RNA-containing droplets are useful models for testing how compartmentalization might concentrate molecules, alter RNA behavior, and enable selected reactions. Their limit is fundamental: a droplet that performs one cell-like function is not a replica of an early cell, and it is not evidence that this chemistry produced the first cells.
Experiments show that coacervate and related membrane-free droplets can retain and exchange RNA, support some RNA catalysis, and grow through chemical turnover. They also reveal serious constraints: composition changes the physical environment, crowded interiors can impair ribozyme folding, nutrient access may be poor, and retention or growth does not demonstrate replication, heredity, or Darwinian evolution.

What an RNA droplet model actually represents
In these experiments, “RNA droplet” usually means a condensed, water-rich compartment rather than a lipid membrane. Coacervates form when oppositely charged polymers or other macromolecules separate from the surrounding solution into concentrated droplets. Related systems include aqueous two-phase compartments and droplets built from intrinsically disordered proteins (IDPs) mixed with RNA.

The attraction is straightforward. A dilute prebiotic soup may not allow molecules to meet often enough to react. A condensed phase can increase local concentration, hold some molecules together, and create a chemical environment different from the bulk solution. A review of RNA-containing membraneless compartments describes these systems as possible settings for origins-of-life chemistry, not as established historical intermediates. The 2020 review is therefore best read as a framework for testing mechanisms.

A model can answer a narrow question—such as whether RNA remains in a compartment long enough to encounter a catalyst—without answering the much larger question of how the first cells formed. The distinction between function and history is the key to interpreting every result.
What the experiments support
Concentration and compartmentalization
Droplets can organize RNA and other molecules into a smaller volume. This makes it plausible that a compartment could change reaction rates or protect selected molecules from immediate dilution. Such concentration is a proof of principle for one possible protocell function, not proof that the same chemistry existed on early Earth or that it was the route taken by life.
RNA retention and exchange
Experiments have measured how RNA oligonucleotides enter, remain in, and leave membrane-free compartments. The 2014 study of aqueous two-phase systems and coacervates directly examines rapid RNA exchange: Rapid RNA Exchange in Aqueous Two-Phase System and Coacervate Droplets. These measurements establish transport behavior under the tested formulations. They do not show that a droplet copies RNA, preserves a sequence across generations, or selects advantageous variants.
Selected RNA catalysis
Compartmentalized RNA catalysis has been demonstrated in membrane-free coacervate protocell systems. The 2018 study reports catalytic activity inside droplets while also identifying a problem that limits the result: highly charged, crowded interiors can cause simple ribozymes to adopt inactive conformations. The study’s results therefore support conditional catalysis, not a general claim that droplets improve every RNA reaction.
Growth driven by chemical turnover
Active droplets can grow when chemical reactions produce or supply more of the material that makes up the condensed phase. A nucleotide-based coacervate model reported composition-dependent growth and resistance to Ostwald ripening in 2021: Active coacervate droplets are protocells that grow and resist Ostwald ripening. A 2017 study likewise used active droplets to model growth and division linked to chemical activity: Growth and division of active droplets provides a model for protocells. These are important demonstrations of possible physical mechanisms. They are not records of an ancient population growing, reproducing, and evolving.

Limit 1: A droplet is not automatically a cell
Cells combine several functions in one integrated system: a boundary, selective transport, energy management, chemical maintenance, information storage, replication, and some route to reproduction with inherited variation. An RNA droplet normally supplies only a subset of these. It may concentrate a polymer or alter a reaction environment while lacking a stable boundary, controlled exchange, a genetic copying process, or a mechanism that passes molecular information to daughter compartments.
Calling a system a “protocell” describes its role as a model for possible early organization. It does not mean that the system meets a modern biological definition of a cell or that it is an ancestor of cells. The appropriate claim is that the droplet exhibits a cell-like function under specified laboratory conditions.
Limit 2: Composition determines the behavior
There is no single universal RNA droplet. The polymers, charges, concentrations, pH, salt, temperature, and fuel molecules determine whether a compartment forms and how it behaves. RNA is not merely cargo: it can change the physical properties of the condensed phase itself.
The 2020 review reports that adding RNA to IDP-based coacervates can reduce local protein concentration and decrease viscosity. A less viscous or less protein-rich interior may change diffusion, reaction encounters, and the stability of folded molecules. Consequently, a result obtained with one RNA sequence and one polymer recipe cannot be generalized to every putative early-Earth condensate. The review’s discussion of RNA-dependent material properties makes composition a first-order experimental variable, not a minor technical detail.
This also complicates comparisons between papers. Two studies may both use the word “coacervate” while testing compartments with different charge densities, water content, viscosities, or exchange rates. A favorable result in one formulation says little about another unless the physical conditions are comparable.

Limit 3: Retention is not replication, heredity, or evolution
Keeping RNA inside a droplet can help a reaction by preventing immediate dilution, but retention alone has no informational consequence. For heredity, a sequence must be copied with enough fidelity, copies must persist, and differences must affect survival or reproduction of compartments. A droplet that traps an RNA oligonucleotide has not demonstrated any of those steps.

Repair Windows errors before they cause bigger problemsFix Now →Scan for outdated or missing drivers - takes under a minuteDriver Scan →Exchange creates the opposite trade-off. If RNA leaves rapidly, a compartment may fail to maintain a useful catalyst. If exchange is too slow, substrates may not enter and products or waste may not leave. The measured exchange behavior in aqueous two-phase and coacervate systems therefore informs a transport question; it does not establish a complete life cycle.
The same caution applies to division. Physical splitting of a droplet can distribute material unevenly, but unequal partitioning is not equivalent to copying a genome and transmitting a heritable trait. To argue for Darwinian evolution, an experiment would need to connect molecular information, variation, differential persistence, and repeated inheritance—not just observe changing droplet sizes.
Limit 4: Crowding can undermine RNA function
Concentration is not always beneficial. RNA catalysts need to fold into particular three-dimensional structures. Electrostatic interactions and crowding in a coacervate can stabilize alternative structures that do not catalyze the intended reaction. The 2018 catalysis study explicitly discusses this risk for simple ribozymes. Its findings show why measuring catalytic activity is essential; assuming that all RNA remains functional inside a concentrated phase is not justified.
Activity can also depend on where a molecule resides. A catalyst may partition into the droplet while a required substrate stays outside, or both may enter but encounter inhibitory ions. Therefore, “RNA is present” and “RNA catalyzes a reaction” are separate experimental claims requiring separate measurements.

Limit 5: Nutrient access and sustained activity may be poor
A compartment that concentrates molecules can still starve if useful inputs cannot cross its interface. A review of protocells and RNA self-replication identifies limited access to nutrient sources as a major problem for aqueous emulsion droplets. The review notes that such droplets may remain valuable laboratory models for evolutionary phenomena even when they are incomplete cell models.
This limitation matters for any claim about sustained metabolism or replication. A one-time reaction in a prepared mixture is different from a system that continuously receives activated building blocks, removes waste, maintains energy supplies, and keeps catalysts active. Experiments should report transport and feeding conditions rather than treating the surrounding solution as an unlimited nutrient supply.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Limit 6: Growth is not a historical record
Active coacervates can increase in size through chemical turnover, and growth rates can vary with composition. Those observations demonstrate that a physical system can couple chemistry to compartment growth. They do not establish that the same mechanism operated in the relevant early-Earth environment.
Historical inference requires more than showing that a mechanism is chemically possible. Researchers would need evidence that the ingredients could form naturally in the proposed setting, remain stable under its environmental conditions, and interact with other necessary processes. The cited droplet studies present their systems as models of protocell behavior. They do not identify those droplets as the first cells.

How droplets compare with other protocell models
There is no single “best” protocell model. A useful comparison asks what each system can test and what it leaves out.

A practical checklist for interpreting a droplet claim
- Identify the material recipe. Record the RNA type, partner polymers, concentrations, charge balance, pH, salts, temperature, and any chemical fuel. Without these details, “RNA droplet” is too broad a category.
- Separate localization from function. Ask whether the experiment measured RNA partitioning only, or also measured folding, catalysis, product formation, and turnover.
- Check transport in both directions. A useful compartment must retain relevant molecules while allowing substrates in and products or waste out. A retention result alone is incomplete.
- Look for controls outside the droplet. Compare the reaction with the same RNA and substrates in the surrounding solution. Concentration can improve one step while inhibiting another.
- Distinguish growth from reproduction. Growth through chemical accumulation, physical splitting, sequence copying, and heritable selection are different milestones and should not be reported as synonyms.
- Test environmental plausibility separately. A laboratory demonstration establishes possibility under its recipe. It does not establish availability of those ingredients, conditions, or energy sources on early Earth.
- Ask what remains missing. A strong paper states whether it addresses compartment formation, transport, catalysis, metabolism, replication, or evolution—and avoids extending the conclusion beyond that function.
What these models cannot yet explain
The cited work does not resolve how long RNA polymers first formed, how functional ribozymes arose, or how a chemical compartment acquired a coupled system of replication, metabolism, and inheritance. The 2020 review describes nonenzymatic RNA polymerization as an active research area rather than a solved step. That unresolved status is important: a compartment can help organize molecules after they exist without explaining their initial origin.
Nor do these sources settle the historical competition between membraneless droplets, lipid-bound vesicles, mineral surfaces, or other protocell proposals. The meaningful comparison is functional and conditional: which model provides the required chemistry, transport, stability, and inheritance under a specified environment? Evidence that one droplet performs a task does not eliminate the need for other compartments or establish a unique pathway.
Bottom line for readers
RNA droplets are powerful reductionist tools. They let researchers isolate questions about concentration, exchange, catalytic activity, and chemically driven growth in compartments without membranes. Their limits are equally instructive: the physical recipe is highly specific, crowding can disable ribozymes, nutrient access may fail, and neither RNA retention nor droplet growth supplies genetic replication or heredity.
