About 43% of the families do not appear in the Pfam database, and 33 families occur in eukaryotic pathogens but not in humans. Those pathogen-specific proteins could eventually guide selective drug research, while the broader comparison adds evidence to the unresolved question of how mitochondria evolved.
What the atlas compared
The project assembled mitochondrial protein catalogues from six newly analysed organisms: five parasites and one plant. Researchers then compared those catalogues with known human and yeast mitoproteomes. A mitoproteome is the full set of proteins associated with an organism’s mitochondria or mitochondrion-like organelles.

| Measure | Finding | What it indicates |
|---|---|---|
| Combined mitochondrial proteins | 8,619 | The total catalogue assembled from the newly analysed organisms plus human and yeast references |
| Protein families | 3,199 | Groups of related mitochondrial proteins across the compared species |
| Families absent from Pfam | About 43% | Many proteins lack a match in a widely used protein-family resource, so their biology is still poorly described |
| Pathogen-conserved families absent from humans | 33 | Possible selective-target leads, not validated medicines |
The Pfam result does not mean that 43% of the proteins were proven to be entirely new molecules. It means that roughly 43% of the identified families had no representation in that database, leaving their evolutionary relationships or functions unresolved.

What was found in Giardia lamblia
Giardia lamblia received particular attention because it lives in the oxygen-poor human gut and relies on mitochondrion-like organelles rather than the textbook mitochondria found in oxygen-using cells. These reduced organelles are difficult to investigate, yet they still perform essential chemistry.
The atlas identified 59 mitochondrial proteins in Giardia. Of those, 32 corresponded to previously recognized proteins and 27 were newly identified in the atlas. Researchers could not assign a function to 34 of the 59 proteins; the categories overlap because some of the newly identified proteins are also the ones whose roles remain unknown.

Nature’s report cites an estimate of about 280 million Giardia infections worldwide each year. That public-health burden makes basic knowledge of the parasite’s unusual organelles relevant even before any drug candidate exists.

It could provide starting points, but it is not evidence that a malaria or sleeping-sickness treatment has been discovered. The strongest medical idea is selectivity: a protein family present in a pathogen but absent from humans may be easier to inhibit without directly blocking a human counterpart.

Mitochondrial biologist Vamsi Mootha described the pathogen-specific families as potentially “ideal drug targets” because a therapy could act on parasite proteins rather than human ones. That is a target-discovery hypothesis. Each candidate would still need to pass several tests:
- demonstrate that the protein is essential to the parasite;
- determine its three-dimensional structure and active site, if applicable;
- show that a chemical or biological inhibitor can reach the parasite organelle;
- measure selectivity against human proteins and cells;
- test efficacy and toxicity in animal models; and
- complete human clinical trials before any treatment claim is justified.
The atlas therefore narrows the search space. It does not show that any of the 33 families already has a working inhibitor, is safe, or has entered clinical testing.

Why parasite mitochondria matter
Parasites have adapted mitochondrial systems to very different environments and lifestyles. A parasite living in an oxygen-limited host compartment may retain only a reduced, specialized organelle, while another lineage may preserve a more conventional mitochondrion. Comparing those states can expose biochemical pathways that are invisible in human cells.
That contrast also helps separate universal mitochondrial machinery from lineage-specific additions. Proteins shared by parasites, plants, yeast and humans are more likely to represent deeply conserved functions. Proteins restricted to pathogens may reflect adaptations that parasites need but people do not.

What the atlas says about mitochondrial origins
Mitochondria retain small genomes distinct from nuclear DNA, although mitochondrial proteins are encoded by both genomes. The broad explanation accepted by cell biology is endosymbiosis: roughly two billion years ago, an archaeal cell engulfed a bacterium, and the bacterium became a permanent resident that evolved into the mitochondrion.
The new atlas does not overturn that model. Its value is that it compares mitochondrial proteins across widely separated eukaryotic lineages, including organisms with highly reduced mitochondrion-like organelles. Such comparisons can help researchers distinguish proteins inherited from the original bacterial partner from proteins added later by different host lineages.
They also reopen questions about timing and mechanism. If a protein family appears in some distant eukaryotes but not others, scientists must consider whether it was lost repeatedly, evolved independently, or was present in an ancestral organelle and became difficult to detect. The atlas supplies a much larger set of sequences for testing those possibilities; it does not by itself establish a single revised origin story.
Cell biologist Pavel Doležál summarized the significance of the uncharacterized proteins this way: “it means that there is some new metabolism that we don’t know about. And it was overlooked”.
How to read the numbers
- 8,619 proteins is the size of the combined catalogue, not the number found in one organism.
- 3,199 families groups related proteins, so one family can contain members from multiple species.
- 43% absent from Pfam describes database coverage, not a measured percentage of proteins with no biological function.
- 33 pathogen-specific families identifies a possible selectivity advantage, not a list of approved or tested drug targets.
- 59 Giardia proteins includes both known and newly identified proteins, with unknown-function assignments overlapping the new-protein count.
Taken together, the atlas is both a reference resource and a set of hypotheses. It gives evolutionary biologists a broader map of mitochondrial diversity and gives infectious-disease researchers a prioritized list of parasite proteins that may deserve laboratory validation.
