Vitamin D2 vs. D3: Which Supplement Is Right for You?

Vitamin D2 (ergocalciferol) and vitamin D3 (cholecalciferol) both raise blood vitamin D. The NIH Office of Dietary Supplements says most evidence finds that D3 raises serum 25-hydroxyvitamin D (25(OH)D) more and keeps levels higher for longer than D2. That difference does not make D3 the universally correct choice, however. The right form depends on whether you are treating a diagnosed deficiency, considering routine prevention, or choosing ingredients that fit your dietary preferences.

Current guidance also has a narrower scope than many supplement claims suggest. The Endocrine Society’s 2011 deficiency guideline accepted either form for treating or preventing deficiency. Its 2024 guideline concerns disease prevention in people without an established indication for vitamin D treatment or 25(OH)D testing, and gives population-specific—not universal—recommendations.

What vitamin D2 and D3 have in common

Vitamin D2 and D3 are two forms of vitamin D used in supplements and clinical care. D2 is the chemical name for ergocalciferol; D3 is cholecalciferol. Both can increase serum 25(OH)D, the usual blood marker used to assess vitamin D status.

That shared effect matters because a supplement label, a laboratory result and a treatment plan are not separate decisions. A higher 25(OH)D response may be useful in one setting, while ingredient sourcing or an established medical indication may matter more in another. The form should therefore be chosen in context rather than by treating a single biochemical difference as a universal health verdict.

What the blood marker does—and does not—tell you

Serum 25(OH)D is the marker discussed in the major guidance documents. It is a way to evaluate vitamin D status, not a complete measure of every health outcome. The Endocrine Society’s 2024 prevention guideline specifically notes that routine testing is not supported in the populations it considered when outcome-specific benefits based on a measured level have not been identified. That statement does not cancel testing or treatment ordered for an established clinical indication.

D2 versus D3 at a glance

Question Vitamin D2 Vitamin D3 What it means for a decision
Scientific name Ergocalciferol Cholecalciferol Both names may appear on a supplement label or prescription.
Raises serum 25(OH)D? Yes Yes Both forms can increase the usual blood marker of vitamin D status.
Relative response in most evidence summarized by NIH ODS Generally produces a smaller or less persistent rise than D3 Generally raises 25(OH)D more and maintains higher levels longer This is a biochemical difference, not proof that D3 is best for every person or outcome.
Deficiency guidance Accepted as an option in the Endocrine Society’s 2011 deficiency guideline Accepted as an option in the same 2011 guideline A clinician-directed deficiency plan may use either form.
Typical source described by NIH ODS Ergosterol in yeast, exposed to ultraviolet light 7-dehydrocholesterol from lanolin obtained from sheep’s wool, exposed to ultraviolet light Source and processing can matter for dietary, ethical or religious reasons.
Animal-free option Not established in the supplied evidence Lichen-derived D3 is available Ask the manufacturer how the ingredient and capsule or tablet are sourced and processed.

Why D3 often produces a stronger, longer-lasting blood response

The NIH Office of Dietary Supplements summarizes the evidence this way: both forms raise serum 25(OH)D, but most evidence indicates that D3 raises the measure to a greater extent and maintains higher levels for longer than D2. The practical implication is that two products with the same stated vitamin D amount may not produce identical 25(OH)D patterns over time.

“Higher” and “longer-lasting” describe the measured blood response. They do not automatically establish better protection against every disease, nor do they determine what an individual should take. The Endocrine Society’s prevention recommendations are based on populations and outcomes, not simply on choosing the form that tends to produce the larger laboratory increase.

What the evidence does establish about rickets

The NIH fact sheet states: “Both vitamins D2 and D3 raise serum 25(OH)D levels, and they seem to have equivalent ability to cure rickets.” This is a specific statement about rickets. It should not be expanded into a claim that D2 and D3 are identical for all conditions, doses, blood-level patterns or long-term outcomes.

The crucial distinction: prevention versus treatment

Before choosing a form, identify the question you are actually trying to answer. “Which supplement should I buy for routine prevention?” is not the same as “Which prescription or supplement regimen should treat my diagnosed deficiency?” The two questions are covered by different guidance.

Clinician-directed treatment of diagnosed deficiency

The Endocrine Society’s 2011 guideline for vitamin D deficiency suggested using either D2 or D3 for treatment and prevention of deficiency. That historical recommendation means the form alone does not invalidate a clinician’s treatment plan. If a clinician has diagnosed deficiency, follow the prescribed or recommended regimen and ask whether changing forms would alter the intended monitoring plan.

A diagnosed deficiency, a disorder affecting absorption or metabolism, or another established indication can require individualized decisions about form, amount, timing and follow-up. The supplied guidance does not establish one personal dose, and it does not replace medical assessment.

Routine disease prevention without an established indication

The Endocrine Society’s 2024 guideline has a different scope. It addresses people without an established indication for vitamin D treatment or 25(OH)D testing and asks whether empiric supplementation can prevent disease in specified populations. It is not a universal instruction to take high-dose vitamin D, and it does not replace dietary reference intakes.

The guideline suggests against empiric supplementation above current reference dietary intakes to lower disease risk in healthy adults younger than 75. For this group, taking more simply because D3 tends to raise the blood marker more is not supported by that recommendation.

Who the 2024 prevention guideline specifically discusses

The 2024 Endocrine Society guideline suggests empiric vitamin D supplementation for several populations because of potential population-specific benefits. These are prevention recommendations for the groups considered, not individualized treatment orders.

  • Children and adolescents aged 1–18. The recommendation covers this age range as a population, while a child’s medical history, diet and existing treatment still determine what a clinician may advise.
  • Adults older than 75. The guideline identifies this age group for empiric supplementation in prevention settings.
  • Pregnant people. Pregnancy is included among the populations for which empiric supplementation is suggested.
  • People with high-risk prediabetes. This is a defined risk category, not a blanket recommendation for everyone with a mildly abnormal glucose result.

The guideline says optimal empiric doses remain unclear for the populations it considered. That uncertainty is important: the recommendation to consider supplementation does not supply a single dose that can safely be applied to every person in a listed group.

When testing makes sense—and when not to assume it is needed

The 2024 guideline suggests against routine 25(OH)D testing in the prevention populations it considered when no outcome-specific benefit based on a measured level has been established. In other words, a test is not automatically required before every prevention decision.

That is not permission to disregard testing when a qualified clinician has identified an established indication. Symptoms, a diagnosed deficiency, a condition affecting vitamin D handling, a medication issue or a treatment-monitoring plan can change the clinical question. Ask why a test is being ordered, what result would change management and when follow-up would occur.

Questions to ask about a proposed test

  • Is the test for a diagnosed deficiency or another established indication, or is it routine screening?
  • What treatment decision would change if the result were low, adequate or high?
  • Will the result be interpreted alongside medical history, diet, medications and conditions that affect absorption or metabolism?
  • When should the result be rechecked, if rechecking is clinically necessary?

Ingredient source and dietary preference

The two forms can also differ in how their active ingredients are made. NIH ODS says D2 is manufactured by exposing ergosterol in yeast to ultraviolet light. D3 is typically produced by exposing 7-dehydrocholesterol from lanolin obtained from sheep’s wool to ultraviolet light.

People who avoid animal-sourced ingredients should not rely on the “D3” name alone. NIH ODS notes that animal-free D3 sourced from lichen is available, but the finished product may also contain other ingredients that matter to a person’s dietary, ethical or religious requirements. Check both the vitamin source and the capsule, tablet or liquid formulation.

How to verify a lichen-derived product

  1. Look for an explicit statement that the D3 is sourced from lichen rather than lanolin.
  2. Check whether the manufacturer identifies the source and processing of the finished product, not only the active vitamin.
  3. Review capsule and tablet ingredients for materials that conflict with your requirements.
  4. If the label is ambiguous, ask the manufacturer before purchasing or ask a pharmacist to help interpret it.

Lichen-derived D3 is a sourcing option, not evidence that the form is medically necessary for everyone who follows a plant-based diet.

A practical decision framework

Use the following sequence to keep the choice tied to the question you need answered.

  1. Define the purpose. Write down whether you are following a clinician-directed plan for diagnosed deficiency, considering prevention, or trying to meet a dietary preference. Do not treat these as interchangeable goals.
  2. Check for an established indication. If a clinician has identified deficiency or a condition affecting vitamin D, discuss the regimen with that clinician before switching forms or changing the amount.
  3. Place yourself in the 2024 guideline’s scope. The prevention recommendations name children and adolescents aged 1–18, adults older than 75, pregnant people and people with high-risk prediabetes. Healthy adults younger than 75 are not advised to use empiric supplementation above current reference dietary intakes to lower disease risk.
  4. Compare the blood-response consideration. If the decision is otherwise open, recognize that most evidence summarized by NIH ODS favors D3 for a greater and more sustained 25(OH)D rise. Treat that as one factor, not a universal outcome claim.
  5. Check sourcing. If avoiding animal-sourced ingredients matters, look for verified lichen-derived D3 or another form that meets your requirements. Confirm the whole formulation.
  6. Agree on monitoring. Ask whether testing is indicated, what result would change the plan and how follow-up will be handled. Do not order repeated tests simply to chase a number without a clinical reason.

Common mistakes when comparing D2 and D3

Mistake 1: Treating “D3 is stronger” as a universal recommendation

The relative 25(OH)D response is real in the evidence summarized by NIH ODS, but it does not answer every clinical question. A stronger laboratory response is not the same as proof of better prevention for every disease or proof that D2 is inappropriate.

Mistake 2: Applying the 2024 prevention guideline to deficiency treatment

The 2024 guideline concerns people without an established indication for treatment or testing. The 2011 deficiency guideline addressed treatment and accepted either D2 or D3. Mixing these scopes can lead to inappropriate self-treatment or to stopping a clinician-directed plan.

Mistake 3: Assuming a recommendation supplies a universal dose

The 2024 guideline says optimal empiric doses remain unclear for the populations it considered. A population-level suggestion to supplement is therefore not a personal dosing instruction.

  • Gluten Free Vitamins: Sourced from high-quality ingredients, this adult Vitamin D supplement is gluten free and has no color added and no artificial flavors

Mistake 4: Using routine testing as a substitute for clinical reasoning

A test can be useful when it answers a defined clinical question, but the 2024 guideline does not support routine 25(OH)D testing across its prevention populations when level-based outcome benefits have not been identified. Testing and treatment decisions should remain connected to the indication.

Mistake 5: Calling every D3 product vegan

Typical D3 is made from lanolin, although lichen-derived animal-free D3 is available. Verify the source rather than inferring it from the D3 label.

How to read a supplement label without overinterpreting it

  • Identify the form: the label should say ergocalciferol (D2) or cholecalciferol (D3).
  • Look for source information: especially if lanolin, lichen or other animal-free requirements matter to you.
  • Separate form from amount: the evidence here does not establish a dose that is right for all readers.
  • Check the complete ingredient list: the active vitamin is only one part of a capsule, tablet or liquid.
  • Match the label to the plan: if a clinician specified a form or regimen, confirm that the product actually uses it.

A label can tell you what a product contains; it cannot determine whether supplementation is indicated or whether a blood test is needed.

Situations that call for a clinician conversation

Seek qualified clinical advice before making a change when you have a diagnosed vitamin D deficiency, are pregnant, are choosing supplementation for a child or adolescent, are older than 75, have high-risk prediabetes, or have a condition or treatment that could affect vitamin D absorption or metabolism. These categories overlap with the prevention guideline or with the kinds of established indications that the guideline does not replace.

Bring the product label, the reason you are considering supplementation, any previous 25(OH)D results and the name of the form you are currently using. Ask whether D2 or D3 is intended, whether the amount is individualized, and whether follow-up testing has a defined purpose.

What can be concluded—and what cannot

Three conclusions are well supported. First, D2 and D3 both raise serum 25(OH)D. Second, most evidence summarized by NIH ODS finds that D3 raises the marker more and maintains higher levels longer. Third, the 2011 deficiency guideline accepted either form, while the 2024 prevention guideline makes population-specific recommendations and says optimal empiric doses remain unclear.

Several broader claims are not established by this evidence. It does not identify a personal dose, rank brands, prove that D3 prevents every disease better than D2, or show that everyone needs a supplement or a 25(OH)D test. It also does not make lanolin-derived D3 unsuitable for every person or lichen-derived D3 necessary for every plant-based eater; those are sourcing decisions to verify against individual preferences.

Bottom line

D3 is usually the stronger choice if your only comparison is the typical serum 25(OH)D response, because it generally raises the marker higher and keeps it elevated longer. D2 remains a legitimate option, including in the Endocrine Society’s 2011 deficiency guidance, and both forms have equivalent ability to cure rickets according to the NIH fact sheet. For prevention, use the Endocrine Society’s 2024 population-specific scope rather than assuming a high-dose or routine-testing recommendation. For diagnosed deficiency, pregnancy, childhood, older age, high-risk prediabetes or a sourcing requirement, make the form and regimen part of a clinician-guided decision.
