What are the main cell types in the small intestine and what do they do? The small intestine uses a crypt–villus epithelium in which different cells divide the work of digestion, absorption, barrier protection, microbial control, hormone release and immune surveillance. Enterocytes absorb most nutrients, ions and water; goblet cells make mucus; Paneth cells defend the crypts and support stem cells; enteroendocrine cells release gut hormones; tuft cells sense chemical signals and help initiate immune responses; M cells transport sampled material to lymphoid tissue; and crypt stem cells continually replace the lining.
These categories describe the principal epithelial populations emphasized in mammalian biology. Exact lists vary depending on whether a source counts stem and progenitor cells, immune-interface cells such as M cells, or less common populations separately.

The crypt–villus layout that organizes the lining
The small-intestinal lining is not a flat sheet. It folds into fingerlike villi that project into the lumen, while the spaces between villi contain tubular invaginations called crypts of Lieberkühn. The National Institute of Diabetes and Digestive and Kidney Diseases described villi in 2017 as structures that increase internal surface area and help the gut absorb materials.

Villi are dominated by mature, post-mitotic cells that face digested food. Crypts contain the renewal machinery and specialized defense cells. New epithelial cells arise near a crypt base, pass through a transit-amplifying progenitor stage, differentiate as they migrate, and are eventually shed at a villus tip. In the mammalian intestine, the complete epithelial surface is reported to renew in approximately four to seven days, although the exact rate depends on species, location and physiological state.

Enterocytes: the main absorptive cells
Enterocytes are the most numerous cells on small-intestinal villi. Their apical surface carries dense microvilli, collectively called the brush border. Microvilli greatly expand the membrane area exposed to the lumen, giving the intestine more space for terminal digestion and transport.
At the brush border, enterocytes complete the final steps of breaking food components into absorbable forms. They then move nutrients across the epithelial cell and into the body. They also absorb ions and water, so their work affects both nutritional uptake and the fluid composition of the intestinal contents.

Enterocytes are highly polarized: the apical side faces the lumen, while the basolateral side communicates with underlying tissue and circulation. Tight epithelial junctions and the coordinated arrangement of neighboring cells allow useful molecules to cross while limiting uncontrolled passage between cells. As enterocytes migrate toward villus tips, they become mature absorptive cells; when their useful lifespan ends, they are shed and replaced by cells generated in crypts.
Goblet cells: building the mucus barrier
Goblet cells are interspersed among enterocytes and specialize in secreting mucins. When mucins mix with water and other secreted material, they form a mucus layer over the epithelial surface.
This layer creates physical separation between the epithelial membrane and much of the material in the lumen. It helps limit direct contact with digestive contents, particles and microorganisms while still allowing nutrients and chemical signals to reach the epithelial surface. Mucus is therefore a barrier system, not simply a lubricant.

Goblet cells occur along the crypt–villus epithelium and are replenished from the same stem-cell system that produces other epithelial lineages. Their output must be balanced: enough mucus to protect the surface, but not so much that it prevents normal exchange between the lumen and the epithelium.
Paneth cells: crypt defense and stem-cell support
Paneth cells are concentrated at the bases of intestinal crypts rather than on the exposed villus surface. They release antimicrobial peptides and proteins that help control interactions between the host and the resident microbial community.
Their position lets them protect the compartment where new epithelial cells are produced. Paneth-cell products can influence which microbes remain near the crypt and can help prevent damaging overgrowth close to stem and progenitor cells. This is local defense, distinct from the mucus barrier supplied by goblet cells.
Paneth cells also release factors that support neighboring intestinal stem and progenitor cells. In this way, they contribute to the stem-cell niche: the surrounding microenvironment that helps maintain renewal and appropriate differentiation. A 2012 review reported that Paneth cells in a healthy mouse persist for about two months. That lifespan is a mouse observation and should not be treated as a universal human value.
Enteroendocrine cells: turning luminal information into hormones
Enteroendocrine cells are chemosensory secretory cells. They detect nutrients and other signals in the intestinal lumen and respond by releasing gastrointestinal hormones.
Those hormones act locally or travel to other tissues to coordinate secretion, intestinal motility, digestion and appetite-related physiology. A meal can therefore change gut behavior before its nutrients have been fully absorbed: chemical information detected by enteroendocrine cells is converted into signals that adjust how the digestive tract handles its contents.

Enteroendocrine cells are relatively uncommon compared with enterocytes. Gerbe, Legraverend and Jay reported in 2012 that they account for approximately 1% of intestinal epithelial cells. The percentage is an approximate value from that review, not a fixed count for every intestinal segment, species or physiological condition.
Tuft cells: epithelial chemosensory sentinels
Tuft cells are rare epithelial cells with a sensory role. They detect chemical information in the lumen and can initiate type 2 and other immune responses.
Rather than serving primarily as absorptive or barrier cells, tuft cells act as an information-gathering interface. Recent reviews describe them as a link among epithelial sensing, immune responses, other epithelial cells and neuronal pathways. Their signals can help the tissue recognize a change in its luminal environment and recruit an appropriate response.
Because tuft cells are sparse, they do not contribute most of the intestine’s surface area or secreted material. Their importance comes from what they sense and the downstream circuits they activate. The exact responses depend on the stimulus and the surrounding immune and epithelial context.
M cells: sampling material for intestinal immunity
Microfold, or M, cells are specialized immune-interface cells found in follicle-associated epithelium, particularly over Peyer’s patches and related lymphoid tissue. They are not distributed uniformly across ordinary villus epithelium.
M cells capture particles, antigens and microorganisms from the lumen and transport them across the epithelial layer to immune cells in the underlying lymphoid tissue. This process, called transcytosis, gives the immune system access to information about material present in the gut without requiring widespread disruption of the epithelial barrier.

Their sampling function has two sides. It helps intestinal immune tissues monitor the lumen and develop responses, but the same pathway can be exploited by some pathogens to reach underlying tissue. M cells are therefore best understood as controlled gateways between luminal contents and organized immune surveillance.

Intestinal stem cells reside near crypt bases. They generate transit-amplifying progenitors, which divide rapidly and then differentiate into enterocytes, goblet cells, enteroendocrine cells, tuft cells and Paneth cells. Specialized epithelial populations such as M cells arise in follicle-associated regions under additional local signals.
This renewal system replaces cells lost through normal shedding and responds to injury or changing physiological demand. The crypt is consequently both a production site and a decision-making environment: signals from neighboring epithelial cells, Paneth cells, immune cells and the tissue beneath the epithelium influence whether descendants continue dividing or adopt a mature identity.
The often-cited four-to-seven-day renewal interval describes the approximate turnover of the intestinal epithelium in mammalian studies. It is not a promise that every cell type, every villus or every human intestine follows exactly the same clock.
How the cell types work as one system
From digestion to absorption
Food-derived molecules encounter the brush border of enterocytes after passing through the lumen. Enterocytes complete terminal digestion and absorb the resulting nutrients, along with ions and water. Their large microvillar surface makes this high-volume exchange possible.
Separating the lumen from living tissue
Goblet-cell mucus and epithelial junctions establish a protected interface. The barrier is selective rather than absolute: it keeps much luminal material from directly contacting cells while allowing controlled nutrient transport and signaling.
Best Value
Controlling microbes near the crypt
Paneth-cell antimicrobial products add a chemical defense at crypt bases, where stem and progenitor cells are maintained. This places microbial control alongside, rather than instead of, the broader mucus barrier.
Coordinating physiology and immunity
Enteroendocrine cells translate nutrient information into hormonal instructions for motility, secretion and appetite-related processes. Tuft cells detect chemical changes and connect epithelial sensing to immune and neuronal responses. M cells deliver selected luminal material to lymphoid tissue, where immune surveillance can occur.
Why different sources list different “main” cells
Many reviews emphasize five principal post-mitotic epithelial lineages: enterocytes, goblet cells, Paneth cells, enteroendocrine cells and tuft cells. That list focuses on mature cells produced by the crypt–villus program.
Quick wins for a faster PC:
Repair Windows errors before they cause bigger problemsFix Now →Fix the driver behind crashes, sound loss and screen glitchesFind Drivers →Clear out junk files and repair common Windows errorsFree Scan →Other classifications add M cells because they are important epithelial cells with a specialized immune-interface function. They may also count stem cells and transit-amplifying progenitors, even though these are renewal stages rather than mature villus populations. Less common populations, including cells sometimes called cup cells, are classified differently across studies. A list is therefore meaningful only when it states whether it is describing mature epithelial lineages, all epithelial populations, or the complete crypt environment.
Species and evidence limits
The functions above are based mainly on mammalian studies. General principles such as epithelial renewal, absorptive enterocytes, mucus-producing goblet cells and crypt-localized Paneth cells are broadly conserved, but cell proportions, marker definitions and timing can vary among species and intestinal regions.
Numbers need particular care. The approximately 1% estimate for enteroendocrine cells and the four-to-seven-day epithelial renewal interval are reported values from the cited literature, not universal constants. The approximately two-month Paneth-cell persistence figure was specifically reported for healthy mice. Human intestinal cell lifespans and proportions should not be inferred from that mouse measurement without direct human evidence.
