state datasets, 273 participants and more than 550 connectomes; uniform preprocessing and Bayesian hierarchical modeling Increased transmodal–unimodal connectivity, including visual and somatomotor links; altered subcortical and cerebellar coupling Within-network decreases were selective and variable, and the underlying neural mechanisms remain unresolved

These are different kinds of evidence. A scoping review maps a field; a systematic review evaluates a defined literature; a meta-analysis combines results under statistical assumptions; and a mega-analysis can reprocess individual datasets more consistently. Their apparent differences are not necessarily contradictions. They may reflect different samples, compounds, scan times, preprocessing choices and definitions of connectivity.

Why perception changes during a psychedelic state

Sensory information may be integrated differently

Classic psychedelics act primarily through serotonin systems, especially 5-HT2A receptors, but the imaging evidence does not reduce the experience to one receptor or one circuit. Increased coupling between transmodal networks and visual or somatomotor networks provides a plausible systems-level context for unusual combinations of sensation, imagery, body awareness and attention. The data are consistent with altered communication between high-level expectations and incoming sensory signals.

That interpretation remains a model, not a proven causal chain. A visual distortion could reflect changes in retinal processing, cortical prediction, attention, memory, emotional salience or several processes at once. Group-level connectivity cannot determine which explanation applies to one person at one moment.

Self-related processing and meaning can shift

Changes involving DMN, frontoparietal and limbic networks are relevant to self-referential thought, cognitive control and emotional meaning. Reduced integrity within a network could accompany less rigidly maintained mental patterns, while increased cross-network coupling could allow normally separated associations to interact. This may help contextualize reports of altered identity, unusual significance or a less familiar boundary between inner thought and external perception.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

It would be an error to convert that interpretation into a promise of psychological benefit. Imaging associations do not show that psychedelics permanently improve personality, erase trauma or “rewire” the brain. Clinical outcomes require controlled trials with appropriate participants, preparation, supervision and follow-up.

Acute findings are not the same as later effects

A scan taken during intoxication addresses the acute brain state. A scan weeks or months later addresses persistence, adaptation or recovery. These time points should never be combined casually. A temporary connectivity change during a session does not prove a lasting biological change, and a later behavioral effect cannot automatically be attributed to an acute scan pattern.

What the evidence covers for different substances

Compound or preparation How it appears in the cited evidence How to interpret the imaging findings
Psilocybin Included in the 2023 systematic review, 2024 reviews and 2026 mega-analysis Among the most represented classic psychedelic compounds, but results still vary by dose, timing, task and analysis
LSD Included in the 2023 systematic review, 2024 reviews and 2026 mega-analysis Shows recurring sensorimotor–association effects in the systematic review; pooled patterns are not identical across studies
DMT Included in the 2026 mega-analysis Contributes to the pooled transmodal–unimodal findings, but the mega-analysis does not establish a unique DMT connectivity signature from the summary alone
Ayahuasca Included in the 2024 scoping review and 2026 mega-analysis A preparation containing DMT and other compounds; findings should not be assumed equivalent to isolated DMT
Mescaline Included in the 2026 mega-analysis Part of the pooled dataset, with substance-specific variation retained in the hierarchical analysis
MDMA Included in the 2024 scoping review Pharmacologically and experientially distinct from classic psychedelics; it should not be treated as interchangeable with psilocybin, LSD or DMT

The table describes study scope, not a ranking of substances or a guide to use. A preparation, dose and route can change pharmacokinetics and subjective effects, so results from one form cannot automatically be generalized to another.

How researchers compare studies responsibly

When reading a claim about “connectivity,” check four axes before comparing it with another result:

Free tools Windows power users keep installed

One-click scans. No signup required.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  1. Compound and preparation: identify whether the study used psilocybin, LSD, DMT, ayahuasca, mescaline or another substance, and whether it examined an isolated compound or a mixture.
  2. Measurement and analysis: distinguish resting-state fMRI, task fMRI, regional BOLD activation and functional-connectivity analysis. Different metrics answer different questions.
  3. Time point: separate scans during acute effects from scans after the experience.
  4. Network or outcome: note whether the result concerns the DMN, visual system, somatomotor system, thalamus, a whole-brain measure or a subjective rating.

Two papers can both report “increased connectivity” while measuring different links, at different times, under different preprocessing choices. The phrase alone is not enough to establish that they found the same thing.

Why the current evidence remains uncertain

Small and reused samples

The 2023 systematic review identified 91 papers but only 51 unique samples, meaning multiple publications sometimes drew on the same participants. A large paper count can therefore overstate the amount of independent evidence.

Motion and statistical-control problems

Head motion can create or exaggerate apparent connectivity changes. The systematic review judged that 57 studies, or 54% of those assessed, did not meet its contemporary standards for Type I error correction or motion-artifact control. This does not prove that every result from those studies is wrong; it means confidence should be lower and replication more important.

Different protocols produce different signals

Studies vary in dose, scanner timing, participant expectations, eyes-open or eyes-closed instructions, task demands, sample characteristics and preprocessing pipelines. Even the brain atlas used to define a network can change the measured connectivity. The 2024 meta-analysis found too much procedural and data heterogeneity to validly pool BOLD activation and required a specific re-parcellation approach for its connectivity synthesis.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Correlation is not mechanism

A connectivity change can correlate with a participant’s rating of visual intensity or ego dissolution without causing it. Both could arise from another unmeasured process, and the direction of influence may differ across people. Establishing mechanism requires converging evidence from pharmacology, physiology, behavior, longitudinal designs and carefully controlled experiments.

How to read headlines about psychedelic brains

  • Prefer “associated with altered connectivity” to “causes the brain to rewire.”
  • Ask whether the result came from healthy volunteers, a clinical sample or a mixed population.
  • Check whether the scan captured the acute drug state or a later follow-up.
  • Look for the actual network pair and metric rather than accepting “the brain became more connected” as a complete description.
  • Treat a pooled average as a population pattern, not a prediction of one person’s experience.
  • Separate a neural observation from a treatment claim; imaging alone cannot establish safety, effectiveness or a lasting therapeutic mechanism.

Current human imaging supports a picture of temporarily altered communication across brain systems, especially stronger links between higher-order transmodal networks and sensory or motor networks, alongside selective changes within networks. It does not yet explain consciousness under psychedelics in full. The field’s most useful next steps are larger preregistered studies, consistent motion and error controls, transparent data reuse, substance-specific analyses and designs that connect brain measures to carefully measured perception over time.
