What Is Regioselective Hydroformylation and How Does It Work?

Regioselective hydroformylation is the controlled addition of carbon monoxide (CO) and hydrogen (H2) to an alkene so that the reaction favors one of two possible aldehyde structures. In the usual case, a terminal alkene can produce either a linear (normal, or n) aldehyde or a branched (iso, or i) aldehyde. A catalyst determines which carbon of the original C=C bond receives the formyl group and therefore controls the linear-to-branched (L:B, also written n:i) ratio.

The reaction is widely used to make aldehydes for plasticizers, detergents, fine chemicals, pharmaceuticals and agrochemicals. Its selectivity is not a fixed property of hydroformylation: metal, ligand, substrate, solvent, gas pressure, temperature, catalyst geometry and even confinement can change the product distribution.

What hydroformylation does

Hydroformylation, also called the oxo process, converts an alkene, carbon monoxide and hydrogen into an aldehyde. Overall, the alkene gains a formyl group (–CHO) and a hydrogen atom across its carbon–carbon double bond. The carbon in the new aldehyde group comes from carbon monoxide, while hydrogen is supplied by H2.

For an unsymmetrical alkene there are two constitutional possibilities. Consider a terminal alkene written as R–CH=CH2:

The products have the same atoms but different connectivity, so they are regioisomers. “Regioselective” describes the preference for one connectivity over the other; it does not by itself describe the three-dimensional arrangement of atoms.

Hydroformylation is often described as an “ideal atom economic C/C-bond forming reaction,” a phrase used by Bernhard Breit in a 2003 Accounts of Chemical Research article. In practice, however, high atom economy does not guarantee a simple process: unreacted alkene, branched and linear aldehydes, hydrogenation products and catalyst residues may still have to be separated.

Why regioselectivity matters

The aldehyde is frequently an intermediate rather than the final product. Its carbonyl group can be hydrogenated to an alcohol, oxidized to an acid, converted into an amine or used in condensation and coupling chemistry. Choosing the linear or branched isomer at the hydroformylation stage determines the carbon skeleton available for every downstream step.

Industrial plants often seek a high proportion of linear aldehyde because normal aldehydes can be valuable precursors to linear alcohols and other bulk chemicals. Synthetic chemists may instead want the branched isomer, a stereodefined product or an aldehyde at a particular position in a complex molecule. A catalyst that is excellent for one target can therefore be unsuitable for another.

Selectivity also affects economics and environmental performance. A larger fraction of the desired aldehyde reduces distillation or extraction duty, minimizes waste from unwanted isomers and simplifies downstream purification. The relevant performance is multidimensional: alkene conversion, aldehyde selectivity, L:B ratio, catalyst productivity, operating severity, lifetime and metal recovery all matter.

How the catalytic cycle works

The most commonly discussed model is a homogeneous rhodium(I) cycle. The exact intermediates can vary with ligand, solvent and gas composition, but the sequence below explains how regioisomers arise.

  1. Formation of a rhodium hydride. Under syngas conditions, a coordinatively unsaturated Rh(I) hydride is generated. The metal has an open site where the alkene can bind.
  2. Alkene coordination and insertion. The C=C bond coordinates to rhodium and then inserts into the Rh–H bond. Insertion can occur in either orientation. One orientation gives a linear Rh–alkyl intermediate; the other gives a branched Rh–alkyl intermediate. This is the principal branching point for regioselectivity.
  3. Carbon monoxide insertion. CO coordinates to the Rh–alkyl species and inserts into the metal–carbon bond, producing a rhodium acyl complex. The alkyl group is thereby converted into the carbon chain attached to the future aldehyde carbonyl.
  4. Hydrogen activation. H2 undergoes oxidative addition at the acyl complex, placing hydrogen atoms on the metal.
  5. Aldehyde release and catalyst regeneration. Reductive elimination (often described as hydrogenolysis of the acyl) releases the aldehyde and regenerates a rhodium hydride capable of entering another cycle.

The first insertion step creates the two regioisomeric pathways, but the product ratio is not determined there alone. Linear and branched metal–alkyl or acyl intermediates can interconvert, and their rates of CO insertion, hydrogenolysis and aldehyde release can differ. Consequently, a catalyst can alter the observed L:B ratio through kinetic preference, intermediate equilibration, or both.

What controls linear versus branched products

Metal identity and catalyst phase

Rhodium catalysts usually provide high activity under relatively mild conditions, while cobalt catalysts have long been important in large-scale oxo chemistry and can tolerate different operating regimes. The comparison is not simply “rhodium is better”: metal price, pressure, temperature, ligand stability, catalyst lifetime, product distribution and recovery method determine the practical choice.

In a homogeneous system, the metal complex and substrate share one liquid phase, which gives precise ligand control but can make metal separation difficult. Supported or heterogeneous catalysts simplify recovery and may enable continuous processing, yet the support can create mass-transfer limitations, nonuniform active sites or metal leaching. A 2026 review in ACS Catalysis (published online 20 April 2026; issue dated 1 May 2026) identifies geometric, electronic, synergistic, defect and confinement effects as key design levers. It also notes that controlling regioselectivity within heterogeneous hydroformylation catalysts remains a persistent challenge.

Ligand sterics, electronics and bite angle

Phosphorus ligands define the environment around the metal. Bulky ligands can block one alkene approach, while electronic donation or withdrawal changes the rates of alkene insertion, CO binding and hydrogenolysis. Monophosphines offer one set of coordination geometries; chelating diphosphines hold two phosphorus atoms on the metal and impose a characteristic bite angle.

A narrow or wide diphosphine bite angle changes the relative placement of the alkene, hydride and CO. That geometric bias can favor a linear or branched metal–alkyl, but the outcome depends on the substrate and on competing steps later in the cycle. It is therefore unsafe to label one ligand family as universally “linear-selective” or “branched-selective.”

Substrate structure

Terminal alkenes generally present a clearer linear-versus-branched choice than internal alkenes. Substitution near the double bond changes steric congestion and electronic polarization, and functional groups may coordinate to the metal or react under syngas conditions. Internal alkenes can give several positional products and may isomerize before or during hydroformylation. The catalyst must then control alkene position as well as the orientation of insertion.

Solvent, syngas composition and operating conditions

CO and H2 pressure affect the concentration and coordination state of metal hydride, CO-bound and acyl species. Excess CO can stabilize saturated carbonyl complexes and slow alkene binding; too little CO can change catalyst speciation and increase undesired pathways. The H2:CO ratio influences hydrogenolysis and the rate at which active hydride is regenerated.

Solvent polarity, coordinating ability and viscosity affect ligand exchange, gas solubility and mass transfer. Temperature changes both reaction rates and the relative stability of regioisomeric intermediates, so a condition that improves conversion can reduce L:B selectivity. In a real process, agitation, gas–liquid contact, substrate concentration and heat removal are part of the selectivity problem, not merely engineering details.

Supports, pores and supramolecular confinement

In supported catalysts, pore size, surface functionality and local defects can orient an alkene before insertion. Molecular cages and other confined environments can similarly make one approach easier than another. A supramolecular rhodium system reviewed in 2018 produced outer-to-inner product ratios of 1:9 for trans-2-octene and 1:4 for trans-3-octene. Those values belong to that encapsulated catalyst and those substrates; they are not general hydroformylation constants.

Normal versus iso hydroformylation

Feature Normal (linear, n) Iso (branched, i)
Connectivity for R–CH=CH2 R–CH2–CH2–CHO R–CH(CH3)–CHO
Formyl placement Gives the unbranched carbon chain Creates a branch adjacent to the aldehyde carbon
Typical reason to target it Linear alcohols, plasticizer and detergent intermediates, or predictable chain extension Branched building blocks, altered physical properties, or a specific synthetic target
How it is reported The linear-to-branched ratio, L:B (also n:i), together with conversion and total aldehyde selectivity

An L:B value is a ratio, not a universal percentage. For example, 9:1 corresponds to 90% linear only if the two aldehydes are the sole products being counted; side products and analytical basis can change the calculated percentage. Reports should state substrate, catalyst, conversion, aldehyde selectivity and the conditions used.

How to evaluate a regioselective catalyst

A meaningful comparison records more than the headline L:B number. Use the following checklist when reading a paper or planning an experiment:

  • Conversion: What fraction of the starting alkene reacted?
  • Aldehyde selectivity: How much of the converted material became aldehydes rather than alcohols, hydrocarbons or isomerized alkene?
  • Regioselectivity: Is the L:B ratio measured by calibrated gas chromatography or another quantitative method?
  • Conditions: What temperature, total pressure, CO:H2 ratio, solvent and residence time were used?
  • Catalyst productivity: How many moles of aldehyde are produced per mole of metal, and does performance persist over repeated cycles?
  • Substrate scope: Was the result obtained only with a simple terminal alkene, or does it survive internal alkenes and coordinating functional groups?
  • Recovery: Can the metal and ligand be separated without unacceptable loss or contamination?

Comparisons should also distinguish homogeneous rhodium mechanisms from heterogeneous systems. The former may offer a well-defined molecular cycle; the latter may gain recovery and process advantages while introducing transport and site-distribution effects.

Regioselectivity is not stereoselectivity

Regioselectivity answers “which connectivity?” Linear and branched aldehydes are constitutional isomers. Stereoselectivity answers “which three-dimensional arrangement?” If a reaction creates stereocenters, a catalyst may favor one diastereomer. Enantioselectivity is the preference for one enantiomer over its mirror image.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

These goals can conflict. An asymmetric hydroformylation may need to control regioselectivity and enantioselectivity simultaneously, while maintaining useful activity and suppressing alkene isomerization. A high L:B ratio therefore does not demonstrate enantioselective performance, and an enantioenriched branched aldehyde is not automatically the major regioisomer under every condition.

Common failure modes and how to diagnose them

Good conversion but poor L:B ratio

The catalyst may be very active but allow both insertion orientations. Examine ligand sterics, bite angle and CO loading, then test whether temperature or solvent changes the relative pathways. Confirm that the ratio is not being altered by aldehyde isomerization or analytical overlap.

Low conversion and apparent high selectivity

At low conversion, a ratio based on a small amount of product can look impressive while hiding slow turnover. Report conversion and aldehyde selectivity beside L:B, and check whether the catalyst is inhibited by excess CO or substrate coordination.

Selectivity drifts during a run

Drift can indicate ligand degradation, metal aggregation, support restructuring, changing gas composition or accumulation of strongly coordinating products. Sampling over time and analyzing both liquid products and catalyst-containing phases helps separate deactivation from a simple change in conversion.

Internal alkene gives many products

Allow for alkene migration, multiple insertion sites and geometric isomers. A catalyst optimized on a terminal alkene may not control these additional pathways. Substrate-directed ligands, confinement or a different metal phase may be needed, but each choice must be tested under the intended operating conditions.

Where the chemistry is used

Hydroformylation supplies aliphatic aldehydes for downstream alcohols, acids, amines and other intermediates used in plasticizers, detergents, fine chemicals, pharmaceuticals and agrochemicals. In manufacturing, the preferred catalyst balances linearity with throughput, catalyst cost and metal recovery. In research synthesis, the priority may be a branched or enantioenriched aldehyde that would be difficult to obtain by rearranging a linear product.

The reaction has been studied for decades; a review statement reproduced in a 2018 supramolecular-catalysis article calls it “one of the most intensively explored reactions in the field of homogeneous transition metal catalysis.” Current work extends that foundation to supported catalysts, defect engineering, cooperative metal sites, ligand redesign and confined reaction spaces.

Bottom line for choosing a system

Choose a catalyst by matching the desired aldehyde structure and process constraints, not by searching for a universally best L:B value. Define whether the target is linear, branched or enantioenriched; select the metal phase and ligand environment accordingly; then optimize substrate, solvent and syngas conditions while measuring conversion, aldehyde selectivity, regioselectivity, lifetime and recovery together.

Frequently Asked Questions

Is hydroformylation the same as the oxo process?

Yes. “Oxo process” is the established industrial name for hydroformylation, the addition of CO and H₂ to an alkene to make an aldehyde.

Why can an internal alkene be harder to hydroformylate selectively than a terminal alkene?

An internal alkene can isomerize, present more than one insertion orientation and form several positional or geometric products. The catalyst must control these pathways in addition to choosing linear or branched connectivity.

Does a high L:B ratio prove that a catalyst is better?

No. L:B must be considered with alkene conversion, total aldehyde selectivity, productivity, catalyst lifetime, operating severity and metal recovery. A very selective catalyst that barely reacts may be less useful than a slightly less selective, durable system.

Can regioselective hydroformylation also be enantioselective?

Yes, but the catalyst must control constitutional isomer formation and mirror-image product formation at the same time. Those objectives can compete with activity and often require specialized chiral ligands or substrate-directed designs.
