Under anoxia, the researchers report an anaerobic pathway that converts pyruvate through ferredoxin oxidoreductase to hydrogenase, and they experimentally demonstrated hydrogen gas production by a mitochondria-localized, oxygen-labile hydrogenase. The findings reveal a possible avenue for studying the organism, not a treatment for infection.
What the study mapped
The researchers combined mitochondrial immunoprecipitation, density-gradient purification, mass spectrometry, protein-correlation profiling and microscopy to assemble AcMitoCarta, a high-confidence inventory of A. castellanii mitochondrial proteins. The Broad Institute reports that the inventory contains 1,122 proteins; 381 lacked readily identifiable homologs in human and yeast mitochondria. Complexome analysis also highlighted 20 macromolecular assemblies. These are inventory and analysis counts, not measures of disease frequency or clinical risk.
The work was published in Cell in 2026 as “Mitochondrial proteome of Acanthamoeba delineates aerobic and anaerobic pathways dynamically regulated by oxygen” (volume 189, issue 20, pages 6246–6266.e19; DOI 10.1016/j.cell.2026.08.056).
How mitochondrial energy metabolism changes with oxygen
Proteomic and transcriptomic profiling showed extensive oxygen-linked rewiring of mitochondrial bioenergetic machinery. In the study’s account, oxygen availability is associated with a shift between aerobic machinery and anaerobic energy pathways. Under anoxia, the researchers report induction of a pathway involving pyruvate:ferredoxin oxidoreductase and hydrogenase.

The distinction between profiling and functional evidence matters: changes in protein or transcript patterns indicate which machinery is associated with oxygen conditions, while the hydrogen-production experiment demonstrates a function under anoxia. The available source summaries do not provide the precise oxygen levels, sample counts or effect sizes needed to quantify the transition.
Why the findings matter—and what they do not show
A. castellanii can cause sight-threatening corneal infection, including keratitis. A detailed mitochondrial map could help researchers investigate how the organism adapts and identify candidate targets for future study. That therapeutic relevance is prospective. The reported work does not establish a new drug, test a therapy, or show clinical efficacy or patient outcomes. Its findings concern A. castellanii; they should not be assumed to apply identically to every Acanthamoeba species or to oxygen conditions inside an infected human cornea.
