What De-Extinction Can—and Cannot—Tell Us About Aging and Longevity

Short answer: De-extinction can improve our understanding of ancient DNA, comparative genetics and selected traits in living relatives. It does not show that aging can be reversed, that extinct animals can be recreated as exact genetic copies, or that humans can live longer. The key is to keep three questions separate: how a trait is reconstructed, how biological age is measured, and whether an intervention extends healthspan or lifespan.

De-extinction is trait reconstruction, not a time machine

In common usage, de-extinction means using ancient genetic information and modern biotechnology to make a living animal express selected characteristics associated with an extinct relative. It usually involves a living host species, genome analysis and targeted genetic changes. It does not mean recovering an intact extinct genome, producing a perfect clone and restoring the original species in every genetic, behavioral and ecological detail.

That distinction matters for longevity claims. A project can demonstrate that researchers identified variants associated with a historical trait and introduced some of those variants into a living animal. That is evidence about genetic reconstruction. It is not evidence that the animal has been rejuvenated, that its aging process has changed, or that the same edits would extend human life.

What a de-extinction project can demonstrate

  • Ancient DNA can preserve information useful for comparing extinct and living lineages, even though it is often fragmented and chemically damaged.
  • Researchers can infer candidate variants linked to visible characteristics and alter a living relative’s genome.
  • The resulting animal can be evaluated for resemblance in traits such as body size, coat characteristics or skull morphology.
  • Comparisons among extinct and living mammals can reveal evolutionary relationships and biological differences worth studying.

What it does not demonstrate

  • It does not recreate an extinct animal’s complete genome, development, behavior, microbiome or environment.
  • It does not establish that the edits affect aging pathways, lifespan or age-related disease.
  • It does not provide a human treatment, a consumer anti-aging test or a proven route to longer life.

Colossal’s own description of its approach says the goal is to make close genomic approximations of extinct species rather than clones. The company also acknowledges that resulting animals cannot be genetically, behaviorally and psychologically identical to their extinct ancestors. That is a useful description of the company’s method and goals, not independent proof of a longevity effect.

What the dire-wolf example actually shows

A prominent case illustrates the difference between a reconstructed trait and a resurrected species. In an April 14, 2025 report (modified May 27, 2025), the University of California, Santa Cruz described pups called Romulus and Remus as genetically modified gray wolves endowed with traits of the extinct dire wolf. Pieces of genetic code identified in ancient dire-wolf DNA were used to alter gray-wolf genomes. The university’s caption and text make clear that the pups were not exact replicas.

The report says the dire-wolf genome analyzed by the study team appears to derive roughly two-thirds from an ancestor of wolves and coyotes, with the remaining third from an ancient lineage related to South American dogs. The team presented this pattern as evidence consistent with hybrid speciation around 4 to 5 million years ago. Those proportions and the evolutionary interpretation should be understood as findings reported by that study team, rather than as an unqualified, timeless fact about every dire-wolf genome.

Beth Shapiro, an evolutionary biologist and Colossal chief science officer, described the achievement in the same University of California, Santa Cruz account:

“Ancient DNA is notoriously difficult to work with because it is often fragile and degraded, but paleogenomics has made tremendous leaps forward in the past 20 years, leading to the recent groundbreaking achievement of the world’s first de-extinction with the birth of Colossal’s dire wolves,” Shapiro said.

“World’s first de-extinction” is Shapiro’s characterization, quoted by the university. The operational result described in the account is more precise: genetically modified gray wolves carrying selected traits inferred from an extinct relative. Calling them identical dire wolves would erase the role of the host species, the limited set of edits and the missing historical environment.

Why the distinction matters for aging

A visible trait and an aging phenotype are different endpoints. A researcher can measure whether an edited wolf has a particular coat pattern or body form. Showing a change in aging would require a separate study: defined age groups, validated biomarkers, health outcomes, survival tracking and an appropriate unedited comparison. None of those requirements is satisfied merely by producing an animal with selected dire-wolf characteristics.

What an epigenetic clock is

An epigenetic clock is a statistical model that estimates age-associated biological patterns, commonly by measuring DNA methylation at many locations in the genome. DNA methylation is a chemical mark that helps regulate gene activity. Its patterns change with age in ways that can be modeled across tissues or species.

The output is an estimate or prediction, not a stopwatch and not a therapy. A clock may estimate chronological age, identify that a sample looks older or younger than expected, or correlate with later health outcomes. Those uses can be scientifically valuable while remaining different from proving that aging has been slowed or reversed.

The cross-mammal clock study

A peer-reviewed 2023 paper in Nature Aging, “Universal DNA methylation age across mammalian tissues,” assembled a large comparative dataset: 11,754 methylation arrays from 59 tissue types across 185 mammalian species. The Mammalian Methylation Consortium reported predictive models with correlation r greater than 0.96. In the evaluated models, cross-validation results included median absolute error below one year and relative error below 3.3%.

Those figures describe model performance in the study’s dataset and validation framework. They are not universal guarantees for every species, tissue, laboratory method or consumer test. A model trained across mammals can be powerful for comparative biology while still requiring careful validation when applied to a new organism or sample type.

The paper also reported that age deviations were associated with human mortality risk, mouse somatotropic-axis mutations and caloric restriction. “Associated with” is the critical wording. A statistical relationship between a methylation-based age measure and an outcome does not show that changing the clock reading will produce that outcome, or that the clock itself is the biological cause.

What human studies add

The National Institute on Aging summarized a separate body of human research in 2024, reporting that DNA-methylation-based biological-age measures can help predict age-related health outcomes, chronic disease, cognitive and functional limitations, and mortality in older adults. The strength and pattern of association differed somewhat by clock type. This supports the use of methylation measures as research indicators, while leaving open questions about causation, clinical thresholds and which interventions, if any, can reliably change meaningful health outcomes.

De-extinction and longevity answer different questions

Comparison axis De-extinction work Aging and longevity research
Organism and material Extinct DNA fragments, reconstructed sequences and a living relative used as a host Living humans or animals, tissues, cells, methylation data and longitudinal health records
Typical method Ancient-genome analysis, candidate-variant selection and gene editing DNA-methylation measurement, physiological tests, genetic studies and controlled interventions
Primary endpoint Resemblance to selected historical traits or clarification of evolutionary relationships Age prediction, disease risk, healthspan, survival or lifespan
Evidence example Institutional reporting on genetically modified gray wolves with selected dire-wolf traits; company explanations of genomic approximation Peer-reviewed model validation and government summaries of associations with health outcomes
Main extrapolation limit A trait in a living relative does not recreate the extinct organism or its life history An age estimate or association does not prove that an intervention extends life

The fields overlap in one legitimate way: comparing species can reveal conserved biological processes. A shared molecular pattern may suggest a mechanism worth testing. It remains a hypothesis until an intervention study shows that changing the mechanism improves healthspan or lifespan in a specified organism.

Can bringing back extinct animals extend human life?

No evidence in the sources reviewed shows that de-extinction itself reverses human aging or extends human lifespan. The dire-wolf work concerns reconstructing selected traits in gray wolves. The methylation-clock work concerns estimating age and studying associations with health outcomes. Neither is a human longevity intervention.

A credible claim that a discovery extends human life would need to identify the intervention, the biological target, the population, the control group, the duration and the endpoint. Depending on the claim, endpoints could include disease-free survival, functional capacity, validated healthspan measures or lifespan. A resemblance study, a correlation coefficient or a company target cannot substitute for that evidence.

How a comparative finding could become a longevity hypothesis

  1. Identify a conserved mechanism. Researchers would first need evidence that a molecular pathway is shared across relevant species and plausibly connected to aging.
  2. Test causality in a model organism. Editing or pharmacologically changing the pathway would need controlled experiments with appropriate untreated and sham-edited groups.
  3. Measure meaningful outcomes. A lower methylation age alone would not be enough; studies would need health, function, disease and survival measures.
  4. Check safety and durability. Gene edits can have off-target effects, tissue-specific consequences and uncertain long-term risks.
  5. Translate cautiously to humans. Human trials would need their own evidence because species differ in metabolism, immune response, development and lifespan.

How to read claims about de-extinction and aging

Use five checks before accepting a headline or promotional statement.

  1. Name the organism. Is the result about an extinct species, a living host, a mouse, a human tissue sample or people?
  2. Identify the biological material. Ancient DNA, edited nuclear DNA, methylation arrays and clinical measurements answer different questions.
  3. Find the endpoint. Is the study measuring a visible trait, an age prediction, a disease association, healthspan or lifespan?
  4. Classify the evidence. A company explanation, a university report, a peer-reviewed model paper and an intervention trial have different evidentiary roles.
  5. Check the extrapolation. Ask what must be true for a result in one species or tissue to apply to another.

For example, “the clock predicts age with correlation r greater than 0.96” is a model-performance statement. “The clock makes animals younger” is an intervention claim and does not follow from it. Likewise, “edited gray wolves express selected dire-wolf traits” is supported by the university account; “dire wolves have been perfectly resurrected” is a broader claim that the account does not support.

Common mistakes and how to correct them

Mistake: treating a host animal as an exact copy

Correction: Describe the host species, the edits and the traits observed. Include what remains unknown about behavior, development, physiology and environment.

Mistake: treating biological age as a literal age reversal

Correction: Say that a methylation clock estimates age-associated patterns. If an intervention changes the estimate, that still requires independent evidence of improved function or longer survival.

Mistake: turning an association into a treatment

Correction: Preserve the study’s wording. Associations can generate hypotheses; randomized or otherwise controlled intervention evidence is needed to establish an effect.

Mistake: ignoring species and tissue limits

Correction: State whether a result was measured in blood, another tissue, a cell culture, a mouse or a human cohort. A clock trained on one context may not transfer unchanged to another.

Mistake: presenting a company’s framing as a consensus

Correction: Attribute the statement to the company, then use independent institutional or peer-reviewed sources for what was actually measured.

What this research can contribute to aging science

De-extinction projects can expand comparative datasets and force researchers to reconstruct how traits evolved. Ancient DNA may reveal which variants appeared, disappeared or moved among lineages. Living relatives provide experimental systems in which candidate variants can be studied. These are legitimate contributions to evolutionary and comparative biology.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The strongest bridge to aging is methodological rather than therapeutic. Cross-species datasets can help identify molecular patterns that remain stable across mammalian evolution. Methylation clocks can quantify age-associated change and provide a common measurement framework. Together, those tools may help researchers formulate better questions about conserved aging biology.

The bridge ends where causal claims begin. The reviewed evidence does not show that making a gray wolf more like a dire wolf rejuvenates it, that an extinct-animal genome contains a transferable human longevity program, or that a high-performing age clock is itself a way to extend life.
