A 2013 study reported that an experimental iron-containing molecule called 1-Fe both acted as a catalytic antioxidant and interfered with cholesterol synthesis in preclinical research. It did not show that the molecule prevents heart attacks, treats heart disease in people, or works better than statins.
What is 1-Fe?
1-Fe is an iron(III) complex of an amphipolar corrole, a type of molecule investigated for its antioxidant properties. In a 2013 Chemical Communications paper, researchers from Technion – Israel Institute of Technology examined its antioxidant activity and its effects on cholesterol-related processes. What are the two reported actions?
Catalytic antioxidant activity
The researchers described 1-Fe as a catalytic antioxidant: it was proposed to eliminate free radicals through catalytic action. In the contemporaneous report, corresponding author Zeev Gross said, “Our antioxidant eliminates free radicals in a catalytic fashion.” This describes the proposed molecular activity, not a demonstrated reduction in cardiovascular disease.
Interference with cholesterol synthesis
The study also reported that 1-Fe and a non-redox-active analogue allosterically inhibited HMG-CoA reductase, an enzyme involved in cholesterol synthesis. In addition, macrophages harvested from mice treated with 1-Fe had reduced cholesterol-biosynthesis ability. These are biochemical and animal-research findings, not evidence of a treatment effect in people. The RSC paper’s abstract describes these results.

How does the proposed mechanism differ from statins?
The 2013 report characterized statins as competitive inhibitors of HMG-CoA reductase, while the corroles were reported to inhibit the same enzyme allosterically—that is, by interacting with the enzyme in a different way. Gross described it as “a new mode of action.” This is a comparison of proposed mechanisms only; the study did not establish that 1-Fe is more effective, safer, or otherwise clinically superior to statins. What the research does—and does not—show
- Reported: antioxidant activity attributed to 1-Fe, allosteric inhibition of HMG-CoA reductase by 1-Fe and a non-redox-active analogue, and reduced cholesterol-biosynthesis ability in macrophages harvested from treated mice.
- Not established: prevention of heart attacks, treatment of cardiovascular disease in people, reduced cardiovascular events, clinical safety or efficacy, or a basis for replacing prescribed statins.
The authors’ findings were published in 2013. Chemistry World reported that further work in other cholesterol-related diseases was planned at the time; the sources cited here do not establish whether 1-Fe later advanced to human testing or what its current development or approval status is. The early description of it as a possible statin alternative should therefore be understood as a research prospect, not a clinical conclusion. Chemistry World’s 2013 report covers that context.

Why the headline needs a qualification
“Two fronts” refers to the two proposed lines of action—antioxidant activity and interference with cholesterol synthesis. The evidence supports an experimental molecular finding, not a demonstrated way to fight heart disease in patients. There is no numerical clinical effect or patient-outcome result in the cited study and report.
