Use dead-end filtration when your main job is straightforward pass-through filtration and the feed will not rapidly block the membrane. Use tangential-flow filtration (TFF) when you must retain a target while concentrating it, exchanging buffer, removing smaller solutes, fractionating by size, or processing a feed that would foul quickly. TFF often maintains usable flux longer because flow runs across the membrane surface, but the benefit depends on the sample, membrane, pressure, crossflow, temperature, and equipment. Neither mode is universally faster or better.

What the two filtration modes actually do

Dead-end (direct-flow) filtration

In dead-end filtration, the feed is driven toward the membrane and generally perpendicular to it. The process attempts to send the liquid through the membrane as filtrate. Material larger than the membrane pores or otherwise retained stays at the membrane surface. As that retained layer grows, it adds resistance, so flow can decline and pressure can rise.

This arrangement is useful when the objective is simply to pass a suitable liquid through a filter, such as removing particles from a compatible feed. It can be compact and relatively simple, but the filter must be selected for the actual solids load, viscosity, concentration, and required throughput. A feed that quickly deposits protein, cells, aggregates, or other solids can lose flux rapidly.

Tangential-flow (crossflow) filtration

In TFF, also called crossflow filtration, the feed travels parallel to the membrane surface. Part of the liquid crosses the membrane as permeate; the material that remains in the circulating stream is the retentate. In many laboratory systems, retentate returns to the feed vessel and recirculates through the module.

Parallel flow can carry retained material along the membrane instead of allowing all of it to settle directly on the surface. That can reduce the rate of cake or concentration-layer formation and help sustain flux. It does not eliminate fouling: adsorption, concentration polarization, viscosity, particulates, temperature, and unsuitable operating conditions can still reduce performance.

A Cytiva/Pall technical guide summarizes the membrane choice this way: "There are two main membrane filtration modes which can use either microfiltration or ultrafiltration membranes." The mode describes flow direction; microfiltration (MF) and ultrafiltration (UF) describe membrane separation ranges. Do not treat TFF as synonymous with UF or dead-end filtration as synonymous with MF.

Dead-end vs. TFF at a glance

Decision axis Dead-end filtration Tangential-flow filtration
Flow path Feed moves toward and through the membrane, generally perpendicular to the surface. Feed moves along the membrane; permeate crosses while retentate remains in the flow path.
What happens to retained material It can accumulate on the surface and form a restrictive layer. Crossflow can sweep some material away from the surface, slowing rapid flux decline under suitable conditions.
Best-known process goals Pass-through filtration when the feed and filter are compatible. Concentration, diafiltration, fractionation, and selected clarification or cell-removal operations.
Fluid handling Often a single feed-to-filtrate path, with the filter changed or stopped as resistance increases. Requires a feed, retentate, and permeate path, usually with recirculation and controlled pumping.
Equipment burden Depends on the selected filter and feed; the setup may be comparatively simple. Needs a suitable module, tubing, pump or pump head, pressure monitoring, and a flow path designed for the module.
Scale Must be sized for the required duty and solids load. Systems are available from benchtop to production scale, with membrane area and flow ranges that vary by configuration.

Choose by the result you need

Choose dead-end filtration for a clean pass-through duty

Dead-end filtration is the sensible starting point when the feed is relatively low in foulants, the retained material is not the product you need to concentrate, and the desired result is a filtrate stream. It may be appropriate for a one-time clarification or particle-removal step where the filter can handle the expected load without an unacceptable pressure increase.

  • Use it when the process has one principal output: filtered liquid.
  • Confirm that the membrane chemistry, pore or molecular-weight rating, area, and pressure limit fit the feed.
  • Allow for filter capacity and a defined endpoint, such as a pressure limit or minimum flow.
  • Plan a staged or larger-area configuration if a small filter would blind quickly.

Choose TFF for concentration

Concentration removes solvent or other permeable liquid while retaining the target in the retentate. The circulating retentate becomes smaller in volume and higher in target concentration. This is useful for biomolecules and other macromolecules that must be concentrated without repeatedly transferring the sample.

For a retained biomolecule, select a UF membrane with a molecular-weight cutoff (MWCO) below the target’s molecular weight. The Cytiva/Pall guide gives a starting rule of thumb of an MWCO roughly three to six times lower than the target molecular weight. That is guidance for initial selection, not a guarantee of recovery: shape, aggregation, concentration, adsorption, membrane chemistry, and operating conditions can alter retention. If substantial diafiltration will follow, a lower MWCO may be advisable to limit target passage during the longer wash.

Choose TFF for diafiltration

Diafiltration washes smaller, permeable solutes from a retentate while retaining a larger target. In continuous diafiltration, fresh buffer is added as permeate leaves, keeping the retentate volume approximately steady while replacing salts, solvent, or other small molecules. The technical guide gives an example in which five diafiltration volumes reduce ionic strength by approximately 99 percent. That figure belongs to the guide’s continuous-diafiltration example; actual reduction depends on retention, mixing, volume control, and process execution.

Concentration and diafiltration are different operations even when performed on the same TFF system. Concentration primarily removes liquid to reduce volume. Diafiltration primarily replaces the liquid phase to wash out permeable components. A process may concentrate first, diafilter, and then reconcentrate, but the sequence should be chosen around product stability, viscosity, recovery, and the required final formulation.

Choose TFF for fractionation or selected clarification

TFF can separate larger from smaller molecules when the membrane retains one population and allows another to pass. It can also support certain clarification or cell-handling steps, provided the module, membrane, shear environment, and crossflow are appropriate for the material. These applications require more than choosing a nominal MWCO: cell sensitivity, debris loading, viscosity, and the desired product location (retentate or permeate) determine whether the process is practical.

A decision framework for a real sample

  1. Define the product stream. Decide whether the desired material should be in the filtrate/permeate or retained in the retentate. If the target must be concentrated or washed while retained, TFF is usually the relevant mode.
  2. Characterize the feed. Record approximate target size, concentration, particulate or cell content, viscosity, temperature sensitivity, adsorption risk, and expected volume. A high-protein or particulate feed can foul either mode, but dead-end operation often shows the decline sooner because all retained material loads the surface directly.
  3. Set the separation requirement. For UF, compare the target’s molecular size with candidate MWCOs. For MF, define the particle or cell-removal requirement. Remember that the same membrane class can be used in direct-flow or tangential-flow operation.
  4. Estimate the process sequence. Identify whether you need only clarification, concentration, diafiltration, fractionation, or a combination. Include hold-up volume, transfer steps, cleaning or disposal requirements, and the final recovery target.
  5. Check operating limits. Verify membrane area, allowable pressure, crossflow range, pump compatibility, minimum retentate volume, tubing size, and temperature range for the exact module. Vendor values are configuration-specific, not universal settings.
  6. Run a small, measurable trial. Track permeate flow, retentate volume, pressure, temperature, and product concentration or recovery. Use the trial to set an endpoint rather than assuming that a nominal MWCO or flow rate will deliver the desired result.

Membrane, module, and process variables

MWCO and retention

MWCO is a practical screening specification, not a sharp molecular sieve boundary. A target near the stated cutoff may show incomplete retention, and actual passage can change with conformation, aggregation, concentration, pressure, and membrane material. Starting with an MWCO several-fold below the target’s molecular weight can improve retention, but it may also reduce permeate flux and increase the time or area required. Test recovery and passage with the actual feed.

Membrane area and hold-up volume

More membrane area can support more throughput, while hold-up volume matters when the sample is scarce or expensive. TFF modules, tubing, connectors, and the feed vessel all contribute to residual volume. A module that works for a liter-scale process may be wasteful for a small sample if its minimum retentate volume is too high.

Crossflow, transmembrane pressure, and temperature

Crossflow helps manage surface buildup, but excessive pumping can damage shear-sensitive material or create unnecessary heat. Transmembrane pressure (TMP) drives permeation; increasing it does not always increase useful flux once a fouling or concentration-polarization layer controls the process. Temperature changes viscosity and can affect product stability. Establish an operating window experimentally instead of treating a maximum pressure as a recommended setpoint.

Feed concentration, particulates, and adsorption

High protein concentration, particulates, adsorption to the membrane, and lower temperature can slow a cassette workflow, as noted in Thermo Fisher’s cassette guidance. Pre-clarification, compatible membrane chemistry, controlled concentration, and a suitable rinse or conditioning step can help, but each intervention can affect recovery. Measure the target in feed, permeate, retentate, and system rinses when recovery is critical.

How to plan a laboratory TFF run

The exact instructions belong to the selected module and pump manual. The following is a process-development checklist, not a substitute for those instructions.

  1. Confirm compatibility. Check membrane material, MWCO or pore rating, chemical compatibility, pressure limit, temperature range, tubing, connectors, and the target’s stability.
  2. Size the module. Match membrane area and minimum working volume to the feed volume and desired endpoint. Avoid choosing solely by the nominal sample volume; account for retentate hold-up and expected fouling.
  3. Install the flow path. Connect the feed vessel, pump, module inlet, retentate return, and permeate collection. Include pressure measurement where specified. Verify that permeate and retentate cannot be accidentally crossed.
  4. Use the correct pump and head. Thermo Fisher’s Pierce cassette FAQ states that its cited setup requires a peristaltic pump and pump head, which are not supplied with that cassette. Other modules may have different requirements, so confirm the exact configuration before starting.
  5. Condition and flush as directed. Remove storage solution and equilibrate with the process buffer using the module’s instructions. Capture or discard the flush according to product-recovery and safety requirements.
  6. Start circulation gently. Establish retentate flow before applying the intended permeate-driving pressure. Watch for leaks, trapped air, unstable pressure, and unexpected permeate flow.
  7. Record a baseline. Log feed volume, temperature, pressure, retentate flow, permeate flow, and target concentration. Without a baseline, a later flux decline is difficult to interpret.
  8. Concentrate or diafilter. For concentration, remove permeate until the target volume is reached. For diafiltration, add fresh buffer at the planned rate as permeate leaves and maintain the required retentate volume. Keep the two operations conceptually separate in the batch record.
  9. Stop before the process becomes unstable. A rising pressure, falling flux, excessive temperature, foaming, or product loss can signal that the chosen endpoint or conditions are unsuitable.
  10. Recover and document. Flush the retentate and module as appropriate, measure final volume and concentration, and account for material in tubing, the module, permeate, and rinses.

How to run and monitor dead-end filtration

  1. Select the filter for the feed load. Consider membrane chemistry, pore or molecular-size rating, area, pressure limit, and expected solids.
  2. Prepare the feed. Remove easily settled or oversized material when the process allows, and control temperature so viscosity remains within the filter’s practical range.
  3. Wet or condition the filter. Follow the filter’s instructions for wetting, flushing, and compatibility. Record any pre-rinse volume if product recovery matters.
  4. Filter at a controlled pressure or flow. Avoid forcing the feed through a rapidly blinding filter. Monitor pressure and filtrate flow rather than relying on elapsed time.
  5. Define the endpoint. Stop at the specified pressure, minimum acceptable flow, maximum process time, or volume processed. If the endpoint is reached early, investigate feed fouling or filter sizing instead of simply increasing pressure.
  6. Assess the result. Check filtrate clarity or particle load and, where relevant, measure target recovery. A clear filtrate does not prove that a valuable retained product was recovered.

Fouling and troubleshooting

Flux falls quickly in dead-end mode

Likely causes include a high solids load, concentration polarization, adsorption, an undersized filter, or a feed that is too viscous or cold. Confirm the feed condition, reduce the load with an upstream clarification step if compatible, increase filter area, or evaluate TFF if retaining the product and sustaining flow are more important than a simple pass-through setup.

TFF flux declines during circulation

Crossflow can limit rapid surface buildup but cannot overcome every fouling mechanism. Check for increasing retentate concentration, particles, aggregation, adsorption, temperature drift, air, or a pressure level that is encouraging a compacted layer. Compare permeate flow with pressure and crossflow records; changing one variable at a time makes the cause easier to identify.

Product appears in permeate

The MWCO may be too high for the target, the target may be smaller than assumed, or operating conditions may be increasing passage. Verify the target’s actual molecular form and assay permeate, retentate, and recovery rinses. A lower MWCO can improve retention but may reduce flux and increase processing time.

Pressure is unstable or the pump loses prime

Inspect tubing connections, air bubbles, vessel level, pump-head installation, and module orientation. Confirm that the retentate return is open and that the permeate line is not blocked. Follow the module’s pressure limits; do not treat a pressure spike as a reason to exceed them.

Recovery is lower than expected

Losses may occur through adsorption, irreversible retention, precipitation, denaturation, hold-up volume, or sampling. Perform a material balance where practical, including permeate and rinse samples. Membrane conditioning and a lower final concentration may help, but the remedy depends on the measured loss mechanism.

What the cited laboratory cassette specifications mean

Sartorius lists the Vivaflow 200 as a research-use TFF cassette for ultrafiltration and diafiltration, with applications including concentration, buffer exchange, desalting, and fractionation. The cited configuration has the following specifications:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
Specification Listed value How to interpret it
Membrane area 200 cm² Area for that configuration; it is not a universal area requirement for laboratory TFF.
MWCO 10 kDa One membrane configuration; select a different rating when the target and process require it.
Maximum operating pressure 4 bar A stated limit for the cited cassette, not a recommended operating pressure.
Minimum retentate volume 20 mL The smallest stated working retentate volume for that configuration; account for hold-up and recovery.
Recommended feed volume 1,000–2,500 mL A vendor recommendation for the cited setup, not a promise that every sample in this range will perform identically.
Recommended flow rate 200–400 mL/min A configuration-specific recommendation that must be checked against the current manual and sample sensitivity.

Sartorius describes its broader TFF portfolio as spanning benchtop to production scale and including single-use and multi-use systems. Overall portfolio ranges vary by system, membrane area, and flow path, so they should not be read as a performance comparison with dead-end filtration. Product configurations and availability can change; verify the current datasheet, tubing, pump compatibility, membrane option, and sample range before purchase.

Scale-up and validation considerations

Scaling TFF is more than multiplying membrane area. Preserve the process conditions that control retention and fouling: feed concentration, temperature, crossflow, TMP, residence time, and the relationship between membrane area and batch volume. A larger system may have different hold-up volume, pump behavior, mixing, and pressure drop. Reconfirm product recovery and permeate quality at each scale.

For a robust process, define measurable acceptance criteria before the run:

  • Final retentate volume and target concentration.
  • Permeate flow or total permeate volume.
  • Pressure and temperature limits.
  • Product recovery and passage into permeate.
  • Buffer-exchange or ionic-strength endpoint.
  • Maximum processing time and acceptable hold-up loss.
  • Membrane integrity or post-use inspection, where applicable.

Dead-end filtration also benefits from validation, particularly when the filter is a critical clarification step. Track throughput, pressure rise, filtrate quality, and product recovery across representative feed lots rather than assuming that a clean laboratory sample predicts production behavior.

Bottom-line selection

Pick dead-end filtration for a compatible, primarily pass-through duty where a simple flow path and disposable filter meet the capacity requirement. Pick TFF when the process must keep a target in the retentate while concentrating, diafiltering, fractionating, or handling a feed likely to foul a direct-flow membrane quickly. In both cases, choose the membrane and operating window from the actual target, feed, scale, pressure limits, and recovery data—not from the filtration mode name alone.
