Adding an active pharmaceutical ingredient (API) supplier improves resilience only when the new source is independently qualified, technically comparable, regulatorily usable and operationally reliable. A second supplier name is not automatically a second supply path: shared starting materials, sites, contractors, ports, utilities or testing laboratories can create one common point of failure. Qualify the actual manufacturing site and map those dependencies before counting the source toward continuity.
Start by defining what diversification must achieve
Set the decision objective before contacting candidates. Is the additional source intended to cover a capacity shortfall, reduce dependence on one geography, provide a registered alternate, support a launch market, or create recovery capacity after a disruption? The answer determines the evidence you need and the acceptable residual risk.

Write down the API, dosage form and finished product, destination markets, relevant marketing applications or filings, manufacturing site, process steps, grade and specification, intended annual volume, and the role the supplier will play in the supply plan. Record whether brokers, distributors, repackers, contract laboratories, storage providers or other outsourced operations are involved. Confirm which legal entity actually manufactures the API and which physical sites perform synthesis, extraction, fermentation or cell culture, recovery, purification, micronization, packaging, testing and release.

A risk-based qualification workflow
1. Qualify the specific manufacturing site, not just the corporate group
Supplier approval needs evidence that the identified manufacturer can repeatedly make material meeting the agreed specification. ICH Q7 states: “Supplier approval should include an evaluation that provides adequate evidence (e.g., past quality history) that the manufacturer can consistently provide material meeting specifications.” Review the manufacturer’s quality history for the actual site and process, not only certificates or a parent company’s reputation.
- Examine the scope and results of recent audits or equivalent evaluations, including open observations and remediation effectiveness.
- Assess process and facility capability against the proposed grade, batch size, containment, cleaning and storage requirements.
- Verify that the quality unit is independent enough to approve or reject materials and batches and has authority over deviations, investigations, changes and release.
- Sample batch records, laboratory data, deviations, out-of-specification investigations, complaints, recalls and change-control records.
- Check data-integrity controls, electronic-system governance, audit trails, record retention and traceability from starting material to dispatched lot.
- Identify critical starting materials and intermediates, their approved sources and the controls used when those sources change.
Q7 expects GMP controls to become more stringent as manufacture advances toward final processing, purification and packaging. A site that is acceptable for an early intermediate may not be suitable for the final API. Where the candidate uses a different route, impurity profile, specification, site or critical material, have technical and quality teams assess comparability and product impact before treating the material as interchangeable. Q7 establishes the need to meet intended quality and appropriate GMP; the comparability conclusion must come from the API- and process-specific data.
2. Put incoming-material controls around the new source
Supplier approval belongs inside the site’s materials-control system. Q7 calls for written procedures covering receipt, identification, quarantine, storage, handling, sampling, testing, approval or rejection and purchasing to agreed specifications from suppliers approved by the quality unit. Translate those requirements into the receiving site’s procedures before the first commercial shipment.

Define how containers and seals are checked, how labels and lot numbers are reconciled, where material is quarantined, who can release it, how samples are taken without compromising the lot, and how rejected or returned material is segregated. Require notification before a source, manufacturing location, specification, test method, packaging configuration or transport condition changes. The approved-source list should identify each API and each permitted manufacturing site, intermediary and repacker.

3. Build an analytical qualification plan before routine supply
Agree the specification, analytical methods, acceptance limits, sampling plan, reference standards, method-transfer or verification work, impurity characterization and treatment of discrepant results before purchase orders are released. Include responsibilities for investigations when an incoming result conflicts with the supplier’s certificate of analysis (COA).
FDA’s component-control Q&A, dated June 9, 2015, says that a finished-drug manufacturer relying on an API supplier’s analysis must perform at least one specific identity test and validate the reliability of the supplier’s analyses at appropriate intervals. The agency explains: “Therefore, if the finished drug product manufacturer accepts the test results from an API supplier’s COA rather than performing the tests itself (other than for identity, which the manufacturer is required to perform), the manufacturer must validate the API supplier’s reliability.” See the FDA Q&A for the agency’s wording and context.
ICH Q7A recommends complete analysis of at least three batches before reducing in-house testing, followed by complete analysis at appropriate intervals compared with COAs and regular verification of COA reliability. Treat the three-batch point as a Q7 recommendation requiring a documented, justified plan; it is not a universal waiver from applicable CGMP requirements or from product-specific controls. Define the interval, attributes, laboratories, acceptance criteria and actions for a failed verification in your procedures.

4. Establish a written quality agreement with usable escalation rules
A quality agreement should make operational responsibility unambiguous. FDA’s November 2016 quality-agreement guidance recommends documenting relevant CGMP roles and activities between owners and contract facilities. Address, as applicable:

- subcontracting, use of laboratories, brokers, repackers and storage providers;

Keep commercial terms separate or severable from quality terms so that a pricing or volume dispute cannot obscure a safety-critical obligation. The agreement allocates activities; it does not transfer legal accountability. FDA states verbatim: “Quality agreements cannot be used to delegate statutory or regulatory responsibilities to comply with CGMP.”
5. Monitor performance after approval
Approval is a starting decision, not a permanent status. Establish a supplier-specific review cadence and use a documented scorecard. Useful indicators include rejected or delayed lots, deviations and investigations, audit findings and overdue remediation, incomplete or late documentation, change notices, complaint or recall signals, on-time delivery, lead-time variance, allocation behavior and actual supply interruptions.
Set escalation thresholds and requalification triggers that fit the API’s criticality. For example, a critical impurity excursion, unexplained data-integrity concern, unapproved source change or repeated late change notification may require immediate quality-unit review, intensified testing or suspension. The exact thresholds are company decisions; do not present an invented percentage or score as an FDA requirement.

6. Test whether the source reduces correlated supply risk
Draw a dependency map for both the incumbent and proposed source. Include API starting materials and key intermediates, manufacturing and testing sites, ownership and contract relationships, distribution and repackaging steps, warehouses, transport lanes, ports, utilities and other dependencies that could fail together. Ask whether both suppliers rely on the same intermediate plant, solvent, catalyst, laboratory, shipping corridor, energy network or allocation decision.
Model realistic disruption scenarios rather than assuming that geographic distance equals independence. Consider regional concentration, capacity and allocation behavior, minimum order quantities, lead times, safety stock, recovery time, qualification time for substitutes and the practical ability to shift production. Compare the consequences of a quality shutdown, natural disaster, cyber incident, transport closure, utility failure, geopolitical restriction and loss of a critical upstream material. Record which controls reduce likelihood, which reduce impact and which residual risks require inventory or a third source.
FDA’s May 2022 Risk Management Plans to Mitigate Potential Drug Shortages document can support this planning. It is explicitly marked “Draft” and “Not for implementation,” so use it as a planning reference, not binding regulation or final guidance. The draft says, “Based on recent publications and reports, the majority of drug shortages are associated with quality issues.” It supplies no percentage on the cited page; do not turn that statement into a numeric statistic.
7. Confirm the regulatory path for the actual product and market
Before placing the new API into commercial manufacture, regulatory affairs must assess the product’s actual application, registered API source and site, approved process and specifications, proposed change and destination jurisdictions. Depending on those facts, the change may require a submission, prior approval, notification, other authority interaction or no filing action. There is no universal filing category or timeline that can be inferred from the number of suppliers.
Best Value
Q7 is a GMP document, not a definition of registration or filing requirements; it also says commitments in registration or filing documents should be met. Therefore, retrieve the current application and commitments, compare the candidate’s process and site details with the registered information, and document the market-by-market conclusion. For products outside the United States, verify the applicable regulator and regional rules rather than assuming FDA guidance controls the filing decision.

Once candidates pass minimum quality and regulatory gates, compare them on the same evidence set. Adjust weights to product criticality and the company’s risk appetite; the sources establish expectations for quality, suitability, oversight and supply risk, but do not prescribe a universal scoring formula.
| Comparison axis | Questions to answer | Evidence to retain |
|---|---|---|
| Quality history and GMP capability | Can the named site consistently meet specifications, and are investigations and data controls effective? | Batch and laboratory records, audit reports, deviations, OOS investigations, CAPA and inspection history |
| Technical and analytical fit | Are route, impurity profile, particle properties, methods, packaging and specifications comparable? | Process description, impurity assessment, method validation or transfer data, qualification batches |
| Regulatory readiness | What change is needed in each application and market, and what commitments constrain implementation? | Current filings, approved source/site details, regulatory assessment and authority correspondence |
| Upstream and geographic concentration | Which starting materials, intermediates, sites, utilities and lanes are shared with the incumbent? | Dependency map, ownership and subcontractor disclosures, continuity scenarios |
| Capacity and allocation risk | Can the source supply the required volume during demand spikes or a competitor’s disruption? | Capacity statement, lead times, allocation policy, minimum order quantities and recovery assumptions |
| Logistics and traceability | Can every lot be traced through brokers, repackers, warehouses and transport, with controlled conditions? | Approved supply-chain diagram, shipping records, temperature or handling controls and chain-of-custody data |
| Oversight workload and transparency | How quickly and completely will the supplier disclose changes, incidents and subcontracting? | Quality agreement, notification commitments, audit access and performance-review records |
| Total landed cost | What is the fully loaded cost after testing, audits, inventory, validation, freight, duties and regulatory work? | Documented cost model with assumptions and sensitivity to volume, delay and failure |
Gate the decision before commercial use
- Document the scope and risk statement. Identify the product, markets, API, sites, process, intended role and the disruption the second source is meant to reduce.
- Collect and verify evidence. Obtain site information, quality history, process and analytical data, upstream disclosures, capacity information and regulatory records. Resolve discrepancies rather than scoring around them.
- Run technical, quality and regulatory assessments in parallel. Do not wait until a purchase order is imminent to discover that impurity differences, method gaps or filing commitments prevent use.
- Complete qualification work. Perform audits or equivalent evaluations, method transfer or verification, identity testing, qualification batches and any product-specific comparability work required by the risk assessment.
- Approve the operating model. Execute the quality agreement, update approved-source lists and procedures, set sampling and COA-verification plans, and train receiving, laboratory, procurement and quality personnel.
- Record the residual-risk decision. State which dependencies remain shared, what inventory or recovery controls address them, which conditions trigger requalification and who owns each action.
- Review after launch. Trend quality and service indicators, verify the source remains the agreed site and supply chain, and reassess the diversification benefit when processes, ownership, markets or upstream sources change.
Common diversification mistakes and their fixes
- Counting supplier names instead of independent paths: map common starting materials, intermediates, laboratories, logistics and utilities before claiming redundancy.
- Accepting a COA without a verification strategy: perform the required identity test and validate supplier analytical reliability at justified intervals.
- Using a quality agreement as a liability transfer: assign activities clearly while retaining each party’s statutory and regulatory CGMP responsibilities.
- Assuming a different route is automatically equivalent: compare impurity profiles, specifications, physical properties and finished-product impact with actual data.
- Skipping ongoing oversight after approval: trend deviations, changes, complaints, documentation and delivery performance and define escalation triggers.
- Assuming supplier count determines filing requirements: review the product’s application, registered commitments and destination-country rules for the specific change.
- Using arbitrary score weights: document why quality, regulatory readiness, capacity, continuity and cost receive their chosen weights for this product.
What a defensible approval file contains
A complete decision record links the business objective to evidence and residual risk. Retain the scope definition, site and supply-chain map, audit or evaluation reports, quality history, process and impurity comparability assessment, analytical qualification and COA-verification plan, quality agreement, regulatory assessment, capacity and logistics evidence, comparison scoring with documented weights, approval decision, conditions, owners and review date. That file lets quality, regulatory, procurement and operations reach the same conclusion and shows why the source is—and remains—an acceptable part of the supply strategy.
