Single-use and reusable tangential flow filtration (TFF) systems use the same filtration principle but have different product-contact lifecycles. In both, feed flows parallel to a membrane; part of the fluid crosses the membrane as permeate while the retained product stays in the process stream. A single-use system discards designated wetted components after a run. A reusable system keeps those components for additional cycles and requires cleaning, validation, and controlled storage between uses.
Neither approach is universally better. The right choice depends on membrane and product compatibility, scale, campaign frequency, realistic reuse cycles, facility utilities, changeover work, qualification requirements, and total cost at your site.

What TFF does before you compare system lifecycles
TFF is a pressure-driven membrane operation used for concentration, buffer exchange (diafiltration), clarification and other downstream steps in bioprocessing. Unlike normal-flow filtration, in which feed is pushed toward and through a membrane, TFF keeps the feed moving across the membrane surface. That crossflow helps limit buildup at the membrane and allows a retentate loop to circulate material while permeate leaves the system.

The filtration method is the same whether the hardware is single-use or reusable. The lifecycle designation describes what happens to the membrane device and other product-contact surfaces after the batch. A “single-use TFF system” may mean only a disposable capsule, or it may include a broader disposable flow path with tubing, a reservoir, pump-contact parts, sensors and connectors. A meaningful comparison must identify exactly which wetted surfaces are disposable.

Single-use versus reusable TFF at a glance
| Decision axis | Single-use TFF | Reusable TFF |
|---|---|---|
| Product-contact components | Designated components are discarded after the run; coverage varies by configuration. | Components are retained for additional cycles. |
| Preparation and changeover | Can remove or reduce cleaning, cleaning validation and storage steps. | Requires a validated cleaning and storage approach between uses. |
| Process performance | Can perform comparably in a particular validated application; this does not prove universal equivalence. | Established formats may fit existing processes and equipment. |
| Economics | Recurring disposable purchases and waste; often attractive when campaigns are short, changeovers frequent or scale is smaller. | Equipment and membrane costs are spread over cycles, while cleaning, labor, utilities, validation and storage add lifecycle costs. |
| Utilities and materials | May reduce process cleaning fluids and associated preparation. | Cleaning and flushing consume water and cleaning agents, while reusable parts avoid some disposable waste. |
| Scale and facility | Can support flexible or multiproduct operation, especially when the complete product-contact path is disposable. | Can align with established infrastructure and repeated, high-throughput campaigns. |
| Product safety and qualification | Requires assessment of materials and extractables and leachables for selected disposable wetted parts. | Requires cleaning validation and control of carryover between uses. |
How a single-use TFF system works in practice
Before processing, operators install a qualified disposable capsule or flow path, connect it to the pump and sensors, and perform the specified integrity, pressure or leak checks. Depending on the design, the disposable assembly can include the membrane device, tubing, bag or reservoir, connectors and some sensors. The exact coverage is a purchasing and qualification specification, not a generic property of every “single-use” system.
After the batch, product-contact parts are removed and discarded according to the facility’s waste procedures. Eliminating the post-run cleaning cycle can shorten turnaround and reduce the need to store a used membrane assembly. It does not eliminate all work: operators still need setup, line clearance, disposal, documentation, and checks that the disposable materials are suitable for the molecule and process.
Disposable wetted materials must be qualified for compatibility, including extractables and leachables information, pressure and temperature limits, and any sterilization or bioburden controls required by the process. A disposable path can also introduce practical constraints such as hold-up volume, connector configuration, tubing flexibility, sensor integration and supply continuity.

How reusable TFF systems work in practice
A reusable cassette or other reusable product-contact assembly remains in service after a batch. Operators must recover product, flush the system, execute a validated cleaning procedure, verify that residues and cleaning agents are removed, and store the assembly under controlled conditions until the next use. The site must define acceptance criteria for cleaning, inspection, integrity and, where applicable, microbial control.
Reusable operation creates a carryover question that single-use operation largely avoids: material from one product or batch must not contaminate the next. Cleaning validation therefore has to reflect the hardest-to-clean product, the worst-case soil, the equipment geometry and the campaign strategy. Storage time and conditions also need controls so that a cleaned assembly does not become a new contamination source.
Reuse is not unlimited. Membrane performance, mechanical condition, cleaning history and validated maximum cycles determine when a cassette is retired. The economically useful cycle count is process-specific rather than a universal rating.

Does single-use perform as well as reusable TFF?
Performance has to be demonstrated for the molecule, membrane, device geometry and operating scale being considered. Compare flux, yield, concentration or diafiltration endpoint, residual clearance, product quality and hold-up volume under matched conditions. A change in membrane area, channel design, pump control or operating pressure can affect results independently of whether the path is disposable.

A peer-reviewed study in the Journal of Chemical Technology & Biotechnology (2022) compared a single-use capsule with reusable cassettes for purification of activated polysaccharides used in conjugate vaccines. In that tested process, the systems showed similar TFF process performance and similar post-purification product attributes. The capsule also required a smaller flush and did not need a compression holder or pre-sanitization. Those are findings for the devices and process studied; they do not establish interchangeability for every molecule, membrane, scale or operating recipe.
Supplier examples illustrate why local testing matters. Merck/Sigma-Aldrich describes one application in which a single-use process took less than 2.5 hours versus 7 hours for a multi-use process, and another example using 20 L/m² of fluid for a capsule versus 130 L/m² for multi-use cassettes. These are manufacturer examples, not industry averages or guarantees for your process.

Cleaning, validation and changeover implications
Single-use changeover
- Remove the used product-contact path and document disposal.
- Install and verify the next qualified path.
- Perform required leak, pressure, integrity and sensor checks.
- Review material certificates, lot records and assembly identification.
Because the wetted path is discarded, many cleaning and cleaning-validation activities can be reduced. The actual benefit depends on whether the facility still reuses a pump head, housing, sensor, reservoir or other wetted component.
Reusable changeover
- Flush product residues using the validated sequence and volumes.
- Apply the approved cleaning chemistry, time, temperature and flow conditions.
- Rinse until chemical and residue acceptance criteria are met.
- Inspect and test the assembly, record its cycle history, then store it under controlled conditions.
Reusable systems can fit a facility that already has validated cleaning skids, water capacity, chemical handling and trained personnel. A site without that infrastructure may find that the apparent hardware savings are offset by qualification and turnaround work.
How the economics really differ
Single-use economics are dominated by recurring consumables: capsules or cassettes, tubing, bags, connectors, sensors and disposal. The model can be favorable when batch sizes are modest, products change often, campaigns are short, or fast changeovers have high value. It can be less favorable at large scale or with long campaigns that would reuse a robust assembly many times.

Reusable economics spread the cost of cassettes and associated hardware over multiple cycles. Against that, include cleaning labor, water, cleaning agents, utilities, cleaning-validation studies, storage space, inspection, testing, downtime and eventual replacement. Include the cost of failed cleaning runs, discarded assemblies that do not reach their expected cycle count, and line capacity consumed by cleaning.
A historical vendor-authored TangenX Technology Corp. model (2008) placed a break-even point at approximately 20 reuse cycles for a 10 m² operation. That figure depends on the model’s assumptions and is not a current quote or a universal threshold. Use it only as an example of how cycle count can change the result. Build a site-specific total-cost model instead of applying a published break-even number.

Single-use paths can reduce process-cleaning water and chemical demand, but they create recurring solid waste and require manufacture and shipment of disposable materials. Reusable paths avoid some disposable waste but consume water, chemicals and energy for cleaning, flushing, drying or storage. The balance depends on local utility sources, waste treatment, transport, assembly materials and reuse count.

Product safety and qualification questions
For a single-use path
- Are all wetted materials compatible with the product, buffers, solvents, temperature and pressure?
- What extractables and leachables data apply to the exact material, lot and sterilization condition?
- Does the assembly meet required bioburden, endotoxin or sterility controls?
- Are connectors, tubing, sensors and bags included in the supplier’s qualification package?
- Can the supplier maintain continuity of supply and consistent assembly quality?
For a reusable path
- What cleaning agents, temperatures, flow rates and contact times are validated?
- How are residues, cleaning chemicals and microbial risks tested?
- What is the validated maximum number of cycles, and how is cycle history recorded?
- How are carryover limits set for the next product or batch?
- What inspection, integrity and storage controls apply between uses?
When each approach tends to fit
Single-use is often a strong candidate when
- The facility runs many products or changes campaigns frequently.
- Fast turnaround is more valuable than minimizing per-batch consumables.
- The operation is small or developmental and does not have extensive cleaning infrastructure.
- A complete disposable flow path can remove difficult-to-clean wetted surfaces.
- Supplier data and process studies support material compatibility and performance at the intended scale.
Reusable is often a strong candidate when
- The same product runs repeatedly in long campaigns.
- The facility already has validated cleaning, water, chemical and storage systems.
- Expected reuse cycles are high enough to spread hardware and membrane costs.
- Existing automation, piping and equipment are designed around reusable cassettes.
- Waste reduction or supply-chain considerations favor retaining the assembly, subject to validated controls.
These are screening conditions, not automatic decisions. A multiproduct facility may still use reusable hardware in one unit operation and a disposable path in another, depending on contamination risk, scale and changeover requirements.
A practical selection method
- Define the disposable boundary. List every product-contact surface: membrane device, tubing, reservoir, pump head, sensors, connectors and housings. Record which items are discarded and which remain in service.
- Set process requirements. Document molecule and membrane compatibility, operating pressure and temperature, target flux, yield, residual clearance, product-quality limits, hold-up volume and required batch size.
- Map the campaign. Estimate annual runs, campaign length, product changes, expected reuse cycles and the time available for setup, cleaning and release.
- Measure the facility workflow. Include setup and teardown labor, line clearance, cleaning and validation, storage, water, chemicals, utilities, waste handling and equipment capacity.
- Compare evidence at relevant scale. Request supplier data for extractables and leachables, membrane performance, pressure limits, integrity testing, cleaning validation and scale-up. Run a side-by-side qualification when a change could affect yield or product quality.
- Build a total-cost and risk model. Include purchase price, consumables, labor, utilities, validation, waste, downtime, failed runs, replacement and supply risk. Treat published examples as assumptions to test, not as universal prices or cycle counts.
- Choose and re-evaluate. Approve the configuration that meets quality and capacity requirements at acceptable lifecycle cost. Revisit the decision when batch size, campaign frequency, product mix, facility utilities or supplier conditions change.
Common mistakes to avoid
- Comparing only the membrane price. Disposable tubing, bags, connectors, sensors and disposal can dominate single-use cost; cleaning, validation and utilities can dominate reusable cost.
- Calling a capsule and a complete system equivalent. The amount of disposable flow path determines how much cleaning and carryover work is actually removed.
- Generalizing one successful study. Similar performance in a conjugate-vaccine purification process does not prove the same result for another molecule or scale.
- Using a vendor example as an industry benchmark. The 2.5-hour versus 7-hour and 20 L/m² versus 130 L/m² figures are application-specific manufacturer examples.
- Assuming “reusable” means unlimited reuse. Cleaning validation, membrane condition and a defined maximum cycle count are essential.
- Ignoring supply and waste logistics. Confirm disposable availability, assembly lead times, storage, disposal routes and the site’s ability to handle cleaning chemicals before approval.
Bottom line
Single-use TFF trades recurring disposable materials for simpler changeovers and less cleaning work. Reusable TFF trades cleaning, validation and storage effort for the opportunity to spread component costs over multiple cycles and use established infrastructure. Select between them by comparing the complete product-contact path, process performance, campaign pattern, facility capabilities and total lifecycle cost for the actual site—not by treating “single-use” or “reusable” as a universal performance or sustainability verdict.
