What Microgravity Does to Plant Growth and Gene Expression

Microgravity changes both how plants grow and how their cells regulate genes. Without a strong gravity vector, roots and shoots lose the directional cue that normally drives gravitropism, so their orientation and growth habits can change. Spaceflight also remodels gene-expression programs, RNA processing, protein abundance and protein modification. Those molecular shifts can affect photosynthesis, metabolism, cell-wall formation and development.

There is no single percentage for how much a plant grows in microgravity. Results depend on species, tissue, genotype, gravity level, light, exposure duration, growth hardware and other spaceflight stresses. Most of the detailed evidence comes from Arabidopsis thaliana, a model plant rather than a food crop.

What changes first: direction, not necessarily overall survival

On Earth, gravity helps plants determine which way to grow. Roots normally display positive gravitropism, growing toward the gravity vector, while shoots generally show negative gravitropism, growing away from it. Moisture and light provide additional directional information. In microgravity, that strong gravity cue is largely absent. Roots and shoots can therefore adopt less predictable orientations, altered branching patterns and different growth habits.

NASA describes microgravity-grown plants as generally normal in appearance while still showing distinctly different growth behavior. A seedling may remain viable and produce ordinary-looking tissues, yet its roots can wander rather than settle into a consistent downward direction. The visible result is not simply “plants stop growing”; it is growth with a changed set of spatial cues.

The TAGES-ISA investigation uses Arabidopsis seedlings to examine root structure, growth and cell-wall remodeling. Its focus matters because a root that grows in an unexpected direction may also experience different mechanical resistance, water distribution and nutrient access. Cell walls must accommodate elongation and bending, so changes in wall construction can connect the plant’s environment to its final shape.

Why roots are especially informative

Roots integrate gravity sensing with water, nutrients, touch and chemical signals. In microgravity, researchers can ask which parts of root development depend specifically on gravity and which are controlled by other cues. Root tissues may also respond differently from shoots, meaning a whole-plant average can hide important tissue-specific effects.

For space agriculture, orientation is practical as well as biological. A crop with roots that fail to explore its growth medium evenly could have trouble obtaining water or nutrients. Researchers therefore study root architecture, not just plant height or biomass.

Does microgravity change plant DNA?

The strongest evidence concerns gene regulation, not a wholesale rewrite of the DNA sequence. Microgravity can change which genes are transcribed, how RNA is processed, which proteins are produced and how those proteins are modified. In other words, the same genome can be used differently under a different physical environment.

NASA summarizes the finding this way: Previous research shows that microgravity conditions during spaceflight affects which genes turn on or off, which proteins are present and in what amounts, and the modifications made to those proteins. This statement appears on NASA’s APEX-07 investigation page, updated May 21, 2023.

Gene expression is a sequence of regulated steps. DNA is transcribed into RNA; RNA can be edited, transported and translated; proteins can then be modified, moved or degraded. A change at any of those stages can alter physiology without changing the underlying genetic sequence. Some responses may also reflect stress from launch, radiation, confinement, airflow or nutrient delivery rather than gravity alone, which is why well-controlled experiments are essential.

RNA regulation adds another layer

Measuring only messenger-RNA abundance does not reveal the entire response. The APEX-07 investigation examines several layers of RNA regulation in roots and shoots:

  • Transcriptome: the collection and abundance of RNA transcripts present in a tissue.
  • Translatome: the subset of RNA molecules being actively used to make proteins.
  • Alternative splicing: different ways of processing one gene’s initial RNA transcript, potentially producing distinct protein versions.
  • Small RNAs: short regulatory molecules that can influence transcript stability and translation.

This design targets post-transcriptional regulation: control that occurs after DNA has been transcribed. Two plants could have similar levels of a transcript but translate it at different rates, splice it differently or suppress it with small RNAs. Looking at transcriptome, translatome and small-RNA landscapes together gives a more realistic picture of how microgravity changes gene function.

Why roots and shoots are measured separately

Roots and shoots perform different jobs and encounter different local conditions. A gravity-sensitive signaling pathway may be prominent in a root tip but not in a leaf. Photosynthetic tissues may respond to light and carbon balance, while roots respond more strongly to water movement and mechanical cues. APEX-07 therefore analyzes both tissues instead of treating the plant as one uniform sample.

From individual genes to whole-plant physiology

Plant Habitat-01 compares Arabidopsis grown at normal Earth gravity (1g) and in microgravity using a multi-omics strategy. The investigation combines four kinds of evidence:

Measurement layer What it can reveal
Phenomics Observable traits such as growth, form and development
Metabolomics Changes in small molecules involved in energy use and biochemical pathways
Transcriptomics Which RNA transcripts are more or less abundant
Proteomics Which proteins are present and in what amounts

NASA identifies photosynthesis, cell-wall formation, growth and development, and gene function as key areas of interest. This integrated approach helps connect a molecular change to a physical outcome. For example, altered expression of cell-wall genes might be evaluated alongside wall-related proteins and the plant’s measured shape. A transcript difference that produces no downstream protein or phenotype may be less consequential than a coordinated change across several layers.

What partial gravity experiments reveal

Microgravity is not the only condition relevant to exploration. The Moon and Mars provide partial gravity, so researchers need to know whether plant responses change gradually with gravity level or switch at a threshold.

A NASA Open Data dataset grew Arabidopsis for six days at microgravity, several intermediate partial-gravity levels and 1g. Blue light was applied during the final 48 hours. Researchers extracted RNA from 20 samples and analyzed differential expression with the HISAT2-StringTie-DESeq pipeline, followed by gene-network and Gene Ontology analyses.

Experimental feature Documented value
Plant Arabidopsis
Growth duration Six days
Gravity conditions Microgravity, several intermediate partial-gravity levels and 1g
Light treatment Blue light during the final 48 hours
RNA material 20 samples
Analysis HISAT2-StringTie-DESeq differential-expression pipeline, gene networks and Gene Ontology enrichment

Because gravity levels were separated experimentally, this dataset can test whether expression patterns track the size of the gravity vector. The blue-light treatment is also important: light is itself a major developmental signal, so the result reflects an interaction between gravity and illumination rather than gravity in isolation.

A root-orientation result after a short exposure should not be treated as equivalent to a proteomic result from a longer experiment. Likewise, a difference in a shoot transcript does not automatically mean the plant’s roots or final yield changed.

Are plants stressed in microgravity?

Often, microgravity activates responses associated with altered mechanical and environmental conditions, but “stress” is not one single molecular signature. Plants may adjust cell-wall remodeling, metabolism, photosynthesis and developmental pathways while remaining alive and producing new tissue. Some changes may be adaptive rather than damaging.

Spaceflight also introduces non-gravity factors. Launch vibration, radiation, confined growth volume, airflow, temperature, water delivery and nutrient distribution can all influence plants. NASA’s controlled comparisons and multi-omics measurements are intended to separate these influences as far as practical. A result observed in orbit should therefore be interpreted alongside its 1g control and hardware conditions.

What this means for growing food and producing oxygen in space

Plants are attractive life-support partners because they can provide edible biomass and contribute oxygen production. NASA connects microgravity plant research directly to those goals for long-duration missions. However, a crop system must do more than keep a few seedlings alive.

Root-zone management

Roots need reliable access to water and nutrients even when gravity does not pull liquid through a medium in the usual way. Irregular root orientation can make uniform delivery harder, so future systems must control moisture, aeration and nutrient flow mechanically.

Predictable development

Food production depends on repeatable timing, architecture and harvestable tissue. Gene-expression changes in photosynthesis, metabolism or development could alter productivity, but the direction and size of those effects will vary by species and environment. Results from Arabidopsis provide mechanisms to investigate; they do not establish a universal response for wheat, lettuce, tomatoes or other crops.

Designing for partial gravity

Moon- or Mars-based agriculture may operate at a partial gravity rather than true microgravity. The partial-gravity dataset shows why those conditions should be tested directly instead of inferred from either Earth or orbit. Intermediate gravity could produce a distinct expression program rather than a simple halfway point between 1g and microgravity.

What the evidence does not establish

  • There is no single cross-study percentage for growth reduction or increase.
  • Microgravity does not automatically mean that plant DNA sequence is rewritten.
  • A gene-expression difference alone does not prove a yield change.
  • Findings from Arabidopsis cannot be generalized quantitatively to every crop.
  • Spaceflight results cannot be attributed to gravity without considering light, radiation, airflow, nutrient delivery, confinement and hardware controls.

The most defensible conclusion is mechanistic: removing the normal gravity cue changes plant orientation and prompts coordinated regulation across RNA, proteins and physiology. The exact phenotype depends on the experimental context.

How researchers build a convincing experiment

  1. Define the gravity comparison. Use microgravity, specified partial-gravity levels and a matched 1g control when the question concerns gravity sensitivity.
  2. Separate tissues. Collect roots and shoots independently so tissue-specific responses are not averaged away.
  3. Record environmental conditions. Document light spectrum and timing, temperature, airflow, water and nutrient delivery, hardware and exposure duration.
  4. Measure more than one layer. Pair visible traits with transcriptomic, translatomic, proteomic or metabolomic data.
  5. Test biological meaning. Use gene networks and Gene Ontology analyses, as in the 20-sample partial-gravity dataset, to determine whether individual changes form coherent pathways.
  6. Validate the phenotype. Link molecular patterns to root architecture, cell-wall properties, photosynthesis, development or another measured plant function.

This workflow prevents a common mistake: treating every altered transcript as proof that microgravity harmed the plant. The meaningful question is whether molecular, cellular and whole-plant measurements point to the same biological process.

Bottom line for readers

Microgravity removes the directional gravity signal that guides roots and shoots, so plants can grow with altered orientation and architecture. At the same time, it remodels gene regulation at transcriptional and post-transcriptional levels, changes protein abundance and modification, and can affect photosynthesis, metabolism, cell walls and development. NASA’s APEX-07, Plant Habitat-01, TAGES-ISA and partial-gravity studies show why the response must be described by tissue, gravity level, light, duration and measurement method. The findings support future space food and oxygen systems, but they do not justify one universal growth percentage or a claim that plant DNA is simply rewritten in orbit.
