Airborne environmental DNA (eDNA) is genetic material released by people, animals, plants and microbes into the air or onto airborne particles. Investigators can capture it with air samplers or settling surfaces, extract the DNA and compare a human profile with reference samples. The method can support an inference that someone recently occupied a room, but it cannot by itself prove when that person was there, what they did or that they committed a burglary.

What airborne eDNA actually is

Environmental DNA is genetic material collected from an organism’s surroundings rather than directly from its body. In air, the material may be carried in skin flakes, hair fragments, saliva droplets, respiratory particles, dust and other microscopic debris. A sampler does not “see” a person. It captures particles that may contain DNA, after which a laboratory determines what genetic information is present.

For human-forensic work, the objective is usually an individual association: does the recovered DNA profile correspond to a resident, a person of interest or an unknown contributor? For ecological work, the objective is often species detection: is DNA from a particular bird, insect, plant, bacterium or other organism present? Those goals require different assays, reference databases and reporting standards.

Airborne eDNA is therefore a sampling and laboratory technology, not a camera-based identification system. Detection is probabilistic. A negative result can mean that no target DNA was present, but it can also reflect poor airflow capture, low shedding, degradation, inhibition or an assay that was not sensitive enough for the conditions.

How a room sample could associate an intruder with a scene

1. Define the room and the time window

Investigators first document which room or zone is being tested, when it was last occupied, whether doors or windows were open, how the ventilation system operated and who had legitimate access. They also decide which comparison people are relevant, such as residents, employees, emergency responders or known suspects. Sampling soon after a disturbance is important because later occupants and cleaning can add, remove or redistribute DNA.

2. Capture airborne particles

An active air sampler pulls a measured volume of air through a sterile filter or collection medium. A passive approach leaves a sterile settling surface in place so particles deposit by gravity and normal air movement. Field blanks, unopened filters and room controls are collected alongside the evidence to reveal contamination introduced by equipment, personnel or the laboratory.

3. Extract DNA under controlled conditions

The filter or settling material is processed in a laboratory that separates pre-amplification and post-amplification work areas. Technicians release DNA from the captured material, quantify or assess it where appropriate and run negative controls. Controls matter because airborne particles can move between rooms and because highly sensitive amplification can detect very small amounts of contamination.

4. Select the genetic test

For a human association, laboratories may use short tandem repeat (STR) or single-nucleotide polymorphism (SNP) profiling. A targeted assay can test a defined species or genetic marker. Metabarcoding and high-throughput sequencing examine many taxonomic groups in one run, which is useful when the question is “what organisms are present?” rather than “which one person contributed this profile?”

5. Compare with validated references

A human profile is compared with reference samples collected from known individuals under documented consent and chain-of-custody procedures. Analysts assess whether the sample is a single-source profile or a mixture, whether alleles are missing and whether the result exceeds validated detection limits. The report should state uncertainty, possible contributors, contamination findings and the strength of the association instead of presenting a binary identification.

Active versus passive air collection

Collection approach How it works Strengths Limitations Best fit
Active filtration A pump draws a defined air volume through a sterile filter or other medium. Known sampled volume, faster deployment and the ability to standardize flow rate and duration. Requires powered equipment, can disturb airflow, and a small sampled volume may miss unevenly distributed particles. Time-sensitive scene work and controlled comparisons between rooms or time points.
Passive settling surface A sterile plate, film or other surface is exposed while particles settle naturally. Simple, quiet and suitable where powered equipment is impractical. Collection depends on gravity, turbulence, exposure time and surface placement; sampled volume is not directly known. Longer deployments and supplementary sampling of dust-producing environments.

Neither method automatically produces a better answer. Active sampling generally improves control over air volume and timing, while passive collection can capture deposition over a longer period. A defensible protocol records sampler location, flow rate when applicable, exposure duration, filter lot, environmental conditions and every person who entered the sampling area.

Human profiling and species metabarcoding answer different questions

Feature Human individual association Species detection or biodiversity survey
Primary question Is DNA consistent with a particular person or set of people? Which species or taxonomic groups are represented?
Typical analysis STR or SNP profiling with comparison to reference profiles. Targeted assays, metabarcoding and high-throughput sequencing.
Interpretation Supports an association, subject to mixtures, transfer and contamination. Indicates biological presence or nearby source, not necessarily an individual animal or a viable pathogen.
Critical controls Reference-sample provenance, negative controls, mixture interpretation and validated statistical reporting. Extraction blanks, sequencing controls, barcode validation and an appropriate reference database.
Operational priority Sampling soon after a defined occupancy event. Repeated, spatially planned sampling to detect arrival, spread or seasonal change.

What the 2026 simulated-burglary study showed

A controlled study reported in Forensic Science International: Genetics in 2026 collected 391 eDNA samples from air, dust and surfaces in 22 households. After 15-minute simulated burglaries, airborne samples collected 30 minutes later identified the intruders in 82% of cases in one laboratory and 36% in the other.

Those figures demonstrate both potential and variability. They are controlled-study detection results, not a universal courtroom accuracy rate. The authors associated the difference between laboratories with factors including household occupancy patterns, how readily people shed DNA, cleaning, environmental conditions and laboratory procedures. A real scene can be more complicated because the time of entry may be unknown, several people may have occupied the room and ventilation may have mixed particles from elsewhere.

Why an air-DNA result can change from one scene to another

Time since occupancy

DNA concentration can fall or become diluted as air is exchanged and particles settle. Sampling immediately after a known event is easier to interpret than sampling after hours or days of normal use.

Ventilation and air movement

Heating, ventilation and air-conditioning systems can carry particles between rooms. Open windows, fans, pressure differences and foot traffic can either remove DNA from the target area or bring material from another location.

Shedding differences

People release different amounts and types of biological material. Skin condition, clothing, activity, breathing and whether a person touched or disturbed dusty surfaces all affect what reaches a filter.

Prior occupants and visitors

A resident’s DNA may remain in a room for a long time. Visitors, cleaners, pets and responders can create mixtures that obscure a recent contributor. A profile consistent with a person is not, on its own, proof that the person arrived during the incident under investigation.

Cleaning and environmental conditions

Vacuuming, wiping, humidity, temperature and dust load alter both particle movement and DNA recovery. Cleaning can lower a target signal while redistributing material to filters, vents or adjacent surfaces.

Contamination and laboratory variation

Because assays can detect tiny quantities, contamination control is central. Field blanks, equipment blanks, extraction negatives and laboratory separation help distinguish scene DNA from background. Different extraction kits, amplification thresholds, sequencing pipelines and analyst decisions can produce different detection rates.

What airborne eDNA can and cannot establish

  • It can support: a finding that DNA associated with a person was present in sampled air or deposited material, and in some circumstances that the person may have occupied the area recently.
  • It cannot automatically establish: the exact time of presence, the route of entry, the person’s actions, ownership of an object, or that the person was the burglar.
  • It requires context: fingerprints, touch DNA, dust patterns, video, access records, witness accounts and other scene evidence may provide the timing or activity information that air sampling lacks.
  • It must account for mixtures: a profile containing several contributors may be difficult to interpret, especially when one contributor is present at a low level.

The 2024 forensic review describes airborne human eDNA as an emerging field and says additional research is needed before routine implementation. That status does not make a properly validated result useless; it means the method’s performance, limitations and statistical interpretation must be demonstrated for the specific laboratory and case type.

A defensible collection and reporting protocol

  1. Define the question. Record the room, event, time window, ventilation state and comparison people or species before opening sampling equipment.
  2. Plan sterile collection. Use active filtration or passive surfaces, include negative controls and document sampler placement, exposure time, air volume where available and environmental conditions.
  3. Protect the chain of custody. Seal filters and surfaces, record transfers and keep pre-amplification work physically separate from amplified products.
  4. Extract and test. Use a validated DNA-extraction method, human STR/SNP profiling or a species assay suited to the question, and repeat or confirm borderline results according to laboratory policy.
  5. Interpret in context. Report mixtures, detection limits, contamination checks, missing data and alternative explanations. Do not convert a probabilistic association into a statement that the sample proves a burglary.

Uses beyond identifying people in rooms

Invasive-species biosecurity

The U.S. Geological Survey is testing airborne eDNA samplers for organisms hidden in shipping containers. Inspections can be difficult, time-consuming and expensive when animals or insects are small or concealed. The agency has described a second phase in which the same air samplers would be applied to an actual cargo shipment, followed by multispecies metabarcoding to identify vertebrate and invertebrate species that may be inside.

Terrestrial biodiversity

Aircraft and fixed-site air samplers can collect DNA from bacteria, plants and vertebrates over broad areas. Repeated sampling can reveal changes in species presence without physically capturing every organism, although wind, altitude, weather and source distance must be modeled when results are interpreted.

Agricultural and pathogen biosecurity

A 2024 study reported in Eurosurveillance detected wild-waterbird DNA at poultry-house air inlets. The finding illustrates how host DNA can reveal a potential pathway for airborne biological threats. Detecting host DNA is not the same as detecting a pathogen, but it can identify where biosecurity attention is warranted.

Invasion management

A 2024 systematic review found that terrestrial eDNA applications are less mature than aquatic applications. Even so, studies have explored arrival detection, spread monitoring, containment, control and eradication. These programs work best when air sampling is combined with conventional surveillance and repeated at locations where a management decision depends on early warning.

How to judge a claim that air DNA “identified” an intruder

  • Ask whether the study or laboratory validated the exact sampler, filter, extraction method and genetic assay used.
  • Check how soon sampling occurred after the suspected entry and whether the room had other occupants.
  • Look for documented field, equipment and laboratory negatives.
  • Determine whether the result is a complete single-person profile, a partial profile or a mixture.
  • Separate “DNA associated with this person was detected” from “this person committed the offense.”
  • Require corroboration from independent evidence before drawing conclusions about actions or timing.

For now, airborne eDNA is most useful as complementary evidence and as a way to generate investigative leads. Its value increases when the occupancy event is tightly defined, the scene is sampled promptly, controls are comprehensive and the laboratory has validated its interpretation procedures.

Bottom line

Airborne eDNA can recover human genetic material from a room without touching a person and can sometimes associate a recent occupant with that space. Controlled burglary simulations show genuine promise, but the large difference between two laboratories in the 2026 study demonstrates why performance is not a single fixed percentage. Airflow, shedding, cleaning, prior occupants, contamination and laboratory protocol all matter. A responsible conclusion treats an air-DNA result as one probabilistic strand of evidence—not a standalone verdict about who entered or what happened.
