A chemical reaction network is a formal map of molecule types, the reactions that convert them, and any catalytic effects that speed those reactions. In origin-of-life studies, such maps let scientists test whether a set of reactions could obtain its ingredients from an environment, regenerate the molecules it needs, and organize into a self-sustaining chemical system. RAF theory makes those tests precise. A network that passes a RAF test is a consequence of the chosen model, not proof that the same network existed on early Earth or became life.
What a chemical reaction network represents
Chemistry is often described one reaction at a time: reactants become products under particular conditions. A chemical reaction network (CRN) looks at many such reactions together. It records which molecular types are present, which transformations are possible, and how the products of one reaction can supply another. The representation can be used for an actual laboratory mixture, a proposed prebiotic pathway, or an abstract mathematical system.

In the formal framework used for reflexively autocatalytic and food-generated (RAF) sets, a chemical reaction system has four ingredients:
| Component | What it means | Question it supports |
|---|---|---|
| Molecule types | The substances that may occur in the system, including reactants, products and catalysts. | What chemical entities are available to participate? |
| Reactions | Rules connecting reactants to products. | Which transformations can occur? |
| Catalysis assignments | Statements that a molecule type catalyzes, or accelerates, a particular reaction. | Which molecules can make other reactions proceed faster? |
| Food set | Molecule types assumed to be supplied by the surroundings. | What can the system draw from its environment instead of making internally? |
The food set is a modeling choice, not a claim that those molecules were abundant everywhere on the early Earth. One model might treat simple gases or small organic compounds as environmental inputs; another might choose a different set. Changing that boundary can change which reactions appear sustainable.

Graph view: more than a list of reactions
A CRN can be drawn as a directed graph. Reaction nodes connect reactant molecules to product molecules, while separate links indicate which molecule types catalyze which reactions. This makes dependencies visible: a product may become the catalyst for a later reaction, creating a feedback loop. The graph can also show dead ends, competing pathways and molecules that must be imported continuously.
The graph is a bookkeeping device. It does not automatically contain concentrations, temperatures, reaction rates, energy supplies or spatial effects. Those details require additional chemical or dynamical models.
What “autocatalytic” means in a network
An autocatalytic system contains feedback in which products help catalyze reactions that produce members of the system. The feedback can be collective. It does not require every molecule to catalyze its own direct copy, and it does not require one reaction to make a complete organism-like entity.

Suppose the environment supplies molecules in a food set F. A selected group of reactions can generate additional molecules from F. The set of everything reachable by repeatedly applying those reactions is called the food-generated closure. A reaction belongs to a RAF set when two conditions hold:

Together, these conditions describe a network-level catalytic organization. They say that the reactions support one another and that their raw materials are reachable from specified environmental inputs. They do not say that each step is fast, favorable, selective or experimentally demonstrated.
Why collective autocatalysis matters
Collective autocatalysis offers a possible bridge between scattered prebiotic reactions and a sustained chemical system. A molecule that is initially rare may be produced by one reaction, then catalyze another reaction that produces a different catalyst. Once enough of the loop is operating, the whole set can reinforce its own activity. The system-level property can therefore exist even when no single molecule is a self-replicating “organism.”
This distinction prevents a common misunderstanding. A RAF result is not evidence that one molecule copied itself exactly. It identifies a closed pattern of catalytic support under the assumptions encoded in the network.
RAF and CAF: two different standards of organization
RAF is not the only way to describe an autocatalytic set. A constructive autocatalytic and food-generated (CAF) set satisfies the RAF conditions and adds an ordering requirement. The reactions must be arrangeable step by step so that, when each reaction is reached, its reactants and at least one catalyst are already in the food set or were produced by earlier reactions.

| Axis | RAF | CAF |
|---|---|---|
| Catalysis and food generation | Every reaction has an available catalyst, and all reactants are food-generated relative to the set. | Meets all RAF conditions. |
| Initiation | The network may qualify structurally even if a spontaneous, uncatalyzed reaction is needed before catalytic activity starts. | Each reaction has a place in a constructive order with its reactants and catalyst already available. |
| Interpretation | A candidate self-sustaining catalytic organization under the model. | A more direct stepwise pathway under the model’s ordering condition. |
The distinction matters for origin-of-life arguments. A RAF can reveal a complete catalytic pattern in the reaction inventory while leaving open how the first member of that pattern appeared. A CAF provides stronger information about a possible sequence of activation, but it is still a formal result rather than a historical record.
How the RAF algorithm finds a self-supporting set
Wim Hordijk’s concise formal definition (2023) describes an iterative procedure. In plain language, it tests whether the proposed reactions can survive repeated checks for food access and catalytic support.
- Specify the system. List molecule types, reactions, catalysis assignments and the food set. The result can only be as meaningful as these inputs.
- Start with the food set. Treat every molecule in
Fas available without requiring an internal reaction. - Expand the closure. Add products of reactions whose reactants can already be made from the current closure.
- Check catalysts. Remove any reaction whose assigned catalysts are neither in the food set nor generated by the reactions still under consideration.
- Repeat. Recalculate the closure and remove newly unsupported reactions until no further change occurs.
- Read the result. If a nonempty set remains, the formal procedure returns the unique maximal RAF contained in the input system. An empty result means no RAF exists under those exact molecule, reaction, catalysis and food-set assumptions.
“Maximal” means that the algorithm retains the largest self-supporting subset identified by the procedure; it does not mean the subset is chemically optimal or historically inevitable. The iteration also does not simulate concentrations over time. It answers a structural question: can these reactions collectively provide their reactants and catalysts from the chosen inputs?
Why this is relevant to the origin of life
From loose chemistry to organized metabolism
Before cells had genetically encoded proteins, any sustained chemical organization would have needed to arise from ordinary molecular reactions. Network models let scientists ask whether simple environmental compounds could feed a web of transformations that maintains its own catalytic machinery. That is a narrower and more testable question than asking a model to produce “life” in one step.

Reviews published in 2018 and 2019 describe autocatalytic networks as possible intermediates between prebiotic chemistry and later systems based on genes and enzymes. The proposed transition is gradual: environmental molecules enter, reactions generate new compounds, some products accelerate further transformations, and a network becomes more organized and persistent than an isolated reaction would be.
Metabolism-first interpretations
A 2020 review of autocatalytic chemical networks at the origin of metabolism discusses RAF analyses of modern metabolic networks. Those analyses have been interpreted as consistent with an autotrophic origin of metabolism, in which a network could build organic compounds from relatively simple environmental inputs. “Consistent with” is the important qualification. The result supports a possible scenario; it does not identify the actual route taken by ancient chemistry.
What a model contributes
- It makes assumptions explicit: which molecules count as food, which reactions are allowed and which catalytic effects are assigned.
- It exposes dependencies that are hard to see in a long list of reactions.
- It provides an algorithmic test that different hypotheses can use and compare.
- It separates network organization from later questions about growth, reproduction and evolution.
What a RAF result cannot establish
A formal RAF or CAF property is necessary evidence for a proposed chemical organization, but it is not a demonstration that the organization operated on early Earth. Several additional issues must be addressed.

Autocatalysis is not yet Darwinian evolution
Self-sustaining chemistry and biological evolution overlap conceptually, but they are not the same criterion. A RAF can maintain catalytic activity without storing an inheritable description of itself. It may also lack variants that compete, reproduce with fidelity or differ in fitness.
A 2021 review of self-reproduction and Darwinian evolution in autocatalytic chemical reaction systems evaluates candidate systems against several requirements:
- Self-reproduction: The system must generate more of the organization, not merely produce a useful catalyst once.
- Variation: More than one heritable form or pathway must be possible.
- Heredity: Differences must persist across cycles well enough to be transmitted.
- Differential fitness: Some variants must reproduce or persist more effectively than others.
A network can therefore be a plausible chemical precursor without being an evolving population. Origin-of-life scenarios need a bridge from catalytic closure to copying, inheritance and selection, whether those functions arise in the same network or through later additions.

Imagine a hypothetical system with food molecules A and B. Reaction 1 combines them to make C; reaction 2 uses C to make D; and D catalyzes reaction 1. If reaction 1 can occur uncatalyzed, the system may form a RAF: the food set supplies the reactants, reaction 1 makes C, reaction 2 makes D, and D supports reaction 1. The loop is collective, because neither reaction alone supplies every catalyst and reactant.
To qualify as a CAF, the reactions would need an order satisfying the catalyst condition at each step. If reaction 1 requires D before any D can be made, no such order exists unless an alternative source of D is placed in the food set or another initiating reaction is added. This example shows why RAF and CAF answer related but different questions.
How to evaluate an origin-of-life claim based on a network
- Inspect the boundary. Ask which molecules are in the food set and whether the environmental assumption is stated for a specific setting.
- Separate reaction existence from catalysis. Check whether each reaction and each catalytic assignment has chemical or experimental support.
- Check initiation. Determine whether the set is merely a RAF or also a CAF, and identify any required uncatalyzed first event.
- Ask about dynamics. Look for concentration, rate, energy, degradation and transport information beyond the topology.
- Test robustness. See whether removing an uncertain reaction or catalyst destroys the set, or whether several alternative pathways remain.
- Look for evolutionary functions. A claim about life must address reproduction, variation, heredity and differential success, not only catalytic closure.
This checklist keeps the conclusion proportional to the evidence. Network theory can show that a proposed form of chemical organization is internally possible under explicit assumptions. Laboratory chemistry and geochemical modeling are needed to determine whether those assumptions describe a real environment, and evolutionary analysis is needed to connect persistence with life.
The practical takeaway
Chemical reaction networks give origin-of-life studies a rigorous language for linking reactions, catalysts and environmental inputs. RAF theory identifies collective catalytic systems whose reactants and catalysts can be generated from a defined food set; CAF theory asks whether that organization can be built in a stepwise order. These tools sharpen hypotheses about how metabolism might have begun, including interpretations compatible with an autotrophic origin, but they do not settle the historical origin of life. The decisive next questions concern chemical plausibility, dynamics, environmental persistence and the emergence of heritable evolution.
