The human superior colliculus—a layered midbrain structure only about 6 millimeters across in each hemisphere—shows which kind of sensation is expected before that sensation arrives, according to a Nature Neuroscience study published October 7, 2026. Using ultra-high-field 7-Tesla fMRI, researchers detected depth-specific activity linked to an upcoming visual or tactile event, extending evidence for predictive processing beyond the cerebral cortex.
What the superior colliculus does
The superior colliculus sits in the midbrain and is best known for integrating sensory information with orienting and motor commands. It contains layered maps rather than functioning as a single undifferentiated relay. The new findings indicate that those layers also participate in preparing for an expected sensation.

Its small size makes the structure difficult to study with conventional imaging. The study reports approximately 1-millimeter spatial resolution with 7-Tesla fMRI, compared with roughly 2–3 millimeters for standard 3-Tesla fMRI. That scale matters when the target itself is only about 6 millimeters in diameter per hemisphere.
How the experiment tested prediction
Participants and sensory tasks
Danlei Chen and colleagues studied 80 adults: 40 in a visual-stimulation group and 40 in a somatosensory group. Visual participants viewed normed affective images. The somatosensory group received mechanical pressure on the left thumb.

Each trial included a decision period before stimulation. Participants viewed shapes and either selected one themselves or let a computer select it. This design allowed the researchers to compare expected sensory modality while keeping immediate decision and motor requirements similar across groups.
Layer-resolved imaging
The researchers mapped activity by depth within the superior colliculus during the decision period and during the subsequent stimulus. They then tested whether spatial activity patterns could identify the expected modality using leave-one-subject-out support-vector-machine classifiers.
What changed across the superior colliculus
Visual and tactile input occupied different depths
During actual stimulation, visual input produced stronger responses in superficial layers, whereas thumb pressure produced stronger responses in deeper layers. The same modality-linked depth organization appeared during the pre-stimulus decision period, before the image or pressure had been delivered.

In practical terms, the superior colliculus did not merely react after the sensory event. Its activity already reflected whether the upcoming event belonged to the visual or touch condition.
Prediction resembled reaction
The anticipatory maps resembled the maps measured during stimulation, although they were not identical. This correspondence supports the idea that the brain prepares using modality-specific patterns rather than generating only a generic alertness signal.
How strong was the decoding evidence?
The study used multivariate classification, which evaluates the joint spatial pattern across measured locations rather than asking whether one location simply became more active. All reported results were above chance with P<0.001.

| Training maps | Test maps | Mean accuracy | Standard error of the mean |
|---|---|---|---|

The cross-condition results are important: classifiers trained on anticipatory patterns could classify stimulation patterns, and classifiers trained on stimulation patterns could classify anticipatory patterns. That transfer is consistent with shared modality structure across expectation and sensation, while the lower anticipatory accuracies show that prediction is weaker and noisier than the response to the delivered stimulus.

Related predictive and reactive effects appeared in the lateral geniculate nucleus, which relays visual information, and the ventral posterolateral nucleus, which relays somatosensory information. Together with the superior-colliculus results, these observations place anticipatory coding in several subcortical structures that pass information toward the cortex.
The authors also reported controls for cross-trial carryover. Those controls are designed to reduce the possibility that the apparent pre-stimulus signal was simply leftover activity from the preceding trial.

Why this matters for predictive processing
Predictive-processing theories propose that the brain continually estimates what is likely to happen and prepares its systems accordingly. First author Danlei Chen described the study as evidence that the brain “doesn’t just wait for sensory input and react to it,” but prepares for what is coming.
Much predictive-processing research has focused on cortical areas. This experiment suggests that deeper structures involved in sensory relaying and orienting also participate in that preparation. Because the superior colliculus maps both sensory experiences and motor commands, anticipatory activity there could help coordinate perception with rapid orienting responses.

What the study does not show
- It does not demonstrate that people consciously knew exactly what they were about to see or feel.
- It does not show that every individual sensation can be predicted reliably. The decoding results are group-level evidence from a controlled experiment.
- It is not a diagnostic test, treatment, or clinical outcome study.
- It does not establish that the superior colliculus alone generates predictions; the observed effects occurred within a broader network that included thalamic relay nuclei and cortical systems.
- Because the experiment compared visual images with pressure on one thumb, the results should not automatically be generalized to every visual or tactile experience, or to other sensory modalities.
Publication and study context
The paper, “Modality-specific reactive and predictive responses in the human superior colliculus,” was published in Nature Neuroscience on October 7, 2026. A reader-facing account appeared in Medical Xpress on October 16, 2026. The central contribution is methodological as well as theoretical: high-resolution, depth-sensitive imaging made it possible to examine a tiny midbrain structure and test whether its anticipatory pattern carried information about the expected modality.

Frequently Asked Questions
Does the brain prepare for a sensation before it happens?
In this controlled experiment, activity in the superior colliculus and related thalamic nuclei differed by the expected visual or tactile modality before stimulation began. That is evidence of neural anticipation, not proof of conscious foreknowledge of every sensation.
Why was 7-Tesla fMRI necessary?
The superior colliculus is only about 6 millimeters across per hemisphere. The study reports roughly 1-millimeter resolution at 7 Tesla, finer than the approximately 2–3-millimeter resolution typical of 3-Tesla fMRI, allowing activity to be examined by depth.
What is the difference between the superficial and deep layers?
Visual stimulation produced stronger superficial-layer responses, while thumb-pressure stimulation produced stronger deep-layer responses. The same depth pattern was present before stimulation during the decision period.
