What Is a Synthetic Cell? How Scientists Build Cell-Like Systems

A synthetic cell is an engineered, cell-like system that reproduces selected features of living cells. It may contain a membrane, genetic instructions, energy-producing chemistry and internal reactions, but it is not automatically a living organism. Scientists use synthetic cells to isolate how life works, test ideas about life’s origins and develop controllable tools for medicine and biotechnology. No laboratory system has yet combined all the capabilities of a natural cell into a fully autonomous artificial organism that functions outside controlled conditions.

What “synthetic cell” means

The term synthetic cell is an umbrella label rather than a single technical specification. Depending on the project, it can refer to a protocell, artificial cell, cell mimic, artificial organelle or minimal cell. These systems overlap in design, so researchers usually judge them by the functions they demonstrate instead of asking whether they satisfy one universal definition.

A typical synthetic-cell project creates a compartment, puts selected molecular machinery inside it and measures a life-like behavior. That behavior might be gene expression, metabolism, sensing, selective exchange with the surroundings, movement, growth or division. A system can be scientifically valuable while demonstrating only one or two of these capabilities.

This distinction matters because a membrane filled with a cell-free reaction is not automatically a living cell. A natural cell integrates many processes at once: it maintains a boundary, obtains and uses energy, stores and reads information, repairs damage, controls its chemistry, grows and reproduces. Most synthetic systems reproduce a subset of those functions under carefully controlled laboratory conditions.

Are artificial cells alive?

There is no agreed point at which a cell-like system becomes alive. “Alive” usually implies an integrated network that maintains itself, uses energy, processes information, adapts to conditions and produces descendants. Synthetic-cell experiments can reproduce individual capabilities without connecting them into a self-sustaining whole.

For example, a vesicle may produce proteins from DNA, while another project may show membrane growth or chemical sensing. Those are life-like functions, but they do not by themselves establish autonomous life. Researchers therefore describe most current systems as cell-like, protocellular or minimal rather than as artificial organisms.

The National Academies’ 2025 assessment notes that synthetic-cell systems have not been shown to function in open, real-world environments. They depend on defined nutrients, energy sources, temperature ranges, equipment and containment. A future system that maintained its chemistry, reproduced with inherited information and adapted without continuous laboratory intervention would represent a much stronger claim than today’s modular demonstrations.

Three ways scientists build synthetic cells

Construction strategies fall into three broad categories. The boundaries are practical rather than absolute, and a single project can combine methods.

Route Starting point Typical objective What distinguishes it
Bottom-up Non-living molecular components Assemble a new compartment with selected cell functions Begins without an intact living cell; behavior emerges from the assembled parts
Top-down A natural organism and its genome Remove genes and processes to define a minimal or refactored cell Retains the architecture and machinery of a living cell while simplifying it
Middle-out Natural modules combined with synthetic components Use cell extracts or purified machinery inside engineered compartments Mixes biological capability with designed membranes, vesicles or control systems

Bottom-up construction

Bottom-up researchers assemble lipids, polymers, nucleic acids, peptides, proteins, enzymes and other ingredients into a compartment. No complete natural cell is required. The challenge is to make the parts organize themselves and then cooperate. A membrane can concentrate reactions, keep incompatible chemistries apart and create an inside that can be regulated independently of the outside.

Top-down construction

Top-down work starts with an existing living organism. Scientists delete genes, redesign the genome or remove cellular functions to determine what is essential. The result is a reduced natural cell, not a cell built from raw materials. Because it inherits much of its machinery from biology, it can perform integrated functions that are difficult to recreate from scratch, while still depending on a living lineage.

Middle-out construction

Middle-out systems occupy the practical space between those extremes. A project might place a cell extract, ribosomes or a purified translation system inside a synthetic vesicle, then add designed control circuits or an engineered membrane. This approach gives researchers access to sophisticated biological modules without requiring an intact organism.

How a synthetic cell is built

Although designs vary, most projects follow a sequence of engineering decisions. The order is useful for understanding what a demonstration actually proves.

1. Form a boundary

The first task is to create an inside and an outside. Lipids can form bilayer membranes similar to those of natural cells. Polymers, peptides, nucleic acids and hybrid materials can also create shells or membrane-like barriers. Components may self-assemble when their chemistry favors a closed structure, or researchers may use a template, interface, support or microfluidic device to impose the desired shape.

A boundary is more than packaging. It controls which molecules enter or leave, prevents useful reactants from diffusing away and allows concentration gradients to develop. Without compartmentalization, reactions that work in a test tube may not interact in a coordinated way.

2. Establish an internal reaction space

Researchers encapsulate purified proteins, enzymes, ribosomes, nucleic acids, cell extracts or complete cell-free transcription-and-translation mixtures. The interior may contain template DNA and an energy donor. In a suitable buffer, the DNA can be transcribed into RNA and translated into protein inside the compartment.

Encapsulation is technically demanding. The compartment must be large enough to hold the machinery but stable enough to avoid leaking it. Mixing, crowding, acidity, salt concentration and temperature all affect whether the internal reactions continue.

3. Add an information system

Some systems use added DNA or RNA as instructions. Others rely on a cell extract that already contains information-processing machinery, or on a reduced natural genome in top-down work. Information can be used to produce a fluorescent reporter, an enzyme, a membrane component or a regulator that changes the compartment’s behavior.

4. Supply energy and materials

Cell-like reactions need an energy source and raw materials. An energy donor can drive RNA and protein production; nutrients or chemical substrates can support metabolism. In current systems, these inputs are generally supplied by the experimenter. A compartment that consumes a defined fuel is demonstrating energy use, but not necessarily an independent metabolism that can find, process and replenish its own resources.

5. Add selected functions

Researchers combine modules according to the question they want to test. Possible functions include:

  • Sensing: detecting a chemical, signal or environmental change.
  • Selective exchange: allowing chosen molecules through while retaining others.
  • Gene expression: transcribing RNA and producing proteins.
  • Metabolism: converting substrates through controlled reaction pathways.
  • Energy management: generating or allocating chemical energy.
  • Information processing: responding to inputs through genetic or biochemical circuits.
  • Movement: changing position or driving internal components.
  • Growth and division: increasing material and splitting into daughter compartments.

Demonstrating one item on this list does not establish the others. A system that expresses a gene may have no metabolism; a system that divides may not copy genetic information into both daughters.

6. Control and measure the behavior

Microfluidic devices can produce uniform vesicles, deliver nutrients and impose controlled changes. Microscopy reveals membrane shape, movement, leakage, growth and division. Biochemical assays measure products, reaction rates and energy use. Researchers may also vary temperature, fuel concentration, membrane composition or genetic instructions to determine which component caused the observed behavior.

Materials used in cell-like systems

The boundary and interior are chosen together. A membrane that is excellent for stability may be poor at exchanging nutrients; a highly permeable shell may allow the reaction products to escape.

Protocell, synthetic cell and artificial organelle: what is the difference?

Protocell

A protocell is a model of an early or prebiotic cell. The emphasis is often on plausible chemistry, compartment formation and primitive metabolism rather than on reproducing the machinery of modern biology. Protocell experiments are central to origins-of-life research.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Synthetic or artificial cell

These terms generally describe an engineered cell-like compartment built to perform selected functions. “Synthetic” often highlights deliberate construction, while “artificial” emphasizes that the system is not a naturally occurring cell. In practice, researchers use the labels with considerable overlap.

Artificial organelle

An artificial organelle reproduces a specialized cellular task, such as catalysis, energy conversion or molecular storage, without attempting to recreate a complete cell. It may operate inside a synthetic compartment or be designed for use in a natural cell.

Minimal cell

A minimal cell is usually produced through top-down reduction of a natural organism. Its genome and machinery are pared down to the functions judged necessary for growth under defined conditions. It remains biologically derived even when its genome has been extensively redesigned.

  1. What construction route was used? Identify bottom-up, top-down or middle-out methods.
  2. What forms the boundary? Check whether it is lipid, polymer, nucleic-acid, peptide or hybrid.
  3. Where does the information come from? Look for added DNA or RNA, a cell extract or a reduced natural genome.
  4. What supplies energy? Determine whether fuel is continuously added, enclosed at the start or generated internally.
  5. Which functions were directly demonstrated? Separate measurements from proposed future capabilities.
  6. How autonomous is the system? Ask how much intervention, nutrient delivery and environmental control it requires.
  7. Can it grow and divide? Check whether division is repeatable and whether information and machinery are inherited by daughter compartments.
  8. What is the containment context? A behavior observed in a microfluidic chip is not evidence that the system works in an open environment.

What synthetic cells can do today

Current systems are useful precisely because they let scientists isolate difficult biological mechanisms. A compartment can show how a membrane controls transport, how a genetic circuit responds to a signal or how enzymes cooperate when concentrated in a small volume. Cell-free expression systems can produce proteins without maintaining a complete organism, making it easier to alter one component at a time.

The same controllability supports miniature bioreactors, biosensors and experimental drug-delivery vehicles. Researchers are exploring compartments that detect chemicals or pathogens, release therapeutic molecules in response to signals, and manufacture proteins or other materials. Origins-of-life studies use related systems to test whether simple chemistry could have become increasingly organized.

These applications remain research directions rather than proof that a general-purpose artificial cell is ready for deployment. Performance depends on the chosen membrane, reaction mixture, energy source and containment method.

Why making a complete artificial cell is difficult

The central problem is integration. It is relatively straightforward to demonstrate gene expression, membrane formation or a metabolic reaction separately. It is much harder to make those processes support one another continuously.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Compartmentalization: The boundary must be stable, yet permit controlled exchange and growth.
  • Energy supply: Reactions consume fuel, and the system must avoid exhausting its energy reserve.
  • Metabolism: Pathways need balanced inputs and outputs rather than a single isolated reaction.
  • Information processing: Instructions must be copied, read and regulated without losing control of the chemistry.
  • Growth: New membrane and internal machinery must be made in the correct proportions.
  • Division: Splitting must produce viable daughters that inherit the necessary components.
  • Environmental control: Temperature, pH, salts and nutrients must remain within workable ranges.

Natural cells solve these problems through networks refined over evolutionary time. Synthetic-cell researchers must design compatible interfaces between modules that were developed separately. A successful demonstration of one module therefore does not remove the remaining engineering barriers.

How close are we to creating life in the lab?

Scientists are closer to reproducing individual cellular functions than to creating a fully autonomous artificial organism. Bottom-up projects have produced increasingly sophisticated compartments and reactions; top-down projects have clarified how much of a natural genome can be removed; middle-out designs combine biological machinery with engineered control. None of these achievements, by themselves, establishes a self-sustaining synthetic life form.

The most defensible description today is that synthetic cells are controlled research systems with selected life-like properties. They are valuable even when they are not alive in the ordinary sense: by reducing a cell to inspectable parts, they reveal which functions require one another and which can operate independently. The field’s long-term goal is an integrated system that can maintain itself, process information, use energy, grow and divide, but that goal has not yet been reached in an open, real-world setting.

Why the distinction matters

Keeping natural, genetically modified cells separate from bottom-up synthetic cells prevents confusion. A modified bacterium is still a living organism descended from a natural cell, even if its genome has been redesigned. A bottom-up protocell begins with assembled components and must earn each capability through demonstrated chemistry. Reporting which functions are present—and which are absent—is more informative than applying the word “artificial” alone.

A 2024 Nature Reviews Molecular Cell Biology viewpoint by Katarzyna P. Adamala, Marileen Dogterom, Yuval Elani, Petra Schwille, Masahiro Takinoue and T‑Y Dora Tang described the prospect of a fully synthetic cell as one that could revolutionize biology, medicine and biotechnology. The viewpoint was published as the version of record on 15 December 2023 for the 2024 volume. An ACS review published online on 26 March 2024 likewise reflects a field focused on combining modules rather than claiming that a complete artificial organism already exists.
