Resistance exercise grows muscle primarily by creating mechanical tension. When a muscle produces force against an external load, structures inside and around its fibers detect deformation, convert it into biochemical signals, and increase the machinery available for making and remodeling protein. Repeated bouts can therefore enlarge muscle fibers, although the exact contribution of each sensor and supporting cell depends on the training context.
Mechanical tension is the initiating signal
During lifting or other resistance exercise, muscle fibers must generate force while their internal structures and connective attachments are loaded. That force is the key external stimulus for hypertrophy. The 2023 American Physiological Society review of mechanical-overload hypertrophy reports evidence for increased mTORC1 signaling, ribosome biogenesis, satellite-cell abundance, myonuclear accretion and post-exercise muscle-protein synthesis after overload.

This does not mean that every sensation associated with training has equal importance. A temporary pump, soreness or an acute hormone change can accompany a workout, but the reviews do not establish any of those effects as a guarantee of long-term hypertrophy. The durable signal is repeated, recoverable mechanical loading that the muscle can adapt to.
How muscle cells detect force
Mechanotransduction is the conversion of a physical force into intracellular information. A muscle fiber is not a simple bag of contractile proteins: it is connected to its surrounding matrix, neighboring structures and internal scaffolding. When those connections are stretched or placed under tension, they can alter the activity of signaling proteins.

Integrins and focal adhesions
Integrins connect the extracellular matrix to the cell’s cytoskeleton. At focal adhesions, force can change the conformation or activity of associated proteins, providing a route from external load to signals that regulate growth and remodeling.
Titin and the sarcomere
Titin is a giant elastic protein that spans each sarcomere. Because it experiences changes in length and tension during contraction, it is a plausible mechanical sensor and signaling platform. Its exact role in initiating human hypertrophy is still being refined.
Filamin-C and BAG3-associated complexes
Filamin-C and BAG3-associated structures help organize the cytoskeleton and respond to mechanical stress. A 2018 review describes their possible regulation of mTORC1, Hippo signaling and autophagy as plausible, but incompletely characterized. That qualification matters: scientists can identify candidate sensors without having established a single master switch or a fixed hierarchy among them.

From force to a growth program
Once mechanical information is detected, a network of pathways changes the cell’s priorities. The 2025 review by Van Every, Lees, Wilson, Nippard and Phillips summarizes the central idea in its abstract: “Mechanical tension is widely recognized as the primary stimulus underlying the molecular mechanisms that influence muscle hypertrophy induced by resistance training.”
mTORC1 and protein-building capacity
mTORC1 is a major coordinator of anabolic signaling. Its activation after overload increases the cell’s capacity to initiate and sustain translation, the process that assembles amino acids into new proteins. This contributes to the post-exercise rise in muscle-protein synthesis, but a single elevated response is not the same as permanent growth; enlargement reflects the accumulated result of many bouts and the remodeling that follows them.
Ribosome biogenesis
Ribosomes are the molecular machines that translate messenger RNA into protein. Mechanical overload can stimulate ribosome biogenesis, increasing translational capacity rather than merely switching existing ribosomes on. More capacity helps a repeatedly trained fiber manufacture the contractile and structural proteins needed to support a larger cell.

Remodeling, autophagy and Hippo-related signals
Growth is also a quality-control problem. Damaged or misassembled components must be removed and replaced while the cytoskeleton and extracellular attachments are reinforced. Autophagy-related processes help manage cellular components, while Hippo-related signaling participates in controlling growth. The precise interaction among these pathways remains an active area of study.
Epigenetic regulation
The 2025 synthesis also identifies epigenetic regulation as part of the hypertrophic response. Mechanical loading can influence how readily particular genes are expressed, helping a fiber maintain adaptations across repeated training exposures. Epigenetic effects complement, rather than replace, the immediate signaling events that follow a loaded contraction.
How signaling becomes a larger muscle fiber
Each workout can raise muscle-protein synthesis and trigger structural repair. If synthesis and remodeling repeatedly exceed breakdown over an appropriate recovery period, the fiber accumulates additional contractile and supporting material and becomes larger.

Satellite cells and myonuclei
Satellite cells are muscle-resident stem cells that can activate, proliferate and fuse with existing fibers. Fusion adds myonuclei, which may support the transcriptional demands of an expanding fiber. Reviews from 2023 and 2025 report increased satellite-cell abundance and myonuclear accretion after overload, but they do not support a universal claim that satellite cells are always required, or that they contribute on an identical timetable in every person and training condition.
Why timing and context matter
Muscle size is influenced by training history, the amount and progression of loading, nutrition, recovery and the specific muscle being trained. A cellular event observed after an unfamiliar overload may not have the same importance after years of training. This is why mechanistic findings explain how adaptation is possible without proving that one pathway or cell type alone determines the final result.

| Training axis | What to evaluate | Why it matters mechanistically |
|---|---|---|
| Amount and progression of tension | Can the exercise impose meaningful resistance and be progressed as it becomes easier? | Repeated overload is the input that keeps mechanotransduction and remodeling relevant. |
| Range of motion and muscle length under load | Which portions of the movement are loaded, and does the target muscle work through a controllable range? | Force and deformation vary with joint position and muscle length. |
| Recoverable volume and frequency | Can you perform enough hard work to stimulate adaptation while recovering before the next demanding session? | Growth requires repeated synthesis and remodeling, not an isolated stress that cannot be repeated. |
| Consistency of loading | Can resistance, technique and progression be reproduced from session to session? | Consistent exposure makes the accumulated adaptive signal more meaningful. |
A practical way to apply the science
- Select a resistance movement for the target muscle. Use a load that requires substantial force while allowing controlled technique.
- Establish a repeatable range of motion. Keep the movement consistent so that changes in performance reflect adaptation rather than a shorter or easier path.
- Progress the mechanical challenge. Add resistance, repetitions, leverage difficulty or another measurable demand when the current workload no longer provides a challenging stimulus.
- Distribute work you can recover from. Organize sessions so the target muscle receives repeated exposure without making fatigue so high that technique and subsequent training deteriorate.
- Track performance over time. A log of load, repetitions, range of motion and perceived effort helps verify that mechanical tension is actually progressing.
Where bands and adjustable dumbbells fit
Resistance bands and adjustable dumbbells are reasonable categories for applying these principles. Both provide external resistance, allow exercise selection for different muscles and can be progressed in practical ways. That is a functional inference from the resistance-training mechanism, not evidence that either category is biologically superior to barbells, machines or other tools.
Choose equipment by asking whether it can provide a stable, measurable and progressively harder stimulus through the range of motion you intend to train. Availability, price, durability and exact loading options vary by product and region; those purchasing details are separate from the cellular mechanism.
What the current evidence does not prove
- There is no established single “mechanosensor” that explains every hypertrophic response. Integrins, focal adhesions, titin and filamin-C/BAG3 complexes are candidate structures whose relative roles are still being mapped.
- An acute increase in muscle-protein synthesis does not by itself prove a particular routine will produce the most long-term growth.
- A pump, soreness or a short-lived hormonal response is not established as a reliable guarantee of hypertrophy.
- The reviews do not identify one universal repetition scheme, frequency, exercise, device or program as best for everyone.
- Satellite-cell activation and added myonuclei are important parts of the observed response in some contexts, but their exact necessity and timing are context-dependent.
Why this matters for interpreting fitness claims
Mechanical tension gives a useful test for evaluating training advice: does the recommendation help a muscle experience a controllable, progressive and recoverable load? If it does, it is consistent with the established initiating stimulus. Claims that rely only on a pump, a named supplement, an acute hormone spike or a particular branded device go beyond what these mechanistic reviews establish.
The overall sequence is therefore force, sensing, signaling, increased protein-building capacity and remodeling, followed by accumulated structural change. Understanding that sequence helps separate the part of training that drives adaptation from the sensations and marketing claims that merely accompany it.
