Leucine is an essential amino acid: your body uses it to make proteins, but cannot make it itself, so it must come from food. It also acts as a nutrient signal linked to muscle protein synthesis, although that signal cannot replace the other essential amino acids needed to build muscle protein.
How leucine supports muscle protein synthesis
Leucine is one of three branched-chain amino acids (BCAAs), alongside isoleucine and valine. Like other amino acids, it is incorporated into proteins. It also signals that amino acids are available, working alongside insulin’s signal of energy availability in the regulation of muscle protein synthesis. Research describes leucine-associated signaling through mTOR and the machinery that initiates protein translation. Garlick’s review and a review of leucine metabolism discuss these mechanisms.

That signaling role helps explain why leucine is studied in relation to muscle, but it does not mean leucine alone builds muscle or guarantees a lasting increase in muscle mass. Muscle protein synthesis requires essential amino acids as building material. A leucine-only supplement does not supply the other amino acids needed to construct proteins.

The 2023 review covers this response.

What leucine does in energy metabolism
Leucine is metabolized through branched-chain amino-acid pathways and can contribute to energy metabolism. Its breakdown also produces metabolites including α-ketoisocaproate (α-KIC) and HMB. These are biochemical roles, not evidence that taking leucine supplements boosts everyday energy, burns fat, or improves health outcomes. A review of leucine and its metabolites discusses findings across experimental settings.
Brown fat offers a specific example of BCAAs being used in energy production: NIH Research Matters describes a study in which the protein SLC25A44 helps transport BCAAs into mitochondria during cold exposure, where they can contribute to heat generation. This is research on brown-fat biology, not a consumer trial showing that leucine supplements make people burn more calories or feel more energetic. NIH Research Matters reports on the study.

Foods and supplements that provide leucine
Leucine comes from protein foods. Animal foods with complete proteins provide it alongside essential amino acids; complementary plant foods can also provide all essential amino acids when eaten in combination. Complete-protein powders are another source. NIH’s Office of Dietary Supplements (ODS) notes that whey contains more leucine than casein or soy, but that comparison does not make whey necessary or establish a particular product as superior. The ODS fact sheet covers these sources.
| Option | What it provides | Practical consideration |
|---|---|---|
| Whole-food protein | Protein foods supply leucine; complete animal proteins provide all essential amino acids, and complementary plant foods can provide them together. | A way to obtain leucine as part of dietary protein. |
| Complete-protein powder | Protein with essential amino acids; ODS notes that whey has more leucine than casein or soy. | One format for complete protein, not a requirement or proof of product superiority. |
| Isolated leucine or BCAA supplement | Leucine alone, or the three BCAAs; it does not necessarily provide all essential amino acids. | It cannot substitute for the full essential-amino-acid supply needed to build protein. |
Do you need a leucine or BCAA supplement?
The practical priority is adequate high-quality protein and a complete supply of essential amino acids, rather than a leucine supplement by default. ODS reports inconsistent evidence that BCAA supplements add muscle-building benefit beyond sufficient high-quality protein. It also says studies have not consistently shown that BCAAs—or any of the three amino acids taken singly—improve exercise performance, build muscle mass, or aid recovery. Read the ODS summary of the evidence.
